Peyer's patches (PPs) består af hundredvis af lymfoid follikler i lamina propria af tyndtarmen. PPs er opdelt i follikler, den interfollicular region, og germinal centre placeret i den nederste del af folliklerne, hvor lymfocytter stimuleres af antigen præsentation. Der er ingen afferente lymfekar, og antigene invaderer lamina propria fra tarm lumen via epitelcellelaget. Den epitel region, der dækker lymfoid follikler kaldes follikel-associeret epitel, inden for hvilken specialiserede afbrudte M-celler optagelse slimhindeantigener. M celler tage i antigener fra luminal side og antigener er derefter fanget af dendritiske celler og præsenteres mod naive lymfocytter, der strømmer ind i PPs gennem endothelium af høje endotel venules (HEVs)1. PPs spiller en vigtig rolle i tarmimmunitet og er relateret til den tidlige fase af inflammation. Mange molekylære interaktioner involverer indgangen til lymfocytter til sekundære lymfoide organer (SLOs), herunder vedhæftningsmolekyler, kemokiner2,3og sphingosin-1-fosfat4; der er således mange forventede terapeutiske mål. Derfor gør observation af lymfocytdynamikken inden for PPs det muligt for os at få et glimt af det meget tidlige stadium af inflammation og undersøge nytten af flere lovende lægemidler.
Metoden her fokuserer på migration af lymfocytter i PPs, som omfatter flere procedurer (cannulation i brystkanalen5 og indsamling lymfocytter og langsigtet observation efter injektionen i indsamlede lymfocytter). Da disse procedurer er komplekse, og det var vanskeligt at se nøjagtigt, hvordan hver procedure blev udført i tidligere rapporter, nævnte vi her nogle tips til at opnå en vellykket observation. For eksempel var cannulation af rørene i brystkanalen meget vanskelig, og den oprindelige succesrate for cannulation var mindre end 50%. Vi forbedrede imidlertid metoden og opnåede en succesrate på over 80 %. Vi nævnte nogle andre tips i dette manuskript, der er nødvendige for en vellykket observation for at muliggøre den kvantitative evaluering af transendotelmigrationen af lymfocytter under flere forhold.
I tidligere rapporter var det svært at forstå de tredimensionelle ændringer over tid, såsom intravenøs injektion af indisk blæk for at plette den vaskulære struktur af PPs6, eller mikroskopet er monofokal7. I de senere år har en observationsmetode ved hjælp af nogle fotokonvertible fluorescensproteintransgene dyr som Kaede mus afklaret systematiske cellulære bevægelser in vivo8. Den anden undersøgelse præciseret CD69 uafhængig nedlukning af lymfocyt udgang fra PPs9. Vi brugte confocal laserscanning mikroskopi (CLSM) på grund af sin høje analytiske kapacitet. Nu kan vi nemt få billeder i høj opløsning og bruge dem til at analysere lymfocytdynamik.
I denne rapport demonstrerede vi en række metoder til evaluering af lymfocytmigration i PPs. For det første viste vi raffinerede metoder til thoraxkanal cannulation at indsamle lymfocytter. For det andet forbedrede vi observationsmetoderne på flere måder for at opretholde objektive organer, når det var muligt under mikroskopisk observation, så vi kunne få billeder i høj kvalitet i 3 timer. For det tredje kvantificerede vi de cellulære bevægelser af lymfocytmigration for at evaluere virkningerne af nogle medikamenter. Disse ændrede protokoller vil bidrage til udviklingen af slimhinde immunologi evalueringer.

Naive lymfocytter recirkulerer fra blodet til lymfoide væv under fysiologisk tilstand, og det er almindeligt anerkendt som et vigtigt fænomen i tarmens immunitet. Stroma af sekundære lymfoide organer, såsom Peyer's patches (PPs) eller mesenteric lymfeknuder, er, hvor naive lymfocytter forstand antigener.
