Los osteocitos son células terminalmente diferenciadas de progenitores osteoblásticos que se incrustaron en su matriz secretada1. Son las células más abundantes y longevas entre las poblaciones de células óseas. Residen dentro de lagunas y tienen una morfología estrellada característica con dendritas que se extienden a través de canales llamados canalículos formando una extensa red de comunicación e intercambio metabólico con su entorno circundante y la superficie ósea2. Los osteocitos coreografian tanto osteoblastos como osteoclastos en la remodelación ósea, son las células mecanosensoriales primarias que confieren adaptación a la carga mecánica3, intervienen en la homeostasis del fosfato4 y en la mineralización de la matriz ósea5, y junto con el sistema lacunocanalicular, actúan como órgano endocrino que señala tejidos distantes6.
Los osteocitos están situados dentro de una matriz mineralizada, lo que limita la accesibilidad y dificulta su aislamiento, lo que dificulta la investigación in vitro. Uno de los primeros métodos de aislamiento describió osteocitos aislados de calvaria de pollo utilizando un anticuerpo monoclonal específico de osteocitos (OB7.3)7 que más tarde se supo que era la variante aviar de PHEX (gen regulador de PHosphate con homología a endopeptidasas en el cromosoma X)8. Otros investigadores utilizaron la digestión secuencial de huesos largos de rata 9 o ratón 10 para obtener fracciones ricas en osteocitos en las que se informó que la pureza de los osteocitos era de alrededor del70%9. Las limitaciones de este procedimiento incluyen la pureza subóptima de los cultivos contaminados con otros tipos de células además de los osteocitos y que los osteocitos podrían ser potencialmente sobrecultivados por otras células en cultivo ya que los osteocitos han perdido la capacidad de dividirse. Estos desafíos restringen la usabilidad de las culturas a largo plazo.
Para superar estas limitaciones, se desarrollaron diferentes líneas celulares de osteocitos. La línea celular MLO-Y411 y la línea celular MLO-A512 son, en particular, las líneas celulares más estudiadas que son útiles para el estudio de los osteocitos en etapa temprana; sin embargo, son menos útiles para estudiar la señalización de osteocitos maduros, ya que expresan bajos niveles de esclerostina y FGF2313, los cuales son marcadores de osteocitos maduros. Otras líneas celulares, incluidas IDG-SW314 y Ocy45415, expresan altos niveles de esclerostina y FGF23 y son útiles para estudiar la etapa tardía de osteocitos. Las líneas celulares demuestran ser herramientas de investigación útiles; Sin embargo, no vienen sin limitaciones, ya que no representan completamente la biología de la célula primaria. Diferentes líneas celulares representan diferentes etapas de desarrollo del espectro de madurez de los osteocitos, y las líneas celulares no representan la heterogeneidad de los osteocitos primarios16,17.
Para obtener cultivos puros de osteocitos primarios, los investigadores aprovecharon el modelo de ratón cre en el que se utiliza el promotor dmp1 de 8 kb para impulsar la expresión de proteína fluorescente verde (GFP) en osteocitos18,19. Para obtener poblaciones de osteocitos se han utilizado ratones transgénicos duales (pOB-Col 2.3-GFP-cian y DMP1-GFP-topaz) por Paic et al.19 y ratones transgénicos dmp1-topacio por Nakashima et al.20. En el que emplearon digestión secuencial y FACS de osteocitos que expresan GFP para adquirir cultivos de osteocitos primarios19,20. Se demostró que la dirección de cre en el ratón reportero dmp1 de 10 kb Ai9, que activa la proteína tdTomato, está presente en osteocitos, osteoblastos, músculos y células dentro de la médula ósea. El promotor dmp1 de 8 kb tenía el mismo patrón de expresión, pero sólo una proporción de osteoblastos y células de médula ósea expresaba la proteína, lo que indica que el promotor dmp1 de 8 kb es más específico21. A pesar de esto, los resultados obtenidos utilizando el promotor dmp1 de 8 kb deben interpretarse con precaución, y los perfiles de expresión génica deben llevarse a cabo de forma rutinaria utilizando marcadores específicos de osteocitos frente a osteoblastos para garantizar que la población obtenida sea de pureza lo suficientemente alta.
Los marcadores de osteoclastos OSCAR y Dcstamp se encontraron en poblaciones hematopoyéticas de osteocitos no agotados frente a poblaciones de osteocitos agotados, este hallazgo llevó a los autores a concluir que los digerios obtenidos del fraccionamiento de la calvaria neonatal de 8 kb dmp1-topacio y la clasificación de GFP están contaminados con células hematopoyéticas. La contaminación con células hematopoyéticas podría haberse mitigado apretando la puerta de clasificación de GFP, ya que las células hematopoyéticas positivas para GFP tenían una intensidad de GFP razonablemente menor que las células mesenquimales positivas para GFP (osteocitos)22.
