three-dimensional motor nerve organoid generation

축삭의 매심은 신경계에서 관찰되는 주요 구조 적분 중 하나입니다. 축 하 매심의 중단 개발 및 신경 퇴행 성 질환을 일으킬 수 있습니다. 축축의 수많은 연구가 수행되었지만, 축축기의 형성과 기능 장애에 대한 우리의 이해는 여전히 강력한 3차원 체외 모델의 부족으로 인해 제한됩니다. 여기서, 우리는 미세유체계 조직 배양 칩에서 인간 유도만능 줄기(iPS) 세포로부터 모터 신경 오르가노이드(MNO)의 신속한 생성을 위한 단계별 프로토콜을 설명한다. 첫째, 방법에 사용되는 칩의 제조가 설명된다. 인간 iPS 세포로부터, 운동 신경 세포 (MNS)가 형성된다. 다음으로, 차별화된 MNS는 칩으로 전달된다. 그 후, 축삭은 자발적으로 스페로이드에서 자라서 칩에 장착된 마이크로채널 내의 근막으로 조립되어 스페로이드에서 확장된 축삭 뭉치를 운반하는 MNO 조직을 생성한다. 다운스트림 분석을 위해, MMO는 형태학적 분석을 위해 고정되거나 생화학 적 분석을 위해 해부될 칩에서 꺼내질 수 있을 뿐만 아니라 칼슘 이미징 및 다중 전극 어레이 기록을 위해 제거될 수 있다. 이 프로토콜로 생성된 MMO는 약물 검사 및 스크리닝을 용이하게 할 수 있으며 축축기 근막의 근본적인 발달 및 질병 메커니즘에 대한 이해에 기여할 수 있습니다.

운동 뉴런은 3D 분화절차(도 4 및 도 5)에서12-14일 이내에 분화되었다. 중요한 것은, 세포의 60% 이상이 분화 중에 모터 뉴런 마커 HB9를 발현하였다. 면역히스토케는 MNS내 세포의 약 80%가 SMI32 양성 운동 뉴런이었다는 것을 밝혔다. HB9 및 SMI32는 확립된 초기 단계 모터 뉴런마커(15,16)이다. HB9 및 SMI32의 발현은 모터 뉴런의 세포 정체성을 보장하기 위해 확인되어야 하는 주요 매개 변수입니다. MNS를 배양 칩에 도입한 후 축축은 채널로 확장되고 축축묶음이 형성됩니다. 물리적 가이드역할을 하는 마이크로채널로 인해 축축은 MNS로부터 길게 문어축되어 축산작용(그림6A)에의해 번들을 형성한다. MNO의 생성을 확인하기 위해서는 현미경 관찰에 의해 축축 묶음의 형성을 확인하는 것이 필수적이다. 성공적인 MNO는 50 μm보다 넓은 축축 묶음과 채널의 번들에서 분리된 축록새가 거의 없습니다. 축축의 초기 신장은 스페로이드의 도입 후 24시간 후에 관찰될 수 있다. 다음 3-4 일 이내에 축축은 마이크로 채널의 중앙에 도달한 다음 추가 10 일 이내에 다른 쪽 끝에 도달합니다(그림 6A). 결과적으로, 축축은 칩에서 2-3주 만에 직선 및 단방향 번들을 조립하고 형성했으며, 그 후 뉴런 활동이 관찰되었다.
모터 신경 오르가노이드는 생물학적 분석을 위해 현미경 유리로부터 PDMS를 분리하여 칩으로부터 수집될 수있다(도 6B). 축삭 번들 및 세포 체는현미경(도 6B)하에서수술용 칼 또는 트위저를 사용하여 절단하여 해부및 격리될 수 있다. RNA와 단백질을 포함하는 이 생물학 물질은 RT-PCR 및 서쪽 블로팅과 같은 일반적인 생화확적인 소사에 사용될 수 있습니다. MNOs의 축축번에서 핵 또는 수지상 제조 자 단백질은 서부 블로팅(도 6C)에서검출되지 않습니다.
칼슘 표시기(Fluo-4AM)와 함께 신경 활성을 조직 배양 칩에 포획할 수 있다. 스페로이드 및 축축전의 모터 뉴런의 자발적인 활성은 MNO 내에서 관찰되었다. 또한, 신경 활동은 다중 전극 어레이 시스템을 사용하여 관찰되었다.
