Western blot is een veel gebruikte analytische techniek om specifieke eiwitten te detecteren die ~40 jaar geleden werd ontwikkeld7,8. Sindsdien is de techniek verder geëvolueerd, waarbij latere innovaties de gevoeligheid, snelheid en kwantificering van de techniekverbeterden 2,9,10,11,12,<s...

Een westerse vlek (ook bekend als een immunoblot) is een krachtige en fundamentele techniek in een breed scala aan wetenschappelijke en klinische disciplines. Deze techniek wordt gebruikt om de aanwezigheid, relatieve overvloed, relatieve moleculaire massa en post-translationele modificaties van eiwitten te onderzoeken1. In combinatie met digitale beeldanalyse kan deze methode op betrouwbare wijze de overvloed aan eiwitten en eiwitmodificaties analyseren2. Hoewel western blot routinematig wordt uitgevoerd, is het een tijdrovende en arbeidsintensieve methode. Lange incubaties van het antilichaam met membranen zijn vereist. Hier beschrijven we een wijziging van de incubatiemethode die deze beperking overwint zonder de gevoeligheid op te offeren.
Tijdens incubatie van het membraan zweven antilichamen in oplossing terwijl antigenen op het membraan worden geïmmobiliseerd. Vanwege hun hoge affiniteit is de snelheid van antilichaambinding aan het antigeen sneller dan de diffusie van antilichamen van de bulkoplossing naar het membraan. Hierdoor ontstaat een lage concentratie "uitputtingslaag" (figuur 1). Het kan uren duren voordat meer verre antilichamen het membraan bereiken via passieve diffusie, wat de belangrijkste factor is die verantwoordelijk is voor lange incubatietijden in deze techniek. Dit effect wordt de massatransportbeperking (MTL)3genoemd. Er is voorgesteld dat het herhaaldelijk aftappen en aanvullen van de antilichaambevattende oplossing de uitputtingslaag kan verstoren en MTL4kan overwinnen . Hier hebben we een unieke techniek ontwikkeld die dit cyclische draining and replenishing (CDR) concept implementeert om het effect van MTL in een traditioneel westers blotting protocol te verminderen en de vereiste incubatietijd te verkorten.
Om de detectiegevoeligheid met een korte incubatietijd te behouden, hebben we gebruik gemaakt van een immunoreactieverhogende technologie die de antigeen-antilichaamreactie versnelt, waardoor de signaal-ruisverhouding5wordt verbeterd. Veel in de handel verkrijgbare immunoreactieverhogende middelen (IRE) bestaan uit 2 componenten (oplossing 1 en 2, zie tabel met materialen). De eigen samenstelling of werkingsmechanisme van IRE is niet bekendgemaakt, maar we hebben eerder vastgesteld dat IRE de dissociatieconstante tussen het antigeen en het antilichaam in vastefasebindende tests heeft verminderd, wat erop wijst dat verhoogde affiniteit ten minste gedeeltelijk verantwoordelijk is voor het versterkende effect van IRE6. De combinatie van CDR met IRE levert een ultrasnel westers vlekkenprotocol op dat de hele proceduretijd vermindert zonder in te boeten aan gevoeligheid.

Een westerse vlek (ook wel immunoblot) is een canonieke methode voor biomedisch onderzoek. Het wordt vaak gebruikt om de relatieve grootte en overvloed van specifieke eiwitten te bepalen, evenals post-translationele eiwitaanpassingen. Deze techniek heeft een rijke geschiedenis en wordt nog steeds op grote schaal gebruikt vanwege de eenvoud. De westelijke vlekprocedure duurt echter uren, zelfs dagen, om te voltooien, met een kritiek knelpunt zijn de lange incubatietijden die de doorvoer beperken. Deze incubatiestappen zijn nodig vanwege de langzame diffusie van antilichamen van de bulkoplossing naar de geïmmobiliseerde antigenen op het membraan: de antilichaamconcentratie in de buurt van het membraan is veel lager dan de bulkconcentratie. Hier presenteren we een innovatie die deze incubatie-intervallen drastisch vermindert door de antigeenbinding te verbeteren via cyclische afvoer en aanvulling (CDR) van de antilichaamoplossing. We gebruikten ook een immunoreactieverhogende technologie om de gevoeligheid van de test te behouden. Een combinatie van de CDR-methode met een commercieel immunoreactieverhogend middel verhoogde het uitgangssignaal en verminderde de incubatietijd van antilichamen aanzienlijk. De resulterende ultrasnelle westerse vlek kan in 20 minuten worden bereikt zonder verlies van gevoeligheid. Deze methode kan worden toegepast op westerse vlekken met behulp van zowel chemiluminescente als fluorescerende detectie. Dit eenvoudige protocol stelt onderzoekers in staat om de analyse van eiwitexpressie in veel monsters beter te onderzoeken.

Dit voorbeeld illustreert de effectiviteit van de CDR-methode samen met immuno-fenhancingtechnologie op western blot. Statische incubatie werd uitgevoerd op een semi-transparante flexibele film waar PVDF-membranen werden geïncubeerd met de antilichaamoplossing, terwijl cdr-incubatie zich in een hybridisatieoven bevond door roterende buizen die de membranen en de antilichaamoplossing bevatten (Movie).
