Die Produktion von Kunststoffabfällen betrug im Jahr 2015 etwa 6300 Mt, von denen ein Zehntel recycelt wurde und der Rest verbrannt oder unter der Erde vergraben wurde. Es wird geschätzt, dass bis 2050 etwa 12.000 Tonnen Plastikmüll unterirdisch vergraben würden1. Mit der Aufmerksamkeit der internationalen Gemeinschaft für Kunststoffabfälle schlug Thompson erstmals 2004 das Konzept des Mikroplastiks vor2. Mikroplastik (MPs) bezieht sich auf kleine Partikelkunststoffe mit einem Partikeldurchmesser von weniger als 5 mm. Derzeit haben Forscher die allgegenwärtige Präsenz von Abgeordneten an der Küste verschiedener Kontinente, der Atlantikinseln, der Binnenseen, der Arktis und der Tiefseelebensräume3,4 ,5,6,7festgestellt . Daher haben mehr Forscher begonnen, die Umweltgefahren von Abgeordneten zu untersuchen.
Organismen könnten Abgeordnete in der Umwelt aufnehmen. MPs wurden im Verdauungstrakt von 233 Meeresorganismen weltweit gefunden (darunter 100% Schildkrötenarten, 36% Robbenarten, 59% Walarten, 59% Seevogelarten, 92 Arten von Seefischen und 6 Arten von Wirbellosen)8. Darüber hinaus können Abgeordnete das Verdauungssystem der Organismen blockieren, sich ansammeln und in ihren Bobies migrieren9. Es wurde festgestellt, dass Abgeordnete über die Nahrungskette übertragen werden können und ihre Aufnahme sich mit den Veränderungen des Lebensraums, des Wachstumsstadiums, der Ernährungsgewohnheiten und der Nahrungsquellen unterscheidet10. Einige Forscher berichteten über die Existenz von Abgeordneten im Kot von Seevögeln11, was bedeutet, dass Seevögel als Träger von Abgeordneten fungieren. Darüber hinaus kann die Einnahme von Abgeordneten die Gesundheit einiger Organismen beeinträchtigen. Zum Beispiel können Abgeordnete im Magen-Darm-Trakt verwickelt sein, wodurch die Mortalität von Walen erhöht wird12.
Abgeordnete allein haben toxische Wirkungen auf Organismen sowie gemeinsame toxische Wirkungen auf Organismen mit anderen Schadstoffen. Die Einnahme von umweltbedingten Konzentrationen von Kunststoffabfällen kann die Funktion des endokrinen Systems erwachsener Fische stören13. Die Größe von Mikroplastik ist einer der wichtigen Faktoren, die ihre Aufnahme und Akkumulation durch Organismen beeinflussen14,15. Die kleinen Kunststoffe, insbesondere die nanogroßen Kunststoffe, sind anfällig für Wechselwirkungen mit Zellen und Organismen mit hoherToxizität 16,17,18,19. Obwohl die schädlichen Auswirkungen von Mikroplastik in Nanopartikelgröße auf Organismen das aktuelle Forschungsniveau übersteigen, ist der Nachweis und die Quantifizierung von Mikroplastik mit Größen unter mehreren Mikrometern, insbesondere der Submikron-/Nanokunststoffe in der Umwelt, nach wie vor eine große Herausforderung. Darüber hinaus haben Nanokunststoffe auch einige Auswirkungen auf Embryonen. Polystyrol kann die Entwicklung von Seeigelembryonen schädigen, indem es Protein- und Genprofile reguliert20.
Um die möglichen Auswirkungen von Abgeordneten auf Organismen zu untersuchen, haben wir diese Studie durchgeführt. Aufgrund der Ähnlichkeit zwischen Vogelembryonen und menschlichen Embryonen werden sie normalerweise in der entwicklungsbiologischen Forschung21 verwendet, einschließlich Angiogenese und Antiangiogenese, Tissue Engineering, Biomaterialimplantat und Hirntumoren22,23,24. Vogelembryonen haben die Vorteile von niedrigen Kosten, einem kurzen Kulturzyklus und einfacher Bedienung25,26. Daher haben wir wachtelembryonen mit einem kurzen Wachstumszyklus als Versuchstier in dieser Studie ausgewählt. Gleichzeitig können wir die morphologischen Veränderungen von Wachtelembryonen, die während der Embryonalentwicklungsphase den MPs ausgesetzt sind, mit einer eierschalenfreien Bruttechnologie direkt beobachten. Die verwendeten Versuchsmaterialien waren Polypropylen (PP) und Polystyrol (PS). Da PP und PS27 den größten Anteil an Polymertypen haben, die weltweit in Sedimenten und Gewässern gewonnen werden, sind ethylen und propylen28die häufigsten Polymertypen, die aus gefangenen Meeresorganismen gewonnen werden. Dieses experimentelle Protokoll beschreibt den gesamten Prozess zur visuellen Bewertung der toxikologischen Wirkungen von Abgeordneten auf Wachtelembryonen, die Den Abgeordneten ausgesetzt sind. Wir können diese Methode leicht erweitern, um die Toxizität anderer Schadstoffe für die Embryonalentwicklung anderer oviparer Tiere zu untersuchen.

Mikroplastik ist ein aufstrebender globaler Schadstofftyp, der aufgrund seiner Aufnahme und Translokation in tierischen Geweben und Organen eine große Gesundheitsbedrohung für Tiere darstellt. Ökotoxikologische Wirkungen von Mikroplastik auf die Entwicklung von Vogelembryonen sind nicht bekannt. Das Vogelei ist ein komplettes Entwicklungs- und Ernährungssystem, und die gesamte Embryoentwicklung findet in der Eierschale statt. Daher ist eine direkte Aufzeichnung der Entwicklung von Vogelembryonen unter dem Stress von Schadstoffen wie Mikroplastik durch die undurchsichtige Eierschale beim traditionellen Schlüpfen stark eingeschränkt. In dieser Studie wurden die Auswirkungen von Mikroplastik auf die Entwicklung von Wachtelembryonen visuell überwacht, indem sie ohne Eierschale schlüpften. Die Ergebnisse zeigen, dass im Vergleich zur Kontrollgruppe das Nassgewicht und die Körperlänge der mikroplastikexponierten Gruppe einen statistischen Unterschied auften und der Leberanteil der gesamten exponierten Gruppe signifikant anstieg. Darüber hinaus haben wir externe Faktoren bewertet, die die Inkubation beeinflussen: Temperatur, Feuchtigkeit, Drehwinkel der Eier und andere Bedingungen. Für die Analyse experimenteller Daten verglichen wir Nassgewicht, Körperlänge, Brustbeinlänge und die Veränderung des hepatosomatischen Index zwischen der Kontrollgruppe und den 6 experimentellen Gruppen, wobei wir das Wachstum und die Entwicklung der Wachtelembryonen aus einer Makroperspektive maßen und reflektieren.
