Malaria is een door vector overgedragen ziekte die bij de mens wordt veroorzaakt bij infectie met vijf verschillende soorten van het geslacht Plasmodium. De meest voorkomende soort is P. falciparum, dat ook verantwoordelijk is voor de meest morbiditeit en mortaliteit1. Malaria klinische presentatie varieert van asymptomatische of goedaardige infecties tot gecompliceerde / ernstige ziekte, de laatste komt meestal voor bij kinderen jonger dan vijf jaar. Blootstelling aan P. falciparum leidt niet tot steriele immuniteit, maar personen die in endemische gebieden leven, ontwikkelen langzaam immuniteit tegen de klinische ziekte. Bescherming is afhankelijk van leeftijd/blootstelling en de immuniteit wordt normaal gesproken verworven tijdens de eerste 5-10 jaar van het leven2. Volwassen vrouwen zijn een belangrijke uitzondering, omdat ernstige malaria kan optreden tijdens de zwangerschap in een klinische presentatie bekend als placentamalaria (PM). PM is een belangrijke oorzaak van abortus, doodgeboorte, vroegtijdige bevalling, laag geboortegewicht, foetale dood, en moederbloedarmoede. Weerstand tegen PM ontwikkelt zich over opeenvolgende zwangerschappen3. Bescherming tegen PM wordt geassocieerd met de verwerving van antilichamen tegen VAR2CSA-type PfEMP14,5, een besmet erythrocyte (IE) oppervlakteantigenen dat zich bindt aan chondroitin sulfaat A (CSA) waardoor IE-sekwestratie in de placenta mogelijk wordt. Antilichamen bemiddelen bescherming bij het uitvoeren van verschillende functionele activiteiten (beoordeeld in6,7) waaronder opsonisatie van IEs om fagocytose te induceren. Vroege in vitro studies toonden aan dat antilichamen de groei van P. falciparum kunnen beperken in aanwezigheid van monocyten via fagocytose8,9. Recentere studies hebben aangetoond dat hogere niveaus van fagocytose-inducerende antilichamen worden geassocieerd met betere zwangerschapsresultaten (in de context van HIV-co-infectie)10,11, wat de relevantie van deze effectorfunctie in de natuurlijk verworven immuunrespons aangeeft.
Hier presenteren we een protocol om deze functie van antilichamen aanwezig in menselijk plasma / serum te meten, met behulp van in vitro gekweekte IT's uitdrukken VAR2CSA samen met de monocyten lijn THP-1. De test is eerder gebruikt11,12,13,14,15,16,17,18 en wordt beschouwd als een verbeterde en gemakkelijkere aanpak in vergelijking met eerdere microscopie gebaseerde protocollen8, omdat het mogelijk maakt het testen van een groter aantal antilichaam monsters in een enkele run met behulp van kleinere volumes van antilichamen en het vermijden van vervelende en bevooroordeelde microscopie tellen. Hoewel de test is gebruikt door meerdere laboratoria en de uitvoering ervan is eenvoudig genoeg, het vereist een zorgvuldige planning en voorbereiding, daarom, een gedetailleerd protocol zou de toepassing ervan door laboratoria en onderzoekers ontbreekt eerdere ervaring. We gebruiken als voorbeeld laat-stadium-gesynchroniseerde IEs uitdrukken VAR2CSA opsonized met antilichamen aanwezig in serum verzameld van vrouwen met natuurlijk verworven PM-specifieke immuniteit. Het protocol kan echter eenvoudig worden gewijzigd om de functionaliteit van antilichamen tegen parasietantigeen op het IE-oppervlak te testen, hetzij veroorzaakt door natuurlijke blootstelling of door vaccinatie.

measuring naturally acquired phagocytosis-inducing antibodies to plasmodium falciparum parasites by a flow cytometry-based assay

Het protocol beschrijft hoe een op stroom gebaseerde phagocytosetest van Plasmodium falciparum-geïnfecteerdeerytrocyten (IEs) op te zetten en uit te voeren, opmeting door natuurlijk verworven IgG-antilichamen die specifiek zijn voor VAR2CSA. VAR2CSA is het parasietantgeen dat bemiddelt bij de selectieve inbeslagname van IE's in de placenta die een ernstige vorm van malaria kan veroorzaken bij zwangere vrouwen, placentamalaria (PM). Bescherming tegen PM wordt bemiddeld door VAR2CSA-specifieke antilichamen die vermoedelijk functioneren door het remmen van placenta-inname en/of door het opsoniseren van IE's voor fagocytose. De test maakt gebruik van laat-stadium-gesynchroniseerde IEs die zijn geselecteerd in vitro om VAR2CSA, plasma / serum-antilichamen uit te drukken van vrouwen met natuurlijk verworven PM-specifieke immuniteit, en de fagocytische cel lijn THP-1. Het protocol kan echter eenvoudig worden gewijzigd om de functionaliteit van antilichamen tegen parasietantigeen op het IE-oppervlak te testen, hetzij veroorzaakt door natuurlijke blootstelling of door vaccinatie. De test biedt eenvoudige en hoge doorvoer evaluatie, met een goede reproduceerbaarheid, van een belangrijk functioneel aspect van antilichaam-gemedieerde immuniteit in malaria. Het is daarom nuttig bij de evaluatie van de klinische immuniteit tegen P. falciparum malaria, een belangrijke oorzaak van morbiditeit en mortaliteit in de tropen, met name in Afrika bezuiden de Sahara.

