1. ブドウ果汁寒天プレートの作製

1 分間すすぎます 次の溶液を追加する前に、洗浄バッファーがスライドから完全に乾燥していることを確認してください。各スライドに100ミリリットルのブロッキング溶液を加え、スライドを30分間インキュベートする。これは、良好な染色を確実にするために、光のない環境で行うのが最善です。次にスライドを20mLの抗体溶液中で再び30分間インキュベートする。抗体溶液をスライドに直接塗布して、組織を完全にカバーします。インキュベーションが完了したら、100ミリリットルの洗浄バッファーを塗布します。光のない条件下で15分間インキュベートし、未結合の抗体コンジュゲートを除去した。洗浄バッファーを取り外します。次に、20ミリリットルの検出バッファーを塗布し、約4分間インキュベートさせた。4 分後、検出バッファーを削除します。スライドを乾燥させて追加の溶液がないようにし、新しく調製したカラー基質溶液を10ミリリットル加える。この反応は数分以内に始まりますが、16時間後に完了します。だから、光のない状態でその期間インキュベートすることを許可してください。バンドが検出される目的のスポットが見つかったら、スライドを 50 ミリリットルの TE
