Choroidale angiogenese dysregulatie wordt geassocieerd met neovasculaire leeftijdsgebonden maculadegeneratie (AMD)1. De choroïde is een microvasculair bed aanwezig onder het retinale pigment epitheel (RPE). Het is aangetoond dat verminderde bloedstroom in de choroid wordt geassocieerd met progressie van AMD2. De ingewikkelde relatie tussen vasculair endotheel, RPE, macrofagen, pericyten en andere cellen is verantwoordelijk voor de homeostase van het weefsel3,4,5. Daarom is een reproduceerbare test modellering choroidale micro-omgeving is van cruciaal belang voor de studie van neovasculaire AMD.
Ex vivo angiogenese testen en in vitro endotheelcelculturen kunnen studies naar microvasculair gedrag in vivo aanvullen, voor het testen van nieuwe geneesmiddelen en voor studies van pathogenese. Endotheelcellen zoals menselijke retinale microvasculaire endotheelcellen (HRMECs), Human Umbilical Vein Endothelial Cells (HUVEC), geïsoleerde primaire dierlijke hersenen of retinale VC's worden vaak gebruikt in vitro studies voor oculair angiogenese onderzoek6,7,8. Met name HRMECs zijn op grote schaal gebruikt als een model van in vitro choroidale neovascularisatie (CNV)9 door de beoordeling van endotheelproliferatie, migratie, buisvormige vorming en vasculaire lekkage om interventies6,10te evalueren . ECs in de cultuur zijn echter beperkt als model van CNV vanwege het gebrek aan interacties met andere celtypen in de chooïde en omdat de meeste EG-maatregelen die in deze tests worden gebruikt, niet afkomstig zijn van chooïde. Muis choroidale ECs zijn moeilijk te isoleren en te onderhouden in de cultuur.
De aorta ring test wordt veel gebruikt als een model van macro vasculaire proliferatie. Vasculaire spruiten van aorta-explants zijn onder andere ECs, pericyten en macrofagen11. De aortaringtest modelleert grote scheeps angiogenese goed12,13,14. Echter, het heeft beperkingen als een model van choroïdale neovascularisatie als aorta ringen zijn een macrovasculair weefsel ontbreekt de karakteristieke choroidale microvasculaire omgeving, en spruiten uit grote vaten kunnen verschillen van spruiten van capillaire netwerken die betrokken zijn bij microvasculaire pathologie. Onlangs publiceerde een groep een ex-vivo retinale test15,16. Hoewel het geschikt is voor retinale neovasculaire ziekte, is het niet zo geschikt voor choroidale neovascularisatie zoals gezien in AMD.
De choroidale kiemende test met muis RPE, choroïde en scleral explanted tissue werd ontwikkeld om CNV beter te modelleren. Het weefsel kan gemakkelijk worden geïsoleerd van de muis (of andere soorten) ogen17. Deze test maakt reproduceerbare evaluatie van pro- en anti-angiogeen potentieel van farmacologische verbindingen en evaluatie van de rol van specifieke trajecten in choroidale neovascularisatie met behulp van weefsel van genetisch gemodificeerde muizen en controles18. In veel latere publicaties 9 , 10,18,19,20. Hier wordt de methode die betrokken is bij het gebruik van deze test aangetoond.

Pathologische choroidale angiogenese, een treffend kenmerk van leeftijdsgebonden maculadegeneratie, leidt tot gezichtsstoornissen en blindheid. Endotheelcel (EC) proliferatietesten met behulp van menselijke retinale microvasculaire endotheelcellen (HRMECs) of geïsoleerde primaire retinale EC's worden veel gebruikt in vitro modellen om retinale angiogenese te bestuderen. Echter, isoleren van zuivere murine retinale endotheelcellen is technisch uitdagend en retinale ECs kunnen verschillende proliferatie reacties dan choroidale endotheelcellen en verschillende cel / cel interacties. Een zeer reproduceerbare ex vivo choroidale kiemen test als een model van choroidale microvasculaire proliferatie werd ontwikkeld. Dit model omvat de interactie tussen choroïde vasculatuur (EC, macrofagen, pericyten) en retinale pigment epitheel (RPE). Muis RPE/choroïde/scleral explants worden geïsoleerd en geïncubeerd in groeifactor-verminderde basale membraan extract (BME) (dag 0). Medium wordt om de andere dag veranderd en choroid ontkiemen wordt gekwantificeerd op dag 6. De beelden van individuele choroid explant worden genomen met een omgekeerde fase microscoop en het ontkiemen gebied wordt gekwantificeerd met behulp van een semi-geautomatiseerde macro plug-in om de ImageJ software ontwikkeld in dit lab. Deze reproduceerbare ex vivo choroidale spruittest kan worden gebruikt om verbindingen voor potentiële behandeling te beoordelen en voor microvasculair ziekteonderzoek om trajecten te beoordelen die betrokken zijn bij de proliferatie van choroidale microvaten met behulp van wild type en genetisch gemodificeerd muisweefsel.

We ontleed de choroid met sclera, ingebed in BME en gekweekt ze voor 6 dagen (Figuur 1).
