an orthotopic resectional mouse model of pancreatic cancer

קיים מחסור במודלים משביעי רצון של בעלי חיים לחקר טיפול אדג'ובנטי ו/או ניאו-ג'ובנטי בחולים הנחשבים לניתוח של סרטן הלבלב (PC). כדי לטפל במחסור זה, אנו מתארים מודל העכבר מעורבים השתלה אורתוטופית של המחשב ואחריו כריתת לבלב דיסטלי ו splenectomy. המודל הוכח כבטוח וגמיש כראוי לחקר גישות טיפוליות שונות בהגדרות אדג'ובנטיות וניאו-אדג'ובנטיות.
במודל זה, גידול בלבלב נוצר לראשונה על ידי השתלת תערובת של תאים סרטניים בלבלב האנושי (לוציפראז מתויג AsPC-1) וסרטן האדם הקשורים תאים כוכבי הלבלב לתוך הלבלב הדיסטלי של עכברי עירום Balb/c אתימי. לאחר שלושה שבועות, הסרטן הוא resected על ידי לפרוטומיה מחדש, כריתת לבלב דיסטלי ו splenectomy. במודל זה, הדמיה bioluminescence ניתן להשתמש כדי לעקוב אחר ההתקדמות של התפתחות סרטן והשפעות של כריתה / טיפולים. לאחר כריתה, ניתן לתת טיפול אדג'ובנטי. לחלופין, ניתן לתת טיפול ניאואדג'ובנטי לפני כריתה.
מוצגים נתונים מייצגים מ-45 עכברים. כל העכברים עברו כריתת לבלב דיסטלית מוצלחת / כריתת טבור ללא בעיות של עצירת דימום. שולי לבלב פרוקסימליים מקרוסקופיים הגדולים מ- 5 מ"מ הושגו ב- 43 (96%) עכברים. שיעור ההצלחה הטכנית של כריתת הלבלב היה 100%, עם 0% תמותה מוקדמת ותחלואה. אף אחת מהחיות לא מתה במהלך השבוע שלאחר כריתה.
לסיכום, אנו מתארים טכניקה חזקה הניתנת לשחזור עבור מודל כריתה כירורגית של סרטן הלבלב בעכברים המחקה את התרחיש הקליני. המודל עשוי להיות שימושי עבור בדיקות של טיפולים אדג'ובנטיים ו neoadjuvant.

חמישים ותשעה עכברים רצופים עברו ניתוח השתלה. דליפת ברוטו התרחשה בשמונה (14%) עכברים. מידת הדליפה בזמן ההזרקה מוערכת כמתואר לעיל בסעיף הפרוטוקול. לאחר שלושה שבועות כדי לאפשר לגידולים מושתלים אלה לגדול, הדמיה ביולומינציה לפני כריתה בוצעה כדי לא לכלול עכברים עם מחלה גרורתית ברוטו לפני כריתה. א...
