Bovendien veroorzaakt S. Enteritidis latente infecties bij volwassen kippen en chronische dragers besmetten pluimveeproducten, wat resulteert in door voedsel overgedragen infecties bij mensen19. Niet-coderende kleine RNA's (sRNA's) zijn 40-400 nucleotiden lang, die over het algemeen geen eiwitten coderen, maar onafhankelijk kunnen worden getranscribeerd in bacteriële chromosomen 1,2,3. De meeste sRNA's zijn gecodeerd in de intergene regio's (IGR's) tussen gencoderende regio's en interageren met doel-mRNA's door middel van base-pairing acties, en reguleren de expressie van doelgenen op het post-transcriptionele niveau 4,5. Ze spelen een belangrijke regulerende rol in het stofmetabolisme, de synthese van buitenmembraaneiwitten, quorumdetectie en virulentiegenexpressie5.
MicC is een 109-nucleotide klein RNA-transcript aanwezig in Escherichia coli en Salmonella enterica serovar Typhimurium, dat meerdere buitenmembraaneiwitexpressie zoals OmpC, OmpD, OmpN, Omp35 en Omp36 6,7,8,9 zou kunnen reguleren. MicC reguleert de expressie van OmpC door ribosoombinding aan de ompC mRNA-leider in vitro te remmen en het vereist de Hfq RNA-chaperonne voor zijn functie in Escherichia coli6. In Salmonella Typhimurium legt MicC ompD-mRNA het zwijgen op via een ≤12-bp RNA-duplex binnen de coderende sequentie (codons 23-26) en destabiliseert vervolgens endonucleolytisch mRNA7. Dit regulatieproces wordt ondersteund door chaperonne-eiwit Hfq10. De OmpC is een overvloedig buitenmembraaneiwit waarvan werd gedacht dat het belangrijk was in omgevingen waar de concentraties voedingsstoffen en toxines hoog waren, zoals in de darm6. De OmpD porine is het meest voorkomende buitenmembraaneiwit in Salmonella Typhimurium en vertegenwoordigt ongeveer 1% van het totale celeiwit11. OmpD is betrokken bij de hechting aan menselijke macrofagen en darmepitheelcellen12. MicC onderdrukt ook de expressie van zowel OmpC- als OmpD-porines. Er wordt gedacht dat MicC virulentie kan reguleren. Om nieuwe doelgenen gereguleerd door MicC te onderzoeken en de virulentieregulatiefunctie van micC te bestuderen, kloonden we het micC-gen in de Salmonella Enteritidis (SE) stam 50336 en construeerden vervolgens de mutant 50336ΔmicC en de aangevulde mutant 50336ΔmicC / p micC. Nieuwe doelgenen werden gescreend door qRT-PCR. De virulentie van 50336ΔmicC werd gedetecteerd door muizen en kippeninfecties.

Een niet-coderend klein RNA (sRNA) is een nieuwe factor om genexpressie op post-transcriptioneel niveau te reguleren. Een soort sRNA MicC, bekend in Escherichia coli en Salmonella Typhimurium, zou de expressie van buitenmembraaneiwitten kunnen onderdrukken. Om de regulatiefunctie van micC in Salmonella Enteritidis verder te onderzoeken, kloonden we het micC-gen in de Salmonella Enteritidis-stam 50336 en construeerden vervolgens de mutant 50336Δ micC door het op λ Red gebaseerde recombinatiesysteem en de aangevulde mutant 50336Δ micC / p micC met recombinant plasmide pBR322 die micC tot expressie brengt