Une tache occidentale (également connue sous le nom d’immunoblot) est une technique puissante et fondamentale dans un large éventail de disciplines scientifiques et cliniques. Cette technique est utilisée pour examiner la présence, l’abondance relative, la masse moléculaire relative et les modifications post-translationnelles des protéines1. Combinée à l’analyse d’images numériques, cette méthode peut analyser de manière fiable l’abondance des protéines et des modificationsprotéiques 2. Bien que la tache occidentale soit exécutée régulièrement, c’est une méthode longue et laborique. De longues incubations de l’anticorps avec membranes sont nécessaires. Ici, nous décrivons une modification de la méthode d’incubation qui surmonte cette limitation sans sacrifier la sensibilité.
Pendant l’incubation de la membrane, les anticorps flottent en solution tandis que les antigènes sont immobilisés sur la membrane. En raison de leur affinité élevée, le taux d’anticorps se liant à l’antigène est plus rapide que la diffusion d’anticorps de la solution en vrac à la membrane. Cela crée une « couche d’épuisement » à faible concentration (figure 1). Il peut prendre des heures pour que des anticorps plus éloignés atteignent la membrane par diffusion passive, qui est le principal facteur responsable des longs temps d’incubation dans cette technique. Cet effet est appelé la limitation de transport de masse (MTL)3. Il a été proposé que le drainage répétitif et le réapprovisionnement de la solution contenant des anticorps puissent perturber la couche d’épuisement et surmonter le MTL4. Ici, nous avons conçu une technique unique qui met en œuvre ce concept cyclique de drainage et de réapprovisionnement (CDR) pour diminuer l’effet du MTL dans un protocole traditionnel de ballonnement occidental et raccourcir de façon remarquée la période d’incubation requise.
Afin de maintenir la sensibilité à la détection avec un temps d’incubation court, nous avons utilisé une technologie d’amélioration de l’immunoréaction qui accélère la réaction antigène-anticorps, améliorant ainsi le rapport signal-bruit5. De nombreux agents d’amélioration de l’immunoréaction disponibles dans le commerce (IRE) se composent de 2 composants (solution 1 et 2, voir tableau des matériaux). La composition propriétaire ou le mécanisme d’action de l’IRE n’a pas été divulgué, mais nous avons déjà constaté que l’IRE a diminué la constante de dissociation entre l’antigène et l’anticorps dans les essais de liaison de phase solide, indiquant que l’affinité accrue est, au moins en partie, responsable de l’effet d’amélioration de l’IRE6. La combinaison du CDR et de l’IRE donne un protocole de ballonnement occidental à très grande vitesse qui réduit tout le temps de procédure sans sacrifier la sensibilité.

Une tache occidentale (également connue sous le nom d’immunoblot) est une méthode canonique pour la recherche biomédicale. Il est couramment utilisé pour déterminer la taille relative et l’abondance de protéines spécifiques ainsi que les modifications des protéines post-translationnelles. Cette technique a une histoire riche et reste largement utilisée en raison de sa simplicité. Cependant, la procédure occidentale de ballonnement prend des heures, voire des jours, pour terminer, avec un goulot d’étranglement critique étant les longs temps d’incubation qui limitent son débit. Ces étapes d’incubation sont nécessaires en raison de la lente diffusion des anticorps de la solution en vrac aux antigènes immobilisés sur la membrane : la concentration d’anticorps près de la membrane est beaucoup plus faible que la concentration en vrac. Ici, nous présentons une innovation qui réduit considérablement ces intervalles d’incubation en améliorant la liaison antigène par drainage cyclique et réapprovisionnement (CDR) de la solution d’anticorps. Nous avons également utilisé une technologie d’amélioration de l’immunoréaction pour préserver la sensibilité de l’essai. Une combinaison de la méthode CDR avec un agent d’amélioration de l’immunoréaction commerciale a stimulé le signal de sortie et considérablement réduit le temps d’incubation des anticorps. La tache occidentale à très grande vitesse qui en résulte peut être réalisée en 20 minutes sans aucune perte de sensibilité. Cette méthode peut être appliquée aux taches occidentales en utilisant à la fois la détection chemiluminescente et fluorescente. Ce protocole simple permet aux chercheurs de mieux explorer l’analyse de l’expression des protéines dans de nombreux échantillons.

Cet exemple illustre l’efficacité de la méthode CDR ainsi que la technologie d’immunoenhancing sur la tache occidentale. L’incubation statique a été effectuée sur un film flexible semi-transparent où les membranes PVDF ont été incubées avec la solution d’anticorps, tandis que l’incubation du CDR était dans un four d’hybridation par des tubes rotatifs qui contenaient les membranes et la solution d’anticorps (Movie).
