Silicon Nanowires and Optical Stimulation for Investigations of Intra- and Intercellular Electrical Coupling

この技術は、標準的な光学顕微鏡のみを用いて、非常に高い空間分解能と細胞特異性を有する細胞内生体電気的尋問を可能にする。この方法は、細胞および細胞内の特定の位置の刺激および電気的尋問を促進し、3D ex vivo組織製剤でだけでなく、インビトロで行うことができる。この技術は、心臓細胞や脳細胞などの多くの細胞アリーナのバイオ電子調査と、すべての非興奮性細胞間の電気的コミュニケーションを可能にします。筋線維芽細胞を筋筋細胞懸濁液から分離するために、100ミリメートルの組織培養皿に分離された細胞を1時間予めプレートし、心筋細胞が付着する繊維をネック化して処理した表面を必要とする。インキュベーションの終了時に、下流共培養用の上清を含む濃縮心筋細胞を収集する。DMEMで筋線維芽細胞をリンスして残りの心筋細胞を排除し、さらに2〜4日間のインキュベーションの前に新鮮な培養培地で細胞を供給する。細胞が80%のコンフルエンシーに達したら、ダイヤモンドの筆記者を使用して、化学気相成長シリコンナノワイヤを持つウエハーから3ミリメートルチップを3%切ります。70%エタノールでチップをすすいるために鋭い鉗子を使用して下さいます。すすい後、バイオセーフティラミナーフローフードの紫外線下で30分間、チップを無菌マイクロ遠心分離管に移す前に空気乾燥させます。完全な細胞培養培地リンスを使用して残りのエタノールを除去し、1〜10分間超音波処理浴中の新鮮な培養培地の1ミリリットルでチップを超音波処理する前に。上清は、ナノワイヤーが放出されると曇るはずです。シリコンナノワイヤー懸濁液を5ミリリットルの新鮮な培養培地に混ぜ、ナノワイヤー溶液をミオ線維芽細胞の皿に種付けします。細胞培養インキュベーターで4時間後、新鮮な培養培地で皿を5回洗い、非内在化したナノワイヤを除去し、さらに1時間細胞をインキュベートし続け、部分的に内在化されたナノワイヤを完全に組み込むことができるようにします。次に、作りたてのコラーゲンコーティング液を500マイクロリットルの35ミリメートルガラス底皿に加えます。摂氏37度で1時間のインキュベーションを行った後、滅菌PBSで皿を洗い上げ、3ミリリットルのトリプシンを3ミリリットルのトリプシンで37°Cで2分間収穫します。細胞が剥離したら、10ミリリットルの培養培地で反応を停止し、ピペット処理により激しくすすぐ、細胞にダメージを与えないように200G以上の遠心分離を行わない。その後、ハイブリッド細胞ペレットを1ミリリットルの新鮮な培地で再中断し、コラーゲンコーティングガラス底皿に細胞を播種する。ナノワイヤの内部化を検証するには、標準的なプロトコルに従って蛍光サイトゾルと膜色素で細胞にラベルを付け、共焦点顕微鏡を使用して細胞を画像化します。シリコンナノワイヤは反射性が高く、反射光を可視化するために蛍光の代わりに使用することができます。筋線維芽細胞シリコンナノワイヤーハイブリッド心筋細胞共培養をセットアップするには、各細胞タイプの適切な数をコラーゲンコーティングガラス底皿に播種します。そして、細胞を摂氏37度で48時間培養し、細胞間ギャップ接合部を形成できるようにした。実験当日、培養上清を、調製したカルシウム感受性染料を1ミリリットルの培地に交換し、摂氏37度で20~30分間培養する。インキュベーションの終わりに、滅菌PBSでプレートを2回洗浄し、プリ温めのフェノールレッドフリーDMEMの1ミリリットルで細胞を処理する前に。その後、細胞内カルシウム色素が37°Cで30分間脱エステル化し、ベースライン画像を取得します。光学イメージングと刺激の前に、加湿したマイクロインキュベーターを顕微鏡に予熱し、光路に連結したレーザーラインを用いて、カルシウムイメージングと光学刺激を摂氏37度と5%気泡時代の二酸化炭素混合物に予熱します。顕微鏡の準備ができたら、ミクロインキュベーターに筋線維芽細胞ハイブリッド心筋細胞共培養液を入れ、明視野顕微鏡でナノワイヤを可視化し、適切な刺激部位を見つけます。サイトが特定されたら、ナノワイヤの事前定義された位置に刺激点を維持しながら蛍光モードへの光路を再構成し、各シリコンナノワイヤーサイズとセルタイプに最適な刺激力とパルス長さを検証します。ベースライン細胞内カルシウム活性の2〜10秒の記録を取得し、1~10メロワットの電力と1〜10ミリ秒の持続時間の単一のレーザーパルスでナノワイヤを刺激する前に、得られたカルシウム波をさらに2〜10秒間記録する。実験の最後に、光学刺激の記録されたムービーを適切なソフトウェアプログラムに移し、さらなる分析を行う。標準的な位相コントラスト光学を使用して、合流細胞は光顕微鏡によって容易に見られる。しかし、シリコンナノワイヤーはほとんど見えないので、この方法ではその位置を定義することは不可能です。しかし、光と暗いフィールド画像のスーパー面付けは、光反射シリコンナノワイヤーのペリー核配置の視覚化を可能にする。共焦点顕微鏡を用いて、蛍光マーカー染色されたサイトゾルや細胞質膜を可視化し、シリコンナノワイヤの細胞内位置をより明白にすることができる。標準的な顕微鏡を用いて、明視野画像を得て、刺激されるシリコンナノワイヤの位置を特定することができる。自発的に鼓動する心筋細胞と休息中の筋線維芽細胞の短いベースラインビデオを取得することができます。刺激後、共培養中のカルシウム伝播を記録することができる。異なるセルが励起される時間は、平均オプティカルフローの変化がその最大値に達した時点で決定できます。オプティカルフローは、各セル領域内の活性化時間の同定に役立つ計算が可能です。細胞内速度は、細胞内伝搬速度の逆を表す線の傾きを用いて、ハイマグラフを使用して細胞ごとに決定することができる。例えば、ここでは、単一の共培養内の細胞相互作用について測定された異なる細胞間および細胞内速度の要約が観察できる。この方法は、細胞シリコンハイブリッドを直接組織に注入し、生体内電気会社を研究するための3D ex vivo製剤を使用することにより、インビボ研究に利用することができる。
