복부 지느러미 상처는 CHT에서 복부 지느러미로 급속 호중구 동원 및 30-60 분 이내에 상처 마진에서 군집화됩니다 (그림 1). 우리는 제브라피쉬 호중구 24에 의해 발현되는 두 개의 케모카인 수용체인 Cxcr1 및 Cxcr2의 분포를 시각화하고 스핀 디스크 공초점 현미경을 사용하여 Cxcl8a 및 Cxcl8b14를 인식합니다. 우리는 두 개의 상응하는 트랜스제닉 라인, Tg(lyz:Cxcr1-FT) 및 Tg(lyz:Cxcr2-FT)를 생성했는데, 여기서 호중구는 수용체의 형광 타이머(FT) 구축물, 즉 sfGFP 및 tagRFP의 탠덤과의 융합을 발현한다(도 2 및 참조14). 두 형광단의 사용은 광범위한 수용체 운명을 모니터링하고 원형질막에서의 단백질 턴오버 시간의 추정치를 제공하기위한 것이었으며, 새로 합성 된 수용체는 녹색으로 형광을 띠고 8,14 세가되면 점진적으로 적색이됩니다. 그러나, 이들 수용체는 호중구 원형질막에서 빠른 구성적 턴오버를 가지며 체류 시간이 tagRFP의 성숙 시간보다 짧았고, sfGFP는 막 국소화를 나타내고 tagRFP는 정상 상태에서 수포 국소화를 나타내는 것으로 밝혀졌다 (보충 비디오 1 및 참조14). 따라서, 우리는 조직 손상 부위에서 리간드-유도된 내재화를 모니터링하기 위해 sfGFP의 분포에 초점을 맞추었다. 수용체 분포의 패턴은 이웃 픽셀들 사이의 강도의 차이를 보고하는 콘트라스트 메트릭을 사용하여 정량화되었다. 그 근거는 수용체 분포가 막강하고 매끄럽게 될 때 대조 값이 낮다는 것입니다. 수용체 분포가 수포성이고 더 많은 구멍을 뚫으면 명암비가 높습니다 (그림 3).
대안적인 방법은 대조군 막 마커, 예를 들어 막 CFP(mCFP)의 수준에 대한 수용체 수준(sfGFP 강도)의 비율을 정량화하는 것이다(도 3). 두 방법 모두 세포에서 전세계적으로 더 많은 수포 수용체 분포 패턴 (더 높은 명암치) 또는 막에서의 낮은 수용체 수준 (더 낮은 sfGFP / mCFP 비율)에 의해 나타나는 것처럼 수용체 내재화를 감지 할 수 있습니다. 그러나, 콘트라스트 메트릭은 또한 상처에서 호중구 클러스터에서 수용체 내재화를 검출할 수 있었는데, 여기서 막 분할은 덜 정확했고 적용 가능하지 않았다(그림 3). 이 메트릭을 사용하여 상처에서 호중구에서 Cxcr1과 Cxcr2 밀매의 가시적 인 차이를 정량화 할 수있었습니다 (그림 4 및 보충 비디오 2).
