脂質異常症は、世界の動脈硬化性疾患の主要な危険因子です。高濃度の低密度リポタンパク質(LDL)および低濃度の高密度リポタンパク質(HDL)は、冠状動脈性心疾患(CHD)1,2の高リスクと密接に関連している。臨床現場では、LDLコレステロール(LDL-C)とHDL-コレステロール(HDL-C)の両方が臨床検査室3,4の自動分析装置を用いて日常的に測定される。しかし、脂質異常症の診断やヒトおよび実験動物における脂質代謝とアテローム性動脈硬化症の研究については、リポタンパク質のプロファイルを詳細に分析することが不可欠です。超遠心分離、サイズ排除クロマトグラフィー(高速タンパク質液体クロマトグラフィー(FPLC)、高速液体クロマトグラフィー(HPLC)、アガロースおよびポリアクリルアミドゲルによる電気泳動、核磁気共鳴、ポリアニオンおよび二価カチオンなどの選択的化学沈殿を分析する方法がいくつか報告されている。1950年代、Havelのグループは、最初に超遠心分離を使用して密度によって定義されるリポタンパク質の概念を提案し、それらをチロミクロン(CM)、非常に低密度リポタンパク質(VLDL)、中間密度リポタンパク質(IDL)、LDL、およびHDL5以降に分類し、この方法は他のグループ6、7によってさらに改変された。これまで、超遠心分離は最も一般的で信頼性の高い方法ですが、実用的なプロトコルはまだ利用できません。本稿では、先に説明した逐次密度の超遠心分離を用いて、微小なプラズマを単離するための使いやすいプロトコルを説明することを試みた。7つの血漿リポタンパク質画分の単離 [VLDL (d<1.006 g/mL), IDL (d=1.02 g/mL), LL (d=1.04 および 1.06 g/mL), HDL (d=1.08, 1.10、および1.21 g/mL))は、研究者がリポタンパク質とその組成アポリポタンパク質9、10、11の両方の広範な分析を行うことを可能にする。無傷の7つの連続した密度リポタンパク質は、リポタンパク質機能を分析し、また細胞ベースのインビトロ戦略にも役立つことができます。このプロトコルは、臨床診断と基礎研究の両方に有用である必要があります。ここでは、他の種からのプラズマを同じように適用しながら、この技術を実証するために、例としてウサギのプラズマを使用しました。
