Le vie cellulari che regolano gli eventi di sviluppo all'interno degli organismi sono altamente conservate tra tutti i metazoi. Una di queste vie è la cascata di segnalazione della proteina chinasi attivata dal mitogeno EGFR-RAS-ERK (MAPK) che è una via critica che governa la proliferazione cellulare, la differenziazione, la migrazione e la sopravvivenza1,2. I difetti in questa via di segnalazione possono portare a stati patologici o patologici come il cancro. Il recettore del fattore di crescita epidermico (EGFR) ha dimostrato di essere altamente espresso nei tumori umani, tra cui il 50% dei carcinomi a cellule squamose orali, e contribuisce allo sviluppo di tumori maligni3,4,5. Mentre le mutazioni nelle tre isoforme RAS H-, K- e N-RAS sono i principali driver per la trasformazione maligna in più tumori umani. Tra queste tre isoforme RAS, le mutazioni oncogeniche in K-RAS sono più diffuse6,7,8. Affinché EGFR e RAS funzionino, devono localizzarsi sulla membrana plasmatica (PM). Prevenire la localizzazione di queste molecole al PM può abrogare completamente l'attività biologica di questa via di segnale9,10. Quindi l'inibizione della localizzazione di queste proteine al PM è una strategia terapeutica per bloccare la segnalazione a valle e i conseguenti esiti avversi. Utilizzando un test di screening ad alto contenuto, la fendilina, un bloccante dei canali del calcio di tipo L, è stata identificata come un inibitore dell'attività K-RAS11. La nanoclustering di K-RAS al foglietto interno del PM è significativamente ridotta in presenza di fendilina. Inoltre, K-RAS viene ridistribuito dalla membrana plasmatica al reticolo endoplasmatico (ER), all'apparato di Golgi, agli endosomi e al citosol. Ancora più importante, la proliferazione delle linee cellulari tumorali pancreatiche, del colon, del polmone e dell'endometrio che esprimono il mutante oncogenico K-RAS è bloccata dall'inibizione della segnalazione a valle da parte della fendilina11. Questi dati suggeriscono che la fendilina funziona come una specifica terapia antitumorale K-RAS che causa l'errata localizzazione della proteina RAS al PM.
Il nematode Caenorhabditis elegans è stato ampiamente studiato nel contesto dello sviluppo. Molte delle vie del segnale che governano lo sviluppo nel worm sono evolutive e funzionalmente conservate. Ad esempio, l'attivazione mediata da EGFR di RAS e la successiva attivazione della cascata di segnali ERK MAPK sono conservate nel worm12. La cascata è rappresentata dalle seguenti proteine: LET-23 > LET-60 > LIN-45 > MEK-2 > MPK-1. LET-60 è omologo a RAS, mentre LET-23 è omologo a EGFR. Nel verme, questo percorso regola lo sviluppo della vulva13. La vulva è un'apertura epiteliale sulla parete del corpo ventrale del verme che consente di depostare le uova fecondate. La formazione della vulva nel worm dipende dall'esposizione delle cellule precursori vulvari (VPC) a un gradiente di attivazione della cascata di segnali EGFR-RAS-MAPK. Durante il normale sviluppo, i VPC prossimali ricevono forti segnali dalle cellule di ancoraggio gonadiche per differenziarsi in destini cellulari di 1° e 2° che danno origine a una vulva funzionale12. Mentre i VPC distali si differenziano in destini cellulari 3° che si fondono con il sincizio ipodermico e non formano la vulva a causa della segnalazione impoverita. In assenza di segnalazione, tutte le VPC si differenziano in destini cellulari 3° con conseguente formazione di nessuna vulva. Tuttavia, la segnalazione costitutiva porta alla formazione di una o più vulve non funzionali a causa dell'induzione di tutti i VPC ad assumere destini cellulari di 1° e 2°.
Mutazioni che causano un'induzione vulvale difettosa o eccessiva sono state identificate per molti dei geni che codificano per le proteine che rappresentano questa via. L'induzione vulvale difettosa provoca un fenotipo vulvaless (Vul), mentre un'eccessiva induzione vulvare si traduce in un fenotipo multivulva (Muv) che è rappresentato dallo sviluppo di numerose pseudovulve ectopiche non funzionali in tutta la parete del corpo ventrale. Il fenotipo Muv espresso dal ceppo let-60(n1046) è dovuto ad una mutazione gain of function in RAS, mentre nel ceppo let-23(sa62) è dovuto ad una mutazione attivante in EGFR14,15. Il fenotipo Muv forte in questi ceppi mutanti ha dimostrato di essere perturbato da interventi farmacologici come dimostrato dal trattamento dei vermi let-60(n1046) con l'inibitore MEK-1 U012616,17. È interessante notare che abbiamo dimostrato che R-fendilina e inibitori che influenzano il metabolismo della sfingomielina sopprimono il fenotipo Muv nel verme18. Per dimostrare che questi inibitori bloccano la segnalazione let-60 a livello di RAS, il ceppo nullo lin-1 è stato utilizzato17. Lin-1 è un fattore di trascrizione inibitorio simile a Ets che funziona come un repressore nello sviluppo della vulva19. La forte inversione del fenotipo Muv nei vermi let-60(n1046) e nessun effetto sui vermi nulli lin-1 suggeriscono che queste inibizioni si verificano a livello di RAS.
In questo protocollo, dimostriamo l'uso di C. elegans come modello per identificare inibitori delle proteine RAS ed EGFR. Utilizzando un test a base liquida, dimostriamo gli effetti inibitori della R-fendilina sopprimendo i fenotipi Muv nei ceppi mutanti let-60(n1046) e let-23(sa62) di C. elegans. Questo test convalida l'uso di C. elegans come strumento nella fase iniziale della scoperta di farmaci per terapie antitumorali.

I cambiamenti nella localizzazione della membrana plasmatica del recettore del fattore di crescita epidermico (EGFR) e del suo effettore a valle RAS sono stati implicati in diverse malattie tra cui il cancro. Il nematode C. elegans a vita libera possiede una cascata di segnali EGFR-RAS-ERK MAP evolutivamente conservata e funzionalmente conservata che è centrale per lo sviluppo della vulva. Le mutazioni di guadagno di funzione nell'omologo RAS LET-60 e nell'omologo EGFR LET-23 inducono la generazione di pseudovulva ectopica non funzionale visibile lungo la parete corporea ventrale di questi vermi. In precedenza, il fenotipo multivulvale (Muv) in questi vermi ha dimostrato di essere inibito da piccole molecole chimiche. Usando questo test, mostriamo che la R-fendilina, un inibitore indiretto di K-RAS, sopprime il fenotipo Muv espresso nei vermi mutanti let-60(n1046) e let-23(sa62). Il test è semplice, poco costoso, non richiede molto tempo per l'installazione e può essere utilizzato come piattaforma iniziale per la scoperta di terapie antitumorali.

Per prima cosa dimostriamo che la R-fendilina è in grado di sopprimere il fenotipo Muv nel ceppo mutante let-60 (n1046) rispetto ai vermi trattati con DMSO. I nostri dati mostrano che la R-fendilina è in grado di bloccare il fenotipo Muv nel let-60(n1046) in modo dose-dipendente (Figura 2A,B).
