an efficient method for adenovirus production

为了确定每mL（TU/mL）的转导单元中表达的病毒滴定器，AD293细胞感染了AdV-GFP的连续稀释。48小时后，受感染的细胞表示GFP，与病毒悬浮的稀释因子成反比。荧光显微镜观察到这一点，GFP阳性细胞的百分比由流动细胞学（图5）确定。为了计算滴定器，考虑了诱导5 -20%GFP阳性细胞的病毒稀释（图5C）。通常，我们获得GFP-腺病毒的病毒滴定剂为±10 10（TU/mL）。
下面，我们提供了一个特定腺苷基滴定器计算的例子，其中 300000 个细胞 （C） 被转导为 1 mL 腺苷溶液 （V），稀释因子为 106 （D），为此获得了 6% 的 GFP 阳性细胞 （F）：

为了测试预制腺病毒的转导潜力，使用了四个细胞系：人类内皮细胞（EA.hy926）、牛主动脉内皮细胞（BAEC）、穆林肝细胞（Hepa 1-6）和穆林间皮质频闪细胞（MSC）。内皮细胞（EA.hy926和BAEC）用25个TU/细胞进行转导，用5个TU/细胞转导肝细胞，用250个TU/细胞转导MSC。
