Zigarettenrauchen ist nach wie vor eine der Hauptursachen für vermeidbare Krankheiten in den USA, trotz des stetigen Rückgangs der Zahl der zigarettenrauchenden Erwachsenen in den letzten 50-60 Jahren1. Es ist allgemein bekannt, dass Rauchen mit mehreren Erkrankungen der Lunge und des Blutes verbunden ist, einschließlich chronisch obstruktiver Lungenerkrankung (COPD), einer Gruppe von Krankheiten, die Emphysem und chronische Bronchitis2,3,4umfasst. Nach Angaben des Center for Disease Control (CDC), im Jahr 2014, COPD war die dritthäufigste Todesursache in den Vereinigten Staaten mit über 15 Millionen Amerikaner n.H.
CS wurde auch vor kurzem mit einem höheren Risiko der Entwicklung klonaler Hämatopoese (CH)6,7verbunden, ein Zustand, in dem eine einzelne hämatopoetische Stammzelle überproportional einen großen Prozentsatz des peripheren Blutes einer Person produziert. Dieser Befund deutet auf einen möglichen Zusammenhang zwischen Rauchen und Knochenmarkfunktion hin. Angesichts der weitverbreiteten und sehr signifikanten gesundheitlichen Auswirkungen von CS und angesichts der Tatsache, dass murine Modelle von Krankheiten ein Eckpfeiler des Fortschritts in der biomedizinischen Forschung sind, ist es nützlich, effiziente und erschwingliche Systeme zu entwickeln, um CS bei Mäusen zu modellieren.
Hier bieten wir eine Schritt-für-Schritt-Anleitung für den Aufbau eines erschwinglichen Systems zur Behandlung und Untersuchung der In-vivo-Effekte von CS auf Lungenemphysem und Knochenmarkhomöostase. Die Montage dieser Geräte erfordert nicht, dass der Benutzer spezielle Kenntnisse hat und ermöglicht somit die DIY-Montage.

Im Jahr 2018 sollen fast 14 % der Erwachsenen in den Vereinigten Staaten Zigaretten geraucht haben. Die Auswirkungen von Zigarettenrauch (CS) auf Lungen und Herz-Kreislauf-Erkrankungen wurden weithin untersucht, jedoch bleiben die Auswirkungen von CS in anderen Geweben und Organen wie Blut und Knochenmark unvollständig definiert. Die Suche nach dem geeigneten System zur Untersuchung der Auswirkungen von CS bei Nagetieren kann unerschwinglich teuer sein und den Kauf kommerziell erhältlicher Systeme erfordern. So haben wir uns vorgenommen, ein erschwingliches, zuverlässiges und vielseitiges System zu entwickeln, um die pathologischen Wirkungen von CS bei Mäusen zu untersuchen. Diese Ganzkörper-Inhalations-Expositions-System (WBIS) Setup imitiert das Atmen und Puffen von Zigaretten durch abwechselnde Exposition gegenüber CS und saubere Luft. Hier zeigen wir, dass dieses Do-it-yourself (DIY) System nach 4 Monaten Zigarettenrauch-Exposition Atemwegsentzündungen und Lungenemphysem bei Mäusen induziert. Die Auswirkungen der Ganzkörperinhalation (WBI) von CS auf hämatopoetische Stamm- und Vorläuferzellen (HSPCs) im Knochenmark mit diesem Gerät werden ebenfalls gezeigt.

Eines der Hauptmerkmale der CS-Exposition ist Emphysem, das durch die Beschädigung und Zerstörung von Luftsäcken (Alveolen) in der Lunge gekennzeichnet ist. So konzentrierten sich erste Experimente auf die Fähigkeit des DIY-Systems, emphysematous Veränderungen in der Lunge weiblicher Mäuse bei wiederholter Ganzkörperexposition gegenüber CS zu provozieren.
