Hier presenteren we een praktisch en eenvoudig protocol om subpopulaties van spermatocyten en spermatids te isoleren van een enkele volwassen mannelijke muis. Om de reproduceerbaarheid van dit protocol te waarborgen, zijn er enkele kritieke stappen die aandacht nodig hebben. Vóór enzymvertering, wasstap is gericht op het verwijderen van interstitiële cellen; na de spijsvertering, deze stap helpt om spermatozoa en puin te verwijderen. Spermatogenese is een complex proces waarbij een kleine populatie spermatogonale stamcellen een continue productie van een groot aantal sperma gedurende het hele volwassen leven gedurende1,2. Tijdens spermatogenese vindt dynamische chromatine remodellering plaats wanneer spermatogene cellen meiose ondergaan om haploïde spermatids3,4,5te produceren . Isolatie van meiotische spermatocyten is essentieel voor moleculair onderzoek en er zijn verschillende benaderingen vastgesteld om meiotische zaadcyten te isoleren, waaronder scheiding op basis van sedimentatie6,7 en fluorescentie-geactiveerde celsordening (FACS)8,9,10,11,12,13,14,15,16,17. Deze methoden hebben echter technische beperkingen. Terwijl sedimentatie gebaseerde scheiding levert een groot aantal cellen5,6,7, het is arbeidsintensief. De gevestigde FACS-gebaseerde methode maakt gebruik van Hoechst 33342 (Ho342) om meiotische zaadtomen te scheiden op basis van DNA-inhoud en lichtverstrooiingseigenschappen en vereist FACS-celsorteerders die zijn uitgerust met een ultraviolette (UV) laser8,9,10,11. Alternatieve FACS-gebaseerde methoden vereisen transgene muislijnen die fluorescerende eiwitten uitdrukken, synchronisatie van spermatogenese12, of celfixatie en antilichaamlabeling die niet compatibel is met isolatie van levende cellen13. Hoewel er een andere alternatieve methode met behulp van een cel-permeabele DNA-bindende kleurstof, DyeCycle Groene vlek14,15,16,17, deze methode wordt aanbevolen voor de isolatie van spermatogene cellen van jeugdige testis. Daarom is er een kritische behoefte om een eenvoudige en robuuste isolatiemethode te ontwikkelen voor levende meiotische spermatocyten die kunnen worden toegepast op elke muisstam van elke leeftijd en die kan worden uitgevoerd met behulp van een FACS-celsorteerder.
Hier beschrijven we zo'n lang gezocht celisolatieprotocol met behulp van de DyeCycle Violet (DCV) vlek. DCV is een lage cytotoxiciteit, cel-permeabele DNA bindende kleurstof structureel vergelijkbaar met Ho342, maar met een excitatie spectrum verschoven naar de violette bereik18. Daarnaast heeft DCV een breder emissiespectrum in vergelijking met DCG. Zo kan het worden opgewekt door zowel UV-en violet lasers, die de flexibiliteit van de apparatuur verbetert, waardoor het gebruik van een FACS cel sorteerder niet uitgerust met een UV-laser. Het DCV-protocol dat hier wordt gepresenteerd, maakt gebruik van tweedimensionale scheiding met DCV-blauw en DCV-rood, waarbij het voordeel van het Ho342-protocol wordt nagebootst. Met dit voordeel stelt ons DCV-protocol ons in staat om sterk verrijkte kiemcellen te isoleren van de volwassen testis. We bieden een gedetailleerd gating protocol om levende spermatogene cellen te isoleren van volwassen muispesten van één muis (van twee teelballen). We beschrijven ook een efficiënt en snel protocol voor de voorbereiding van eencellige suspensie van muistestes die kunnen worden gebruikt voor deze celisolatie. De procedure vereist een korte tijd om te voltooien (voorbereiding van eencellige suspensie - 1 uur, kleurstof - 30 min, en celsorde - 2-3 uur: totaal - 4-5 uur, afhankelijk van het aantal benodigde cellen; Figuur 1). Na celisolatie kan een breed scala aan downstreamtoepassingen, waaronder RNA-seq, ATAC-seq, ChIP-seq en celcultuur, worden voltooid.

