このプロトコルは、さまざまな生体材料に対する宿主の免疫応答を特徴付けるのに役立ち、その後、より良い将来の医療用インプラントの設計に役立ちます。フローサイトメトリーは、生体材料に浸潤した免疫細胞に関する情報を提供し、細胞が損傷や物質の移植にどのように反応するかのメカニズムや、治療を改善するための標的を決定するのに役立ちます。同じプロトコルを変更して、異なる設定での免疫応答を特徴付けることができます。1週間前にECMインプラントを装着し、5ミリリットルの無血清培地を含む50ミリリットルのチューブに採取したマウスの大腿四頭筋を解剖します。最後にハサミでティッシュをさいの目に切った。次に、チューブに5ミリリットルの消化酵素培地を加えます。消化培地とさいの目に切った組織を含むチューブを、摂氏37度、100RPMで45分間振とうインキュベーターに入れます。インキュベーションフィルターの最後に、消化された組織懸濁液を70ミクロンのストレーナーを通して個々の50ミリリットルのチューブに入れます。5ミリリットルのシリンジヘッドを使用して、ティッシュの固い塊を粉砕し、室温のPBSでストレーナーを洗浄します。ストレーナに残っている残留物を廃棄し、室温PBSでチューブの容量を50ミリリットルに調整します。遠心分離により細胞を回収し、PBS中の10ミリリットルの冷たい5ミリモルEDTA溶液にペレットを再懸濁します。サンプル中に血球が存在する場合は、ペレットを1ミリリットルの赤血球溶解緩衝液に再懸濁し、10分間インキュベートしてから、9ミリリットルのEDTAを加えます。懸濁液を氷上に10分間放置します。次に、コールドPBSで容量を50ミリリットルに調整します。遠心分離により細胞を回収し、ペレットと100マイクロリットルの低温PBSを再懸濁します。懸濁液10マイクロリットルを個々のマイクロ遠心チューブに移し、10マイクロリットルのトリップとブルーと混合してカウントします。残りの細胞容量をVボトム96ウェルプレートの各ウェルに分注します。ウェルから20マイクロリットルの細胞を取り出し、それらを新しいウェルに添加して、未染色コントロールとして使用します。次に、PBSを使用して、各ウェルの最終容量を200マイクロリットルにします。遠心分離によりプレートウェルの底部に細胞を回収し、ウェルあたり1〜1000濃度の生存率色素100マイクロリットルに細胞を再懸濁します。インキュベーションの最後に、各ウェルを100マイクロリットルの新鮮なPBSで洗浄し、ウェルあたり200マイクロリットルの染色バッファーにペレットを再懸濁します。2回目の遠心分離後、細胞を1〜100希釈の単球遮断薬50マイクロリットルに再懸濁します。細胞懸濁液を氷上で5分間インキュベートした後、50マイクロリットルの抗体カクテルを加えます。光から保護された氷の上で30分間インキュベートします。次に、ウェルごとに100マイクロリットルの染色バッファーでウェルを洗浄します。細胞を再度遠心分離し、200マイクロリットルの染色バッファーで洗浄します。このプロセスをさらに2回繰り返した後、細胞を400マイクロリットルの染色バッファーに再懸濁します。サンプルを分析する前に、未染色のサンプルを実行して、側方散乱線プロットと前方散乱線プロットで細胞集団を調整できるようにします。次に、染色したサンプルを実行します。自家蛍光シグネチャーを抽出します。次に、FCS ファイルをインポートして、アンミキシングのコントロールとして使用します。アンミキシングアイコンをクリックすると、アンミキシングウィザードが開き、ポジティブとネガティブのポピュレーションをゲートすることで、すべての単色コントロールを使用してアンミキシングを実行します。次に、QCセクションをクリックして、複雑さのインデックスを確認します。複雑度指数は、スペクトル シグネチャが混ざり合っていない場合に、スペクトル シグネチャのコレクションをどの程度区別できるかを示す尺度です。最後に、ライブアンミックスをクリックしてサンプルをアンミックスします。ここでは、対照マウス組織に関する14色の事実の結果を観察することができます。この解析では、ly6g陽性好中球やly6c中発現および高発現単球クラスなど、いくつかのロビイスト集団が観察されました。CD11c高MHCの2つの陽性樹状細胞は、マクロファージや好中球や単球などの他の骨髄系細胞を除外するためにCD11bに対してゲーティングすると容易に明らかになりました。CD206陽性CD86陽性樹状細胞のサブセットは、CD86高M1様樹状細胞とCD206高M2様樹状細胞集団の両方を含むこのCD11c陽性集団に焦点を当てることによって同定できる。F4/80は、さまざまなマクロファージ集団で一般的に観察されるような発現の勾配を示しました。Siglec-Fは、F4-80陽性集団と陰性集団の両方に存在し、それぞれマクロファージサブセットと好酸球に対応する可能性が最も高い。細胞表面マーカーで染色することで、これらの細胞型の指定を行うことができますが、異なるマーカーを発現する細胞は、より二元的な分類とは対照的に、機能的な方法で見るべきであることに注意することが重要です。このプロトコールを試みる際は、パネルを設計する前に、染色されていないサンプルの自家蛍光シグネチャーを理解することが、より良い混合を実現するための鍵となることを念頭に置いてください。単離された細胞は、細胞の解析に加えて、細胞アッセイによるダウンストリーム評価、in vitro、トランスクリプトーム解析、または細胞形態を評価するための顕微鏡解析のために、特定の集団に分類することができます。生体材料のフローサイトメトリー解析の複雑さが増すことで、免疫学の基礎研究と新しい治療法のエンジニアリングの間のギャップを埋めることができます。