Measuring Naturally Acquired Phagocytosis-Inducing Antibodies to Plasmodium falciparum Parasites by a Flow Cytometry-Based Assay

Dit protocol maakt het mogelijk om het vermogen van antilichamen te meten om opsoniseren en induceren de fagocytose van plasmodium falciparum geïnfecteerde erytrocyten. Antilichamen die aanwezig zijn in het serum van personen die van nature zijn blootgesteld aan parasietinfectie, evenals antilichamen die worden veroorzaakt door immunisatie met parasietantigenen, kunnen worden getest. Het aantonen van de procedure met mij zal Maiken Visti, een technicus van mijn laboratorium.Om een THP-1 celcultuur op te zetten voor een fagocytosetest, zaad de dag voor het experiment de cellen in een 25 centimeter kwadraat kweekkolf en een concentratie van 2,5 keer 10 tot de vijfde cellen per milliliter in THP één celcultuurmedium. Op de dag van het experiment, ten minste een uur voor het experiment blok tot ronde-bodem 96-put platen met 150 microliters van steriele 2%FBS in PBS per put. Voor de oogst en zuivering van trophozoiet met midden tot laat stadium, geïnfecteerde erytrocyten, bereid ongeveer 1,2 milliliter verpakte P falciparumparasietencultuur voor in 10 milliliter parasietenkweekmedium, met ten minste 5%parasitemia en voeg toe aan een magnetische kolom.Was de kolom met 50 milliliter parasiet cultuurmedium. Verzamel vervolgens de geïnfecteerde erytrocyten door centrifugatie en resuspend de pellet in een milliliter van verse parasiet cultuur medium voor het tellen.Pas de gezuiverde geïnfecteerde erythrocytensuspensie met 3,3 keer 10 aan op de zevende cellen per milliliter in parasietcultuurmedium dat ethidiumbromiide bevat, tot een uiteindelijke concentratie van 2,5 microgram per milliliter. Verwijder vervolgens de blokkeringsoplossing van een plaat met 96 put door de plaat in een afvalcontainer te vegen. Verwijder vervolgens een overmaat over een papieren handdoek en voeg 30 microliters van de ethidium bromide label geïnfecteerde erythrocyte suspensie tot alle, maar een goed in de bovenste helft van de plaat.Voeg 30 microliters van parasiet cultuur medium aan de lege put, en broeden de plaat gedurende 10 minuten op kamertemperatuur, beschermd tegen licht. Voeg aan het einde van de incubatie 170 microliters van parasietcultuurmedium toe aan elke put en sediment de geïnfecteerde erytrocyten op de bodem van de plaat door centrifugatie. Verwijder de supernatant door de plaat in een afvalcontainer te vegen en was de ethidium bromide-gelabelde geïnfecteerde erytrocyten nog twee keer, met 200 microliters van parasietencultuurmedium per put, zoals net is aangetoond.Gebruik na de tweede wasbeurt een multi-channel pipet om het volledige volume van supernatant zorgvuldig uit elke put te verwijderen, zonder de pellets te storen. En resuspend de ethidium bromide-gelabelde geïnfecteerde erytrocyten in 30 microliter van antilichaam oplossing, met inbegrip van de juiste controles en testmonsters eerder bereid in parasiet cultuur medium. Plaats vervolgens de plaat op 37 graden Celsius gedurende 45 minuten, beschermd tegen licht.Terwijl de geïnfecteerde erytrocyten worden opsonized, het verzamelen van de THP-1 cellen door centrifugatie, en resuspend de pellet in 12 milliliter verse, voorverwarmde THP-1 cel cultuur medium. Na een tweede centrifugatie, resuspend de pellet in een milliliter van THP-1 cel cultuur medium voor het tellen, en pas de celconcentratie tot vijf keer 10 tot de vijfde cellen per milliliter in THP-1 cel cultuur medium. Verwijder de blokkerende oplossing van de tweede 96-put plaat en voeg 100 microliter THP-1 cellen aan elke put.Plaats vervolgens de plaat in de couveuse van de celcultuur. Voeg aan het einde van de opsonisatie incubatie 170 microliters van parasietcultuurmedium toe aan elke put van de opsonisatieplaat en sediment de geïnfecteerde erytrocyten op de bodem van de plaat door centrifugatie. Was de opsonized geïnfecteerde erytrocyten twee keer met 200 microliters van parasiet