Isolatie van meiotische spermatocyten is essentieel om moleculaire mechanismen te onderzoeken die ten grondslag liggen aan meiose en spermatogenese. Hoewel er gevestigde celisolatieprotocollen zijn met Hoechst 33342-kleuring in combinatie met fluorescentie-geactiveerde celsorling, vereist het celsorteerders die zijn uitgerust met een ultraviolette laser. Hier beschrijven we een celisolatie protocol met behulp van de DyeCycle Violet (DCV) vlek, een lage cytotoxiciteit DNA binding kleurstof structureel vergelijkbaar met Hoechst 33342. DCV kan worden opgewekt door zowel ultraviolette als violet lasers, die de flexibiliteit van de apparatuur keuze verbetert, met inbegrip van een cel sorteerder niet uitgerust met een ultraviolette laser. Met behulp van dit protocol kan men drie live-cel subpopulaties isoleren in meiotische profase I, waaronder leptotene/zygotene, pachytene en diplotene spermatocyten, evenals post-meiotische ronde spermatids. We beschrijven ook een protocol voor een eencellige suspensie van muistestes. Over het algemeen vereist de procedure een korte tijd om te voltooien (4-5 uur, afhankelijk van het aantal benodigde cellen), wat veel downstreamtoepassingen vergemakkelijkt.

Een representatief resultaat van dit sorteerprotocol wordt weergegeven in figuur 3. De totale sorteertijd van twee teelballen (één muis) is meestal ongeveer 3 uur, wat afhankelijk is van de concentratie van celsuspensie en de sorteersnelheid. Hier presenteren we een eenvoudige en efficiënte methode om levende meiotische en post-meiotische kiemcellen te isoleren van volwassen muispesten. Met behulp van een lage cytotoxiciteit, violet-opgewonden DNA bindende kleurstof en fluorescentie-geactiveerde celsorde, kan men zeer verrijkte spermatogene celpopulaties isoleren voor vele downstream toepassingen.

Deze nieuwe efficiënte methode voor het isoleren van subpopulaties van spermatocyten en spermatids van volwassen muizen kan worden gebruikt om de moleculaire mechanismen die ten grondslag liggen aan meiose en spermatogenese te onderzoeken. Deze methode maakt gebruik van een lage cytotoxiciteit DNA bindende kleurstof met een breed excitatiespectrum dat kan worden toegepast op de meeste momenteel gebruikte FACS orders. Deze methode is zeer eenvoudig.Het meest uitdagende aspect is de stroom cytometrie gating, maar de gedetailleerde gating strategie moet helpen bij het aanpassen van het protocol aan andere cellen sorteerders. Het aantonen van de procedure zal Yu-Han Yeh, een onderzoeksassistent van Satoshi Namekawa's Laboratory. Na het oogsten van beide teelballen van een acht weken oude mannelijke muis, plaats de weefsels in een 60 millimeter petrischaal met twee milliliter ijskoude PBS en verwijder de tunica albuginea.Scheid de teelballen voorzichtig met tangen om de halfniferous tubuli lichtjes te verspreiden. En breng de tubuli in een druppel verse ijskoude PBS in een 100 millimeter petrischaal. Gebruik de tangen om de tubuli voorzichtig te ontwarren en de tubuli te wassen in drie extra druppels PBS zoals net is aangetoond.Na de laatste wasbeurt, plaats de ontwarrende halfniferous tubuli in een 15 milliliter buis, met twee milliliter dissociatie buffer voor een 20-minuten incubatie bij 37 graden Celsius. Gebruik aan het einde van de incubatie een pipet van 1000 microliter om de tubuli 20 keer voorzichtig te pipetten, voordat u de buis nog eens zes minuten terugzet naar de couveuse. Aan het einde van de incubatie, pipet de weefsels een extra 20 keer, alvorens de buis terug te keren naar de incubator.Na drie minuten, voorzichtig pipette de tubuli 10 keer of totdat er geen zichtbare weefsel stukken blijven, alvorens het stoppen van de dissociatie met 10 milliliter FACS buffer. Vervolgens, centrifuge naar het weefsel suspensie twee keer met behulp van een extra 10 milliliter voor verse FACS buffer voor de tweede wasbeurt om de spermatozoa en zo veel mogelijk puin te verwijderen. Om de cellen te bevlekken voor stromingscytometrische analyse, schort u de pellet opnieuw op in drie milliliter FACS-buffer.En filter de celopening door een 70 micron nylon celzeef, in een buis van 50 milliliter. Na het tellen, breng 10% van de cellen in een nieuwe buis op ijs als de onbevlekte negatieve controle. En meng grondig zes microliters van DCV vlek in de resterende celsuspensie.Na een 30 minuten durende incubatie bij 37 graden Celsius in het donker