Dit document biedt een effectief experimenteel schema om de ontwikkeling van kwartelembryo's te evalueren door de basisontwikkelingsindexen te detecteren. Er zijn echter nog enkele beperkingen aan dit experiment. 
Ten eerste is de mortaliteit van kwarteleibryo's in het latere stadium van het uitkomen hoger vanwege het schelploze uitkomen. Er zijn kunstmatig oncontroleerbare factoren zoals de vernietiging van de normale eiwitverhouding in het experimentele proces. We hebben de blootstellingstij...

De productie van plastic afval was ongeveer 6300 Mt in 2015, waarvan een tiende werd gerecycled, en de rest werd ondergronds verbrand of begraven. Naar schatting zou tegen 2050 ongeveer 12.000 mt plastic afval ondergronds worden begraven1. Met de aandacht van de internationale gemeenschap voor plastic afval stelde Thompson in 2004 voor het eerst het concept van microplasticsvoor 2. Microplastics (Parlementsleden) verwijzen naar kleine deeltjesplastics met een deeltjesdiameter van minder dan 5 mm. Op dit moment hebben onderzoekers de alomtegenwoordige aanwezigheid van parlementsleden ontdekt in de kustlijn van verschillende continenten, de Atlantische eilanden, binnenmeren, het Noordpoolgebied en diepzeehabitats3,4,5,6,7. Daarom zijn meer onderzoekers begonnen met het bestuderen van de milieurisico's van parlementsleden.
Organismen kunnen parlementsleden in het milieu opnemen. Parlementsleden werden gevonden in het spijsverteringskanaal van 233 mariene organismen wereldwijd (waaronder 100% schildpadsoorten, 36% zeehondensoorten, 59% walvissoorten, 59% zeevogelsoorten, 92 soorten zeevissen en 6 soorten ongewervelden)8. Bovendien kunnen parlementsleden het spijsverteringsstelsel van de organismen blokkeren, zich ophopen en migreren in hun bobies9. Het is gebleken dat parlementsleden via de voedselketen kunnen worden overgedragen en hun inname verschilt met de veranderingen van habitat, groeifase, voedingsgewoonten en voedselbronnen10. Sommige onderzoekers meldden het bestaan van parlementsleden in de uitwerpselen van zeevogels11, wat betekent dat zeevogels fungeren als drager van parlementsleden. Bovendien kan de inname van parlementsleden de gezondheid van sommige organismen beïnvloeden. Parlementsleden kunnen bijvoorbeeld verstrikt raken in het maagdarmkanaal, waardoor de mortaliteit van walvisachtigentoeneemt 12.
Parlementsleden alleen hebben toxische effecten op organismen en gezamenlijke toxische effecten op organismen met andere verontreinigende stoffen. Inname van milieugerelateerde concentraties van plastic afval kan de endocriene systeemfunctie van volwassen vissen verstoren13. De grootte van microplastics is een van de belangrijke factoren die van invloed zijn op hun opname en accumulatie door organismen14,15. De kleine kunststoffen, met name de nanosize kunststoffen, zijn gevoelig voor interactie met cellen en organismen met hoge toxiciteit16,17,18,19. Hoewel de schadelijke effecten van microplastics met nanodeeltjesgrootte op organismen het huidige onderzoeksniveau overschrijden, is de detectie en kwantificering van microplastics met een grootte van minder dan enkele micrometers, met name de submicron/nanoplastics in het milieu, nog steeds een grote uitdaging. Daarnaast hebben nanoplastics ook enkele effecten op embryo's. Polystyreen kan de ontwikkeling van zee-egelsembryo's beschadigen door eiwit- en genprofielen te reguleren20.
Om de mogelijke impact van parlementsleden op organismen te onderzoeken, hebben we deze studie uitgevoerd. Vanwege de gelijkenis tussen vogelembryo's en menselijke embryo's worden ze meestal gebruikt in ontwikkelingsbiologieonderzoek21, waaronder angiogenese en antiangiogenese, weefseltechnologie, biomateriaalimplantaat en hersentumoren22,23,24. De embryo's van de vogel hebben de voordelen van lage kosten, een korte cultuurcyclus en gemakkelijke verrichting25,26. Daarom kozen we kwarteleibryo's met een korte groeicyclus als proefdier in deze studie. Tegelijkertijd kunnen we de morfologische veranderingen van kwartelpembryo's die tijdens de embryonale ontwikkelingsfase aan parlementsleden zijn blootgesteld, direct waarnemen met behulp van een eierschaalvrije broedtechnologie. De gebruikte experimentele materialen waren polypropyleen (PP) en polystyreen (PS). Omdat PP en PS27 het grootste deel van de polymeertypen vertegenwoordigen die wereldwijd in sedimenten en waterlichamen worden verkregen, zijn de meest voorkomende polymeertypen die worden gewonnen uit gevangen mariene organismen ethyleen en propyleen28. Dit experimentele protocol beschrijft het hele proces voor visuele evaluatie van toxicologische effecten van parlementsleden op kwartelembryo's die aan parlementsleden zijn blootgesteld. We kunnen deze methode gemakkelijk uitbreiden om de toxiciteit van andere verontreinigende stoffen voor de embryoontwikkeling van andereovipareuze dieren te onderzoeken.

Microplastics zijn een opkomende wereldwijde verontreinigende stof die een grote bedreiging vormt voor de gezondheid van dieren als gevolg van hun opname en translocatie in dierlijke weefsels en organen. Ecotoxicologische effecten van microplastics op de ontwikkeling van vogelembryo's zijn niet bekend. Het vogelei is een compleet ontwikkelings- en voedingssysteem en de hele embryo-ontwikkeling vindt plaats in de eierschaal. Daarom wordt een direct verslag van de ontwikkeling van vogelembryo's onder de stress van verontreinigende stoffen zoals microplastics sterk beperkt door de ondoorzichtige eierschaal in traditionele arcering. In deze studie werden de effecten van microplastics op de ontwikkeling van kwartelembryo's visueel gemonitord door uit te broeden zonder eierschaal. De belangrijkste stappen omvatten het reinigen en desinfecteren van bevruchte eieren, de incubatie vóór blootstelling, de incubatie op korte termijn na blootstelling en de monsterextractie. De resultaten tonen aan dat in vergelijking met de controlegroep het natte gewicht en de lichaamslengte van de aan microplastics blootgestelde groep een statistisch verschil vertoonden en het leveraandeel van de hele blootgestelde groep aanzienlijk steeg. Daarnaast evalueerden we externe factoren die de incubatie beïnvloeden: temperatuur, vochtigheid, eirotatiehoek en andere omstandigheden. Voor de analyse van experimentele gegevens vergeleken we nat gewicht, lichaamslengte, borstbeenlengte en de verandering van hepatosomatische index tussen de controlegroep en de 6 experimentele groepen, waarbij we de groei en ontwikkeling van de kwartelembryo's vanuit een macroperspectief meten en weerspiegelen.