cultuur medium per put, met behulp van een multi-channel pipet om zorgvuldig te verwijderen van de supernatant na de tweede wasbeurt.Resuspend de opsonized geïnfecteerde erytrocyten in 100 microliters van voorverwarmde THP-1 cel cultuur medium per goed, en breng 50 microliter van elke opsonized geïnfecteerde erytrocyten suspensie naar een overeenkomstige put in de faagocytose plaat. Wanneer alle cellen zijn toegevoegd, plaats de plaat in de celcultuur incubator voor niet meer dan 40 minuten, beschermd tegen licht. Aan het einde van de incubatie, stop de fagocytose door centrifugatie bij vier graden Celsius.Verwijder de supernatant en resuspend de pellets met 150 microliters van kamertemperatuur ammonium chloride lysing oplossing per goed. Voeg na precies drie minuten 100 microliters ijskoud toe bij 2%FBS in PBS om de lyse te stoppen en sediment de intacte cellen door centrifugatie. Was vervolgens de cellen drie keer met 200 microliters verse ijskoude 2%FBS in PBS per put, per wasbeurt.Na de laatste wasbeurt, resuspend de cel pellet in 200 microliters van verse ijskoud 2%FBS in PBS per put. Voor stroomcytometrieverwerving en -analyse, laad onmiddellijk de cellen op een flow cytometer en gebruik de lineaire voorwaartse versus lineaire zijverstrooiingsplot voor de putten zonder geïnfecteerde erytrocyten om de THP-1 cellen te bekiseren. Verwerf 10.000 gebeurtenissen in deze poort en gebruik de THP-1 cellen met de geïnfecteerde erytrocyten die zijn gedesoniseerd met een positieve controle om een histogramplot op te zetten om de fluorescentieintensiteit van ethidium bromide te meten.Voor analyse, open de lineaire voorwaartse versus lineaire kant scatter plot voor de putten zonder geïnfecteerde erythrocyten, en poort de THP-1 cellen. Zet vervolgens een positieve poort in een FL-3 histogram en kopieer deze poorten op alle andere monsterputten om het percentage ethidium bromide-positieve THP-1 cellen te bepalen. Voor elk getest monster kan fagocytose worden gerapporteerd als de absolute waarde of als de relatieve fagocytose berekend als een percentage met behulp van de positieve controle als maximum.De THP-1 cellen moeten periodiek worden gecontroleerd op FC gamma receptor oppervlakte expressie door flow cytometrie. De cellen moeten negatief zijn voor CD16, en positief voor CD32 en CD64. In dit opsonisatie-experiment werden de THP-1 cellen eerst afgesloten op basis van hun voorwaartse en zijverstrooiingsprofielen, om kwantificering van het percentage ethidium bromide-gelabelde cellen mogelijk te maken, indicatief voor cellen die ten minste één antilichaam opsonized geïnfecteerde erytrocyte hebben geprocificeerd.De negatieve controlemonsters moeten allemaal een enkele negatieve piek in het FL3-kanaal genereren, met weinig gebeurtenissen waargenomen in de ethidium bromidemarker. Daarom moeten de gemiddelde phagocytosewaarden, zowel als absolute ethidium bromide-positieve THP-1-cellen als als relatieve fagocytosepercentages, zeer laag zijn. De positieve controle moet daarentegen sporen genereren met twee pieken, een negatieve piek en een duidelijk positieve en goed gescheiden piek in de ethidium bromidemarker.De gemiddelde phagocytose waarden zijn normaal gesproken het hoogst voor de positieve controle, gevolgd door de malaria-blootgestelde vrouwelijke zwembad en de enkele malaria-blootgestelde vrouw controles. Hoewel deze methode consistente resultaten oplevert, zijn afwijkingen in de hellingscoëfficiënt van de aangepaste lijnen waargenomen. Daarom wordt het gebruik van relatieve waarden aanbevolen, vooral als het niet mogelijk is om alle monsters in één experiment uit te voeren.Zorg ervoor dat u het experiment van tevoren zorgvuldig plant, zowel de THP-1 cellen als de parasieten dienovereenkomstig voorbereidt, met alleen trophocyten met een zuiverheid van meer dan 80%Bovendien moet u altijd de juiste controles opnemen, zodat de kwaliteit van het experiment kan worden beoordeeld en gegevensanalyse kan worden vergemakkelijkt.
