En alternativ HVI er utviklet, ideell for å utføre SX-eksperimenter ved synchrotron kilder. Den har to viktige fordeler fremfor eksisterende HVIer. For det første er det enkelt å installere på strålelinjen slik at rask veksling mellom konvensjonell krystallografi og SX, bare ~ 30 minutter er nødvendig for installasjon og justering på MX2. For det andre kan prøvesprøytene som brukes til å dyrke krystaller, brukes direkte som reservoarer for injeksjon, noe som begrenser svelhet under prøveoverføring. Protokoll...

Seriekrystallografi (SX) er en teknikk som ble utviklet i utgangspunktet i sammenheng med røntgenfrie elektronlasere (XFELs)1,2,3,4. Selv om faste måltilnærminger kan brukes til SX5,6,7, brukes vanligvis injektorsystemer til å levere krystaller i en kontinuerlig strøm til røntgenstrålen. Fordi den kombinerer data fra et stort antall krystaller, unngår SX behovet for krystalljustering under eksperimentet, og gjør det mulig å samle inn data ved romtemperatur8,9. Ved hjelp av en egnet injektor strømmer krystallene en etter en inn i røntgeninteraksjonsområdet, og de resulterende diffraksjonsdataene samles inn på enområdedetektor 9,10. Hittil har SX vært vellykket i å løse en rekke proteinstrukturer1,11,12,13 inkludertkrystaller for små til å måle ved hjelp av konvensjonell krystallografi. Det har også gitt ny innsikt i tidsløs molekylær dynamikk ved å utnytte femtosekunders pulsvarighet for XFEL. Ved å initiere pumpe-sonde reaksjoner med optiske laserkilder, grundige studier har blitt utført på fotosystem II14,15, fotoaktivt gult protein16,17, cytokrom C oksidase18, samt bakteriorhodopsin19,20,21. Disse studiene har undersøkt elektronoverføringsdynamikken som oppstår etter lysaktivering som viser det betydelige potensialet for seriekrystallografi for å forstå tidsavklarte biologiske prosesser.
Utvikling av seriekrystallografi blir også stadig mer utbredt ved synchrotron kilder9,12,20,22,23,24. Synchrotron basert SX gjør det mulig for et stort antall individuelle krystaller som skal måles effektivt ved romtemperatur ved hjelp av et passende injektorsystem. Denne tilnærmingen er egnet for mindre krystaller derav, i tillegg til å kreve en rask bildefrekvensdetektor for å samle inn dataene, er det også nødvendig med en mikrofokusert stråle. Sammenlignet med konvensjonell krystallografi, involverer SX ikke montering og justering av individuelle krystaller i røntgenstrålen. Fordi data fra et stort antall individuelle krystaller slås sammen, kan strålingsdosen mottatt av hver krystall reduseres betydelig sammenlignet med konvensjonell krystallografi. Synchrotron SX kan også brukes på studiet av tidsoppsagte reaksjoner, selv ned til millisekundregimet, forutsatt at en detektor med tilstrekkelig høy bildefrekvens er tilgjengelig (f.eks. 100 Hz eller mer). Flere seriell krystallografi eksperimenter har blitt utført på synchrotron ved hjelp av injektorer som opprinnelig ble utviklet på XFELkilder 20,22,23. De to vanligste typene injektor er gass dynamisk virtuell dyse (GDVN)25 og høy viskøs injektor (HVI)9,24,26,27,28. GDVN er ideell for å injisere lav viskositet, flytende prøver, men krever høye strømningshastigheter for å oppnå stabile bekker, noe som igjen fører til høye utvalgsforbruksrater. HVI-er er derimot egnet for høyviskositetsprøver som gjør det mulig å lage en stabil strøm ved mye lavere strømningshastigheter, noe som fører til mye lavere utvalgsforbruk. HVI-injektoren favoriserer derfor levering av prøver der en viskøs transportør er å foretrekke (f.eks. lipidbasert for membranproteiner) og/eller store mengder prøve er ikke tilgjengelige. SX injektorer er generelt utfordrende å bruke og krever omfattende opplæring for å operere. De involverer også lange prøveoverføringsprotokoller, da prøven må lastes inn i et spesialisert reservoar, dette har generelt en høy risiko forbundet med at prøven går tapt enten i "dødt volum" eller via lekkasjer i forbindelsene. Derfor er det ønskelig å optimalisere injektordesignen for å redusere eventuelle tap før prøven når røntgenstrålen.
Nylig ble de første SX-resultatene publisert ved hjelp av Lipidico23 med et lysozymmål, ved hjelp av en Eiger 16M detektor. Denne injektordesignen begrenser prøvesvinn ved å minimere antall skritt involvert i å gå fra første krystallisering til overføring av krystaller til injektoren etterfulgt av levering av prøve til røntgenstrålen. Dette manuskriptet beskriver og demonstrerer prøveoverføringsprosedyren fra prøveforberedelse, går videre til injeksjonsprosessen, og til slutt datainnsamling, ved hjelp av samme krystalliseringsfartøy. Et anlegg for å utføre seriekrystallografimålinger er utviklet ved den australske synchrotron. Dette anlegget inneholder en spesialbygd høy viskøs injektor, Lipidico, som en del av den makromolekylære krystallografien (MX2) strålelinjen for å måle et stort antall små krystaller ved romtemperatur. Målet med denne teknikken er å gjøre det mulig for krystaller å bli dyrket/ overført til glasssprøyter som skal brukes direkte i injektoren for seriell krystallografi datainnsamling. Fordelene med denne injektoren inkluderer evnen til å reagere raskt på endringer i strømningshastigheten uten avbrudd i strømmen. Flere begrensninger for denne høyviskositetinjektoren (HVI) finnes som inkluderer en begrensning på de tillatte prøveviskositetene til > 10 Pa.s. Stream stabilitet kan også potensielt være et problem avhengig av de spesifikke egenskapene til utvalget. Lipidico er et HVI bygget som et alternativt leveringssystem for bruk på MX2 (figur 1). Den er ideell for SX hvor krystaller enten dyrkes i lipidisk kubikkfase eller overføres til et høyt viskøs inert media.
