Hier verstrekken we de informatie die nodig is voor de bouw van een apparaat voor WBIS van muizen aan CS. Na installatie van het systeem is het van cruciaal belang dat onderzoekers het systeem kalibreren op basis van de geleverde dosis nicotine of cotinine bij dieren. Het roken van sigaretten blijft een van de belangrijkste oorzaken van vermijdbare ziekten in de VS, ondanks de gestage daling van het aantal volwassenen dat sigaretten rookt in de afgelopen 50-60 jaar1. Het is algemeen bekend dat roken verband houdt met meerdere ziekten van de longen en het bloed, waaronder chronische obstructieve longziekte (COPD), een groep ziekten die emfyseem en chronische bronchitis2,3,4omvat . Volgens het Center for Disease Control (CDC) was COPD in 2014 de derde belangrijkste doodsoorzaak in de Verenigde Staten met meer dan 15 miljoen Amerikanen die aan deze ziekte leden5.
CS is onlangs ook geassocieerd met een hoger risico op het ontwikkelen van klonale hematopoiese (CH)6,7, een aandoening waarbij een enkele hematopoëtische stamcel onevenredig een groot percentage van iemands perifere bloed produceert. Deze bevinding wijst op een mogelijk verband tussen roken en beenmergfunctie. Gezien de wijdverbreide en zeer significante gevolgen voor de gezondheid van CS en gezien het feit dat muriene modellen van ziekten een hoeksteen zijn van de vooruitgang in biomedisch onderzoek, is het nuttig om efficiënte en betaalbare systemen te ontwikkelen om CS bij muizen te modelleren.
Hier bieden we een stapsgewijze handleiding voor het bouwen van een betaalbaar systeem voor de behandeling en studie van de in vivo effecten van CS op longemfyseem en beenmerghomeostase. De assemblage van deze apparatuur vereist niet dat de gebruiker gespecialiseerde kennis heeft en maakt dus zelfmontage mogelijk.

Bijna 14% van de volwassenen in de Verenigde Staten zou in 2018 sigaretten roken. De effecten van sigarettenrook (CS) op longen en hart- en vaatziekten zijn uitgebreid bestudeerd, maar de impact van CS in andere weefsels en organen zoals bloed en beenmerg blijft onvolledig gedefinieerd. Het vinden van het juiste systeem om de effecten van CS bij knaagdieren te bestuderen kan onbetaalbaar zijn en vereist de aankoop van commercieel beschikbare systemen. Daarom wilden we een betaalbaar, betrouwbaar en veelzijdig systeem bouwen om de pathologische effecten van CS bij muizen te bestuderen. Deze opstelling van het hele lichaam inhalatieblootstellingssysteem (WBIS) bootst het ademen en puffen van sigaretten na door afwisselend blootstelling aan CS en schone lucht. Hier laten we zien dat dit doe-het-zelf (DIY) systeem luchtwegontsteking en longemfyseem veroorzaakt bij muizen na 4 maanden blootstelling aan sigarettenrook. De effecten van inademing van cs (WBI) in het hele lichaam op hematopoëtische stamcellen en voorlopercellen (HSPCs) in het beenmerg met behulp van dit apparaat worden ook aangetoond.

Een van de belangrijkste kenmerken van CS-blootstelling is emfyseem dat wordt gekenmerkt door de schade en vernietiging van luchtzakken (longblaasjes) in de long. De eerste experimenten richtten zich dus op het vermogen van het doe-het-zelfsysteem om emfysemateuze veranderingen in de longen van vrouwelijke muizen uit te lokken bij herhaalde blootstelling van het hele lichaam aan CS.