Los métodos para estudiar los osteocitos in vitro han contribuido a la reciente riqueza de información sobre la biología de los osteocitos. Sin embargo, el aislamiento de osteocitos sigue siendo un procedimiento laborioso y prolongado con bajos rendimientos celulares. El método descrito de digestión ósea utilizando colagenasa y EDTA a menudo hasta la fracción 823, toma varias horas en las que se grava la viabilidad de los osteocitos. This protocol reduces the handling time and allows a higher number of osteocytes to be obtained in a single procedure with minimal contamination from hematopoietic or mesenchymal cells.For the isolation of GFP-expressing osteocytes, place euthanized, postnatal day six to seven, transgenic mouse pups into a non-treated culture dish, and use scissors and tweezers to grasp the skin at the base of one's skull. Use the scissors to make an initial incision in the lifted skin, followed by cuts along both sides of the skull in front of the ears. Remove the skin to expose the calvaria and holding the head from the nasal bridge, cut the calvaria along the lamboid suture and along the edges of the parietal bones to the frontal part of the tissue.Separate the calvaria from the underlying brain tissue and place the bones concave side up in PBS. To obtain fraction one, pool up to five calvaria per 50-milliliter tube in five milliliters of freshly prepared two-milligram per milliliter collagenase, and incubate the tissues at 37 degrees Celsius and 300 revolutions per minute for 20 minutes. At the end of the incubation, discard the digest from fraction one and wash the calvaria with five milliliters of PBS.Replace the wash with five milliliters of five-millimolar EDTA supplemented with one milligram per milliliter of bovine serum albumin in PBS for 15 minutes at 37 degrees Celsius with shaking. At the end of the incubation, transfer the digest to a 50-milliliter conical tube and wash the calveria with five milliliters of PBS. Then transfer the wash solution to the digest.To obtain fraction two, place the digest for centrifugation and discard the supernatant. Re-suspend the pellet in eight milliliters of alpha minimum essential medium containing 10%fetal bovine serum, or FBS, and antibiotics, and seed the cells onto a 10-centimeter culture dish. Then place the fraction two cells into the cell culture incubator.After collecting and culturing fractions three and four as just demonstrated. Incubate the calveria in five milliliters of five-millimolar EDTA supplemented with one milligram per milliliter of bovine serum albumin in PBS for 15 minutes at 37 degrees Celsius with shaking. At the end of the incubation, collect and culture fraction five as demonstrated for fractions two, three, and four.Immediately after fractionation, collect the cells for sorting. Alternatively, collect the cells after 24 hours of culture by aspirating the medium and gently wash the cells two times with 10 milliliters of PBS per dish per wash. After the second wash, treat the cells with five milliliters of 0.5%tripsin EDTA in PBS per plate.After five minutes at 37 degrees Celsius, add five milliliters of complete alpha minimum essential medium to each plate and gently detach the cells with pipetting. To combine the cells in each fraction, filter the cell suspensions through a 40-micron cell strainer into a single 50-milliliter conical centrifuge tube. After washing the cells with 10 milliliters of PBS, add them to the tube.Filter the cells through a strainer and collect the cells by centrifugation. Once the cells are pelleted down, re-suspend the pellet in complete minimal essential medium to a one times 10 to the seventh cells per milliliter of medium concentration and filter the cells through a 35-micron nylon mesh capped tube. To test the efficiency of this method, fractions were separately sorted and cultured for 24 hours to determine the percentage and yield of osteocytes from each fraction.The density of the osteocytes in fractions two to five were higher than that observed for fraction one and decreased remarkably in fractions six, seven, and eight. Although the percentage of osteocytes obtained between all of the fractions is not statistically significant, the density of the osteocytes differs dramatically between fractions. Here the gating strategy for isolating GFP-positive osteocytes from their GFP-negative counterparts by fluorescence-activated cell sorting is shown.After sorting, osteocytes can be analyzed for the expression of osteocyte markers of interest. In this analysis, the osteocyte marker genes were expressed at higher levels in the isolated GFP-positive osteocytes compared to presort fraction two, which is known as a high osteoblast fraction. Here the morphology of a GFP-positive osteocyte retaining a stellate shape with dendrites extending from the cell body after 24 hours of culture on a plastic culture dish, post sort, can be observed.Pay attention when aspirating the digests as the calveria will be reduced to minute pellets. Remember to use a small tip aspirator to avoid cell loss. Osteocytes have been enigmatic because they are entombed in bone.This technique has paved the way to studying osteocyte molecular interactions and their control over osteoclasts and osteoblasts.