met zachte schudden om de 10 minuten, voeg vijf microliters van DNase I aan de cellen. En filter de cellen door een 35 micron nylon mesh zeef in een vijf milliliter FACS buis op ijs. Voor de cytometrische analyse van de cellen voor stroomcytometrische analyses maakt u een nieuw experiment in de stroomanalysesoftware en klikt u onder het menu werkbladgereedschappen op nieuwe dichtheid om een forward scatter area versus backscatter area density plot op een lineaire schaal te maken.Klik op nieuw histogram om een DCV-blauw histogramplot op een logaritmische schaal te maken. En klik op start en record om te beginnen met het verwerken van het niet-opgenomen monster. Terwijl het monster wordt uitgevoerd, klikt u op detector- en drempelinstellingen om zowel de PMT-spanningen voor heen als aan de zijkant aan te passen om de niet-gekleurde cellen op de schaal van het voor- en achterstuiverplot te plaatsen.Pas de PMT-spanning voor het jappy-kanaal aan om de positie van de DCV-negatieve populatie in het eerste decennium van de DCV-blauwe histogramlogaritmische plot te lokaliseren. Klik vervolgens op stop om het niet-bevlekte monster te lossen. Nadat u het bevlekte monster kort hebt gevorkt en geladen, klikt u op de volgende buis om een nieuw werkblad voor het monster te maken.Stel de cytometer in om ten minste één keer 10 te verwerven op de zes gebeurtenissen en klik op start en record. Klik vervolgens op nieuwe dichtheid om voorwaartse spreidingshoogte versus voorwaartse spreidingsbreedte te maken. En DCV blauw versus DCV rode dichtheid percelen op een lineaire schaal.Gebruik op het voorste verspreidingsgebied versus backscatter gebiedsdichtheidsperceel het gereedschap veelhoek om een poort te tekenen die cellen wordt genoemd om de meeste cellen op te nemen en om kleine brokstukken uit te sluiten. Pas deze poort toe op de voorwaartse spreidingshoogte versus voorwaartse spreidingsbreedte en gebruik het rechthoeksgereedschap om de poort van een enkele cel te tekenen om niet-enkele cellen uit te sluiten. Pas de poort van de enkele cel toe op het dcv-blauwe versus DCV-rode dichtheidsplot en pas de schaal aan om een uitgebreid profiel vast te leggen.Gebruik het gereedschap veelhoek om een DCV-poort te tekenen om de niet-gekleurde cellen en zijpopulatie uit te sluiten en de poort toe te passen op een DCV-blauw histogramplot op lineaire schaal. De drie grote pieken verwijzen naar de verschillende 1C, 2C en 4C DNA-inhoud. Maak een tweede DCV-blauw versus DCV-plot met rode dichtheid op een lineaire schaal.En achter poort de DCV poort op het perceel om de 1C en 4C populaties te lokaliseren. Maak een ander DCV-blauw versus DCV-plot met rode dichtheid op een lineaire schaal. En gebruik de ellips tool om een poort te trekken op de 1C bevolking.Gebruik het veelhoekgereedschap om de 4C-populatie te bek toegangspoortigen met een continue curve. En maak een ander DCV blauw versus DCV rode dichtheid plot op een lineaire schaal. Gebruik het veelhoekgereedschap om een 4C one gate te maken.In een nieuwe voorwaartse strooi plot, gebruik maken van de ellips tool om pachytene, diplotene en leptoteen, zygotene spermatocyte poorten te creëren. Wanneer alle poorten zijn getekend, maak een nieuwe DCV blauw versus DCV rode kleur stip plot op een lineaire schaal om de voor 4C poort toe te passen om ervoor te zorgen dat de drie populaties zijn in een continue volgorde binnen de poort. Maak vervolgens een nieuw voorwaarts spreidingsgebied versus backscattergebied dichtheid plot op een lineaire schaal, en selecteer de uniforme grootte van cellen als een zuivere ronde spermatid populatie.Na het sorteren liggen de representatieve zuiverheid van het gescheiden leptoteen, zygotene, pachytene en diplotene spermatocytenfracties meestal tussen de 80%tot 90%, zoals bevestigd door SYCP3 en Gamma H2AX immunostaining. De zuiverheid van de ronde spermatid factie kan worden bevestigd door nucleus kleuring met DCV in combinatie met Sp56 en H1t kleuring, en is ook meestal rond 90%De levensvatbaarheid van de geïsoleerde cel populaties is meestal meer dan 95%En de gemiddelde totale opbrengsten van elke fractie van een enkele volwassen muis is meestal voldoende voor verschillende downstream analyse. De gesorteerde cellen kunnen worden gebruikt voor verschillende next generation sequencing analyse om genexpressie en regulatie tijdens spermatogenese beter te verkennen.
