The ideal plating densities for both cell types need to be decided with a series of cell number titration to achieve proper differentiation (Figure 3A). After 4 weeks in co-cultures, the iOPCs are expected to be adequately differentiated into OLs that are positive for specific markers such as MBP and extend processes to ensheath axons (Figure 3B). The co-culture system can robustly boost up the number of synapses, indicating that the iOPCs provide a neuronal support through physical contacts or release of trophic factors (Figure 3C). We can maintain the co-cultures in acceptable health condition for up to 6 weeks and observe that the synapse number and other neuronal attributes plateau around the fifth week. Oligodendrocythärstamningsceller - inklusive oligodendrocytprekursorceller (OPC), myeliniserande oligodendrocyter och övergångstyper däremellan - utgör en stor grupp mänskliga hjärnceller1 som aktivt deltar i många kritiska funktioner för korrekt drift och underhåll av vårt centrala nervsystem under hela neural utveckling och åldrande 2,3,4 . Medan oligodendrocyter är välkända för att producera myelin för att underlätta neuronal aktivitetsöverföring och stödja axonal hälsa i vit substans, är OPC rikliga (~ 5%) i grå substans där myelinering är knapp och utför aktivitetsberoende signalfunktioner för att styra inlärningsbeteende och minnesbildning 5,6,7,8 . Hur oligodendrogliala celler fungerar och dysfunktion i patogenesen av Alzheimers sjukdom (AD) och andra åldersassocierade neurodegenerativa tillstånd har understuderats9. Med tanke på att enskilda hjärncellstyper kan uppvisa distinkta och till och med motstridiga effekter inför samma AD-främjande förhållanden12,13, erbjuder denna stamcellsmetodik unikt celltypspecifik information som tidigare har missats med hjälp av etablerade in vivo- eller in vitro-modeller som endast ger aggregerade avläsningar från samlingar av hjärncellstyper. Under det senaste decenniet har ett stort antal tillförlitliga protokoll utvecklats för att generera mänskliga neuroner från transdifferentiering av ES / iPS-celler eller direkt omvandling från andra terminalt differentierade celltyper (t.ex. fibroblaster)14,15. I synnerhet kan tillämpningen av viktiga neurogena transkriptionsfaktorer (t.ex. neurogenin 2, Ngn2)16 på humana pluripotenta stamceller generera en homogen population av väl karakteriserade neuronala celltyper för rena kulturer utan behov av coculturing med gliaceller12,17,18. För inducerade humana oligodendrocyter finns det några publicerade protokoll som kan generera funktionella celler som mycket liknar deras primära motsvarigheter, med ett brett spektrum av effektivitet och efterfrågan i tid och resurser 19,20,21,22,23,24,25,26,27,28 . Hittills har inget av dessa protokoll tillämpats för att undersöka hur oligodendrogliala celler svarar på och påverkar AD-patogenes.
Här beskriver vi våra förbättrade protokoll för enstaka och blandade kulturer av humaninducerade neuroner (iNs) och OPCs/oligodendrocyter (iOPCs/iOLs). iN-protokollet som beskrivs här är baserat på den allmänt använda Ngn2-metoden16 och har den ytterligare funktionen att vara gliafri. De resulterande iN är homogena och liknar starkt de kortikala skiktet 2/3 excitatoriska neuroner, med karakteristisk pyramidal morfologi, genuttrycksmönster och elektrofysiologiska egenskaper17,18 (Figur 1). För att övervinna några av de grundläggande hindren för riktad differentiering av pluripotenta stamceller har vi utvecklat en enkel och effektiv metod för lågdos dimetylsulfoxid (DMSO) förbehandling29,30 och rapporterat en ökad benägenhet hos humana ES / iPS-celler att transdifferentiera till iOPCs och iOLs31, baserat på ett allmänt anpassat protokoll av Douvaras och Fossati 32 . Vi har ytterligare förenklat protokollet och införlivat en robust differentieringsfrämjande förening, clemastine 7,33,34, för att påskynda processen för oligodendroglial mognad. Som ett resultat (figur 2) kan iOPC: erna genereras på 2 veckor (~ 95% positiva för markören O4) och iOL på fyra veckor (uttrycker mogna markörer MBP och PLP1). Intressant nog fann vi att iOPCs ensamma utsöndrar en anmärkningsvärd mängd amyloid-β (Aβ), i överensstämmelse med de oberoende transkriptomiska data som visar det rikliga uttrycket av amyloidprekursorproteinet (APP) och bearbetningsproteas β-sekretas (BACE1) i oligodendrocythärstamningsceller35,36. Dessutom främjar vårt iN-iOPC-samodlingssystem inneslutningen av axoner genom MBP-positiva iOL-processer och ger betydande stöd för synapsbildning (figur 3). Således har de protokoll som vi har beskrivit nedan tekniska och biologiska fördelar jämfört med tidigare katalogiserade neuron-oligodendroglia co-culturing-metoder och har ett löfte om bättre modellering av neurodegenerationen i AD.

Vid Alzheimers sjukdom (AD) och andra neurodegenerativa sjukdomar är oligodendroglial misslyckande ett vanligt tidigt patologiskt drag, men hur det bidrar till sjukdomsutveckling och progression, särskilt i hjärnans grå substans, är fortfarande i stort sett okänt. Dysfunktionen hos oligodendrocytceller kännetecknas av brister i myelinering och nedsatt självförnyelse av oligodendrocytprekursorceller (OPC). Dessa två defekter orsakas åtminstone delvis av störningen av interaktioner mellan neuron och oligodendrocyter längs uppbyggnaden av patologi. OPC ger upphov till myeliniserande oligodendrocyter under CNS-utveckling. I den mogna hjärnbarken är OPC de viktigaste proliferativa cellerna (som omfattar ~ 5% av de totala hjärncellerna) och kontrollerar ny myelinbildning på ett neuralt aktivitetsberoende sätt. Sådan neuron-till-oligodendrocytkommunikation är signifikant understuderad, särskilt i samband med neurodegenerativa tillstånd såsom AD, på grund av bristen på lämpliga verktyg. Under de senaste åren har vår grupp och andra gjort betydande framsteg för att förbättra för närvarande tillgängliga protokoll för att generera funktionella neuroner och oligodendrocyter individuellt från humana pluripotenta stamceller. I detta manuskript beskriver vi våra optimerade procedurer, inklusive upprättandet av ett samodlingssystem för att modellera neuron-oligodendrocytanslutningarna. Våra illustrativa resultat tyder på ett oväntat bidrag från OPCs / oligodendrocyter till hjärnans amyloidos och synapsintegritet och belyser nyttan av denna metod för AD-forskning. Detta reduktionistiska tillvägagångssätt är ett kraftfullt verktyg för att dissekera de specifika heterocellulära interaktionerna ur den inneboende komplexiteten i hjärnan.
