従来の哺乳類系における生きた白血球における受容体ダイナミクスの追跡は、いくつかの課題に直面している。ライブ研究では、蛍光タンパク質との受容体融合を細胞内で発現させなければならない。これは初代白血球、特に好中球において困難であり、これまでの研究では、ケモカイン受容体を発現するために代理好中球細胞株を使用してきた3,4。白血球が有益なトラフィッキング欠陥5,6を有する蛍光受容体または変異受容体を発現するトランスジェニックマウスモデルの作製には、かなりの時間と資源の投資が伴う。これらの例においても、生きた動物における受容体動態を画像化するための画像化分解能およびコントラストは制限される可能性があり、研究は固定組織切片5で免疫組織化学を用いている。これらの技術的課題を考えると、化学誘引性受容体のダイナミクスが生きた組織環境における細胞挙動にどのように影響するかについての我々の理解は、現在限られている。
ここでは、ゼブラフィッシュ好中球における受容体トラフィッキングを監視するための方法論を提供する。ゼブラフィッシュはマウスと同様に遺伝的に扱いやすいが、効率的なトランスポゾン系と直接受精卵操作の使用により、トランスジェネシスは比較的簡単である7。透明な幼虫は理想的にはイメージングに適している。ケモカイン受容体ダイナミクスは、蛍光レポーター8、9、10との対応する融合体の発現によって始原生殖細胞および側線原基において視覚化されている。ゼブラフィッシュの幼虫は、哺乳類の好中球に関して高度に保存された遺伝的および細胞的特性を有する成熟した好中球を備えている。細胞骨格ダイナミクスおよび極性調節因子などの細胞内シグナル伝達ダイナミクスは、対応するトランスジェニック株11、12、13の生成によってこれらの細胞において視覚化されている。最近、我々は、組織損傷に対する炎症反応の過程で好中球におけるケモカイン受容体動態を可視化し、機能的に解析した14。ここでは、好中球におけるケモカインシグナル伝達のためのトランスジェニックレポーターラインの生成、ライブイメージングのための胚の調製、好中球シグナル伝達を研究するための創傷アッセイ、および画像の取得および分析のためのプロトコルについて説明する。また、候補リガンドに対するケモカイン受容体応答を試験するためのサイドプロトコルも提供しており、これは特徴付けられていない受容体におけるリガンド認識パターンを確立しようとする際に有用である。これらの技術は、内因性ケモカイン発現の阻害またはリガンド誘導性トラフィッキングの変化を伴う変異型受容体の生成などのさらなる遺伝子操作と組み合わせて使用して、特定のシグナル伝達ダイナミクスがインビボでの白血球挙動にどのように影響するかを調査することができる。蛍光標識されたケモカイン受容体を発現するトランスジェニック株は、直接抗体染色では検出が困難な内因性ケモカイン勾配のレポーターとしても使用することができる。記載された方法論は、レポーターの生成を他の免疫シグナル伝達受容体に拡大するための範囲を提供する。

live imaging of chemokine receptors in zebrafish neutrophils during wound responses

化学的勾配による白血球誘導は、免疫応答に不可欠である。好中球は、重要な抗菌機能を実行する組織損傷部位にリクルートされる最初の細胞です。これらの遺伝子座への彼らのトラフィッキングは、ケモカインを含むいくつかの炎症性化学誘引物質によって画策されている。分子レベルでは、化学誘引性シグナル伝達は、対応する受容体の細胞内輸送によって調節される。しかし、ケモカイン受容体の細胞内変化が細胞および組織レベルで白血球遊走ダイナミクスにどのように影響するかは不明のままである。ここでは、組織損傷に対する炎症反応中の好中球におけるケモカイン受容体ダイナミクスのライブイメージングおよび定量分析のための方法論を説明する。これらのツールは、ゼブラフィッシュ好中球における示差的ケモカイン受容体トラフィッキングが、組織損傷部位における好中球クラスタリングおよび分散を調整することを明らかにした。これは、炎症反応がどのように自己解決されるかについての我々の理解に意味を持つ。記載されたツールは、様々な生理学的および病理学的設定における好中球移動パターンを理解するために使用でき、方法論は他のシグナル伝達受容体に拡張することができる。

腹側フィン創傷に続いて、CHTから腹側フィンへの迅速な好中球動員と、30〜60分以内に創傷縁でのクラスタリングが続く(図1)。我々は、ゼブラフィッシュ好中球24によって発現され、Cxcl8aおよびCxcl8b14を認識する2つのケモカイン受容体Cxcr1およびCxcr2の分布を、スピニングディスク共焦点顕微鏡を用いて可視化した。我々は、好中球が受容体の蛍光タイマー(FT)構築物、すなわちsfGFPとtagRFPのタンデムとの融合を発現するTg(lyz:Cxcr1-FT)およびTg(lyz:Cxcr2-FT)の2つの対応するトランスジェニックラインを生成した(図2および参考文献14)。2つの蛍光色素の使用は、新たに合成された受容体が緑色に蛍光を発し、8,14歳になると徐々に赤色になるため、広範囲の受容体運命のモニタリングを可能にし、原形質膜でのタンパク質代謝回転時間の推定値を提供することを目的としていた。しかし、これらの受容体は好中球原形質膜で構成代謝回転が速く、滞留時間がtagRFPの成熟時間よりも短く、sfGFPは膜局在化を示し、tagRFPは定常状態で小胞局在を示すことが見出された(補足ビデオ1および参考文献14)。そこで、組織損傷部位におけるリガンド誘導インターナリゼーションをモニターするために、sfGFPの分布に着目した。受容体分布のパターンは、隣接するピクセル間の強度の違いを報告するコントラスト測定法を用いて定量化した。その根拠は、受容体分布が膜状で滑らかである場合、コントラスト値が低いことである。受容体分布が小胞状でより穿刺性である場合、コントラスト値は高い(図3)。
別の方法は、対照膜マーカー、例えば膜CFP(mCFP)のレベルに対する受容体レベル(sfGFP強度)の比を定量化することである(図3)。どちらの方法も、細胞内の全球的な小胞受容体分布パターン(コントラスト値が高い)または膜での受容体レベルが低い(sfGFP/mCFP比が低い)ことによって示されるように、受容体の内在化を検出することができた。しかし、コントラスト測定は、膜セグメンテーションの精度が低く、適用できない創傷の好中球クラスターにおける受容体インターナリゼーションを検出することもできます(図3)。この測定基準を使用して、創傷における好中球におけるCxcr1とCxcr2のトラフィックの目に見える違いを定量化することができました(図4および補足ビデオ2)。Cxcr1-FTは創傷に位置する細胞にインターナライズされたが、Cxcr2-FTは創傷の好中球において膜性のままであった(図4A-C、補足ビデオ2および補足ビデオ3)。Cxcl8aおよびCxcl8bの抑制は、モルホリノ処置を介して、創傷におけるCxcr1−FTインターナリゼーションに差次的に影響する(図4C、D)。Cxcr1-FTがCxcl8aに応答することをさらに検証するために、我々は初期胚においてケモカイン応答アッセイを実施した。我々は、Cxcr1-FTが、Cxcl8aが共発現した胚において顕著に内在化することを見出した(図5)。全体として、これらの結果は、記載された方法が好中球におけるケモカイン誘発受容体インターナリゼーションを測定し、これらの効果を媒介するリガンドの同一性を確立するために展開され得ることを示している。
