1. 建立和维护胚胎收集笼进行活体分析21

3. 将胚胎安装到盖玻片上
<ol><li>在配备头顶鹅颈照明的立体显微镜下，使用鸡蛋采摘工具在葡萄盘的干净部分组织两排15-20个胚胎。将胚胎在腹侧以相同的方向与前端和后端对齐。使采蛋机将细尖不锈钢钳子弯曲到 45° 角。要查看D/V胚胎的取向，见坎波斯-奥尔特加等人20。 注：对于腹体表面来说，坐在葡萄板上很重要，因为胚胎将被压在步骤3.5的盖滑上，露出后表面进行成像。由于后侧表面比腹体表面更平坦，因此成像后侧表面会增加单个 Z 平面内存在的核数。</li>
	<li>准备 75 mm x 25 mm 硅胶垫片。 注意：可以提前准备。准备多个盖玻片，并将其存放在幻灯片盒中，以保持其清洁。<ol>
<li>将 75 mm x 25 mm 盖玻片的轮廓追踪到 1.6 mm 厚的硅胶垫片上，然后用剪刀切出硅胶垫片（图 2A）。</li>

</li>

4. 脱盐和微缩胚胎
<ol><li>将带胚胎的盖玻片放在滑梯上，以防止盖玻片底部在干燥和微注射过程中变脏。 注：将成像封面滑轨，而不附加幻灯片。因此，倒置显微镜是成像所必需的。</li>
	<li>干燥胚胎5-10分钟，视房间温度不同。用于在代表性结果中成像的胚胎在20°C下在含有1：1层硅胶珠的腔室中干燥7分钟（图3A）。</li>
	<li>干燥后立即用1：1覆盖胚胎 - 卤化油700：卤化油27。将硅胶垫片填到顶部的一半。 注：将高粘度卤化碳油与低粘度卤化碳油混合非常重要。纯卤化碳油 700 （高粘度） 减缓细胞循环进展 30 分钟后应用。卤化油700和卤化碳油27的1：1混合物修复了这一神器。</li>
	<li>用注射剂装载微注射针，然后将一根注射针安装到微注射器上，然后在压力下将其切开。 注：使用任何有价值的试剂进行完整检测之前，先练习步骤4.4。这是微注射过程中最困难的一步。<ol>
<li>使用延长的加载尖端用 2 μL 注射剂加载微注射针。只需要一个微注射针，但好有备用加载的针附近，所以准备2或3针。 注：有关扩展 装载技巧的 规格，请参阅材料表。微注射针不能加载没有延长加载提示，因为我们的微注射技术的独特性质，它依赖于建筑空气压力内密封和加载针之前打开其尖端。</li>
		<li>将针头安装到微注射器上，将柱塞压到尖端的一半。目标是增加玻璃毛细管内部的气压（图4A）。 注：使用柱塞矿物油为基础的微反应器，不带矿物油。用矿物油制造压力，而不是使用柱塞来制造气压。</li>
		<li>将玻璃针放到玻璃滑梯上，用新的剃须刀将针尖#9开。以 45° 角切割针头，以产生锋利的开刀尖。注射剂应向到尖端底部，而不流出针头。小心地在针头上加入 20 μL 的卤化油，然后以 45° 重新切割。注射剂应开始快速流动。</li>
		<li>立即通过缩回柱塞降低气压，但确保保持连续的流速，以防止针头在注射之间用胚胎膜堵塞。（图4B） 注：如果注射剂是明确的，通过将微注射针提起和提起卤化碳油，并观察立体显微镜下注射液滴的形成速率，查看注射液流速。通过一次压压柱塞几下，然后浸入和浸出卤化碳油的微注射尖端，以查看新液滴的形成速率，然后进行调节。</li>

<ol start="5"><li>微基因胚胎<ol>

5. 在高含量分析仪上成像
