脊髄不安定性(SI)は、平衡状態にあっては不調の不安定な脊柱を引き起こします。これは、靭帯や筋肉などの周囲の支持組織の弱さによる腰部運動セグメントの異常な動きによって引き起こされる可能性があります。脊髄融合後の手術19も一般的に見られる。SIはIDDの主な原因と考えられています。そこで、ヒトIDDプロセス20,21を模したSIマウスモデル(腰椎に焦点を当てた)の開発を目指す。
プロトコルでは、腰椎3番目(L3)を腰椎第5(L5)の切除によって腰椎不安定性(LSI)マウスモデルを確立する手順を、上腕および内棘靭帯と共に第5(L5)の脊椎プロセスを腰椎に設定する手順を導入した(図1A,B)。この動物モデルは、エンドプレート(EPs)の肥大および多孔性によって示されるように、手術後早くも1週間のIDDを発症する。IVD容積は、IDDの程度を示す増加したIVDスコアと共に、16週間を通じて手術後2週間減少し始める。詳細で視覚化された手順は、研究者が研究室でLSIマウスモデルを確立し、必要に応じてIDD研究に適用するのに役立つと考えています。
