喘息は気管支収縮および気道過敏性1を特徴とする慢性疾患である。世界中で約3億3,940万人が喘息2と診断されている。喘息の罹患率は世界的に増加しており、今後10年間で1億人の新しい症例が予想される3 .吸入コルチコステロイドおよび長時間作用型β 2-アゴニストは、全患者の症状を効果的に制御しない4.したがって、喘息を治療するための鍼治療などの代替療法の使用は、注目度が高まっている5。喘息を治療するための有効な方法は、針GV14(ダズイ)、両側BL12(風門)および両側BL13(フェイシュ)6.鍼治療は、アレルギー性喘息患者の唾液および鼻分泌物中の分泌免疫グロブリンA(SIgA)のレベルを有意に低下させ、総有効率85%6で末梢血好酸球数を減少させる。慢性喘息を有するマウスにおけるBL13およびST36(Zusanli)における鍼治療は、IL-17のレベルを40%減少させ、オボアルブミン(OVA)群のそれと比較して平滑筋厚を33%減少させ、したがって症状7,8を緩和する。しかし、喘息に対する鍼治療のメカニズムは完全には明らかではない。
動物モデルは喘息治療10の有効性を評価する呼吸機能の連続測定に使用することができるので、9の喘息研究のための重要なツールである。一方、喘息の病態生理学の研究は、主要因子11のレベルの変化を検証するために気管、肺および気管支肺胞洗浄からのサンプルを含む試験サンプルを必要とする。喘息のラットモデルは、長期にわたる気道応答を産生し、即時および後期気道応答12を示すため、喘息病態生理を評価するために一般的に使用される。しかし、繰り返される実験的刺激のために、ラットはしばしば過敏になる。そのため、ラットを固定するための適切な方法が必要である。ラット固定のために一般的に使用される方法には、麻酔および結合13が含まれる。麻酔法は、視力の露出を改善するが、神経伝導に影響を与え、最終的には実験結果14に影響を与える可能性がある。この結合方法は麻酔と同じ生理学的効果を誘発しないが、抑制されたラットは骨格筋の緊張を経験する。また、針は指定位置15に容易に達することができない。ここでは、ラットを制限せずに固定し、背中の指を完全に露出させるために使用できる高いプラットフォームを紹介します。ラットは、鍼治療の投与中に高いプラットフォーム上に配置することができます。高いプラットホームはラットが立っている間だけ収容することができ、そして地面から一定の距離に置かれるので、ラットは高所16の恐怖のために動かないだろう。
