De experimentele protocollen beschreven in dit document bieden nuttige instrumenten om de oncogene transformatie van in vitro gekweekte cellen te beoordelen. Elke techniek evalueert specifieke aspecten van het transformatieproces, en daarom moet bijzondere aandacht worden besteed aan het trekken van conclusies uit een enkele analyse. Groeicurven opbouw is een aanpak die informatie die al beschikbaar is bij het kweken van cellen voor andere doeleinden vereist. Dat maakt deze techniek goedkoper en gemakkelijker toe te pass...

Meerdere opeenvolgende gebeurtenissen zijn vereist voor de ontwikkeling van het neoplasma. In 2011 beschreven Hanahan en Weinberg 10 vermogens die de groei, overleving en verspreiding van getransformeerde cellen mogelijk maken: de zogenaamde "Hallmarks of Cancer"1. De hier beschreven methodologie stelt drie verschillende instrumenten samen om in vitro cellulaire transformatie te evalueren door zich te concentreren op enkele onderscheidende kenmerken van de tumorcellen. Deze technieken beoordelen de celproliferatiesnelheid, het gedrag van cellen wanneer gekweekt in 3D en hun vermogen om kolonies met ankerveronafhankelijkheid te vormen.
Celmodellen zijn cruciaal om hypothese in vitro te testen. Er zijn verschillende benaderingen ontwikkeld om experimentele modellen van cellulaire transformatie te genereren voor de studie van kanker2,3,4. Aangezien borstkanker is de meest voorkomende kanker onder vrouwen wereldwijd en is verantwoordelijk voor ongeveer 15% van de sterfgevallen door kanker onder vrouwen5, het verstrekken van geschikte cellulaire modellen van borstepitheelcellen is van het grootste belang voor verder onderzoek. In dit artikel hebben we het potentieel van drie technieken geïllustreerd om cellulaire transformatie te evalueren met behulp van een experimenteel model van borstwerkelijke epitheelcellen (BPC's) transformatie aanvankelijk beschreven door Ince en collega's in 20076 en later geïmplementeerd in ons laboratorium7. Dit experimentele model is gebaseerd op de sequentiële wijziging van drie gerichte genen (SV40 Large T en kleine t-antigenen hierin aangeduid als Ttag, hTERTen HRAS) op het genoom van niet-getransformeerde BBC's. Bovendien, de methode die wordt gebruikt voor BPECs afleiding bevordert het behoud van borstepileliale cellen met luminale of myoepithelial markers, wat resulteert in een heterogene celcultuur die een aantal van de borstklier fysiologische eigenschappen behoudt.
In de borstklier bevinden zich borstlichaamcellen, die verantwoordelijk zijn voor de melkproductie, in de buurt van het lumen, terwijl myoepitheliale cellen rond luminale cellen worden verwijderd en zorgen voor samentrekkingsbewegingen die de melk naar de tepel leiden. Het verlies van de juiste organisatie tussen deze cellijnen is een kenmerk van tumorale transformatie8 die in vitro kan worden beoordeeld na immunofluorescente detectie van onderscheidende afstammingmarkers in 3D-celculturen. Een ander belangrijk kenmerk van tumorcellen is hun vermogen om te groeien zonder gehechtheid aan andere cellen en aan de extracellulaire matrix1. Wanneer gezonde cellen worden gedwongen om te groeien in suspensie, mechanismen zoals anoikis \u2012 een soort celdood geïnduceerd in reactie op detachement van de extracellulaire matrix \u2012 worden geactiveerd9. De ontduiking van celdood is een van de kenmerkende kenmerken van kanker en dus zijn getransformeerde cellen in staat om anoikis te activeren en op een ankeronafhankelijke manier te overleven. Deze capaciteit kan in vitro worden geëvalueerd met de verankering-onafhankelijke test met behulp van zachte agar. Bovendien is een inherent kenmerk van tumorweefsels hun aanhoudende proliferative signaleringscapaciteit, die gemakkelijk kan worden gecontroleerd onder in vitro omstandigheden door de toename van het aantal cellen langs de tijd te meten, niet alleen in suspensietesten, maar ook door het toezicht op de groeisnelheid van monolaagaanhangende culturen.
Ondanks het beste model om tumorigenic potentieel te testen is de inenting van tumorcellen in murinemodellen en evaluatie van tumorontwikkeling in situ, is het belangrijk om het aantal dieren dat in experimentele procedures wordt aangewend zo veel mogelijk te minimaliseren. Daarom is het hebben van geschikte tests om transformatie in vitro te beoordelen een topprioriteit. Hier bieden we een reeks tools om het tumorigenic potentieel van gedeeltelijk en volledig getransformeerde borsteptropelial cellen te evalueren die gemakkelijk kunnen worden geïmplementeerd in de meeste laboratoria die werken met cellulaire transformatiemodellen.

Tumorigenese is een proces in meerdere stappen waarin cellen mogelijkheden verwerven die hun groei, overleving en verspreiding onder vijandige omstandigheden mogelijk maken. Verschillende tests zijn gericht op het identificeren en kwantificeren van deze kenmerken van kankercellen; echter, ze richten zich vaak op een enkel aspect van cellulaire transformatie en, in feite, meerdere tests nodig zijn voor hun juiste karakterisering. Het doel van dit werk is om onderzoekers te voorzien van een reeks instrumenten om cellulaire transformatie in vitro vanuit een breed perspectief te beoordelen, waardoor het mogelijk is om goede conclusies te trekken.
Een aanhoudende proliferative signaling activatie is het belangrijkste kenmerk van tumorweefsels en kan gemakkelijk worden gecontroleerd onder in vitro omstandigheden door het berekenen van het aantal populatieverdubbelingen bereikt in de tijd. Bovendien maakt de groei van cellen in 3D-culturen hun interactie met omringende cellen mogelijk, die lijkt op wat er in vivo gebeurt. Dit maakt de evaluatie van cellulaire aggregatie mogelijk en, samen met immunofluorescente etikettering van onderscheidende cellulaire markers, om informatie te verkrijgen over een ander relevant kenmerk van tumorale transformatie: het verlies van een goede organisatie. Een ander opmerkelijk kenmerk van getransformeerde cellen is hun vermogen om te groeien zonder gehechtheid aan andere cellen en aan de extracellulaire matrix, die kan worden geëvalueerd met de verankeringstest.
Gedetailleerde experimentele procedures om de groei van cellen te evalueren, om immunofluorescente etikettering van cellijnmarkers in 3D-culturen uit te voeren en om verankering-onafhankelijke celgroei in zachte agar te testen, worden verstrekt. Deze methoden zijn geoptimaliseerd voor borst primary epitheelcellen (BPEC) vanwege de relevantie ervan bij borstkanker; Na enkele aanpassingen kunnen echter procedures worden toegepast op andere celtypen.

Er werd gekozen voor een experimenteel model van cellulaire transformatie met de introductie van drie genetische elementen in BPC's om representatieve resultaten van oncogene transformatie te genereren6,7 (figuur 3).
