Protokollen præsenteres her er tidligere blevet beskrevet ogbrugt 12,15,17,31 til at måle kapaciteten af antistoffer rettet mod overfladen af P. falciparum IEs at fremkalde opsonisation og fagocytose af THP-1 celler. Resultaterne her fokuserer på naturligt erhvervede VAR2CSA-specifikke antistoffer i plasma/serum hos kvinder, der bor i en P. falciparum endemisk region. VAR2C...

Malaria er en vektorbåren sygdom forårsaget hos mennesker ved infektion med fem forskellige arter af slægten Plasmodium. De mest udbredte arter er P. falciparum, som også er ansvarlig for den mest sygelighed og dødelighed1. Malaria klinisk præsentation varierer fra asymptomatisk eller godartede infektioner til komplicerede / alvorlig sygdom, sidstnævnte forekommer for det meste hos børn under fem år. Eksponering for P. falciparum fremkalder ikke steril immunitet, men personer, der bor i endemiske områder, udvikler langsomt immunitet mod den kliniske sygdom. Beskyttelse er alder/ eksponering afhængig og immunitet er normalt erhvervet i løbet af de første 5-10 år af livet2. Voksne kvinder er en vigtig undtagelse, som svær malaria kan forekomme under graviditet i en klinisk præsentation kendt som placenta malaria (PM). PM er en vigtig årsag til abort, dødfødsel, for tidlig fødsel, lav fødselsvægt, fosterdød, og moder anæmi. Modstand mod PM udvikler sig over successive graviditeter3. Beskyttelse mod PM er forbundet med erhvervelse af antistoffer mod VAR2CSA-type PfEMP14,5, et inficeret erythrocyt (IE) overfladeantigen, der binder sig til chondroitinsulfat A (CSA), så IE-binding i moderkagen. Antistoffer mægle beskyttelse udfører forskellige funktionelle aktiviteter (gennemgået i6,7), herunder opsonization af IEs at fremkalde fagocytose. Her præsenterer vi en protokol til måling af denne funktion af antistoffer til stede i humant plasma / serum, ved hjælp af in vitro dyrkede IEs udtrykke VAR2CSA sammen med monocyt linje THP-1. Analysen har tidligere været anvendt11,12,13,14,15,16,17,18 ogbetragtes som en forbedret og lettere tilgang i forhold til tidligere mikroskop-baserede protokoller8, da det giver mulighed for test af et større antal antistof prøver i en enkelt køre ved hjælp af mindre mængder af antistof og undgå kedelige og skæv mikroskopi optælling. Selv om analysen er blevet brugt af flere laboratorier og dens udførelse er enkel nok, det kræver omhyggelig planlægning og forberedelse, derfor, en detaljeret protokol ville tillade dens anvendelse af laboratorier og forskere mangler tidligere erfaringer. Vi bruger, som et eksempel, sene-fase-synkroniserede IEs udtrykke VAR2CSA opsonized med antistoffer til stede i serum indsamlet fra kvinder med naturligt erhvervet PM-specifik immunitet. measuring naturally acquired phagocytosis-inducing antibodies to plasmodium falciparum parasites by a flow cytometry-based assay

Protokollen beskriver, hvordan man opretter og kører en flow cytometri-baseret fagocytose assay af Plasmodium falciparum-inficeredeerythrocytter (IEs) opsonized af naturligt erhvervet IgG antistoffer specifikke for VAR2CSA. VAR2CSA er parasitantigen, der formidler den selektive binding af IEs i moderkagen, der kan forårsage en alvorlig form for malaria hos gravide kvinder, kaldet placenta malaria (PM). Measuring Naturally Acquired Phagocytosis-Inducing Antibodies to Plasmodium falciparum Parasites by a Flow Cytometry-Based Assay

Denne protokol gør det muligt at måle antistoffers evne til at opsonisere og fremkalde fagocytose af plasmodium falciparum inficerede erythrocytter. For hver testet prøve kan fagocytose rapporteres som den absolutte værdi eller som den relative fagocytose beregnet som en procentdel ved hjælp af den positive kontrol som maksimum.THP-1 cellerne skal regelmæssigt kontrolleres for FC gamma receptor overfladeudtryk ved flow cytometri. Cellerne skal være negative for CD16 og positive for CD32 og CD64. I dette opsonisationseksperiment blev THP-1-cellerne først lukket i henhold til deres fremadrettede og sidespistprofiler for at gøre det muligt at kvantificere procentdelen af ethidiumbromid-mærkede celler, hvilket indikerer celler, der har fagocytret mindst ét antistofopsoniseret inficeret erythrocyt.De negative kontrolprøver bør alle generere en enkelt negativ top i FL3-kanalen, med få hændelser observeret i ethidium bromid markør. Derfor bør de gennemsnitlige fagocytoseværdier, både som absolutte ethidiumbromid-positive THP-1-celler og som relative fagocytoseprocenter, være meget lave. I modsætning hertil bør den positive kontrol generere spor med to toppe, en negativ top og en klart positiv og godt adskilt top placeret inden for ethidium bromid markør.De gennemsnitlige fagocytoseværdier er normalt højest for den positive kontrol, efterfulgt af den malaria-eksponerede kvindelige pool og den enkelte malaria-eksponerede kvinde kontrol. Selv om denne metode giver ensartede resultater, er der observeret afvigelser i hældningskoefficienten for de justerede linjer. Derfor anbefales det at bruge relative værdier, især hvis det ikke er muligt at køre alle prøverne i et enkelt eksperiment.Sørg for omhyggeligt at planlægge eksperimentet på forhånd, forberede både THP-1 celler og parasitter i overensstemmelse hermed, ved hjælp af kun trophocytter med en renhed på mere end 80%Derudover skal du sørge for altid at omfatte de relevante kontroller, for at gøre det muligt for kvaliteten af forsøget, der skal vurderes og lette dataanalyse.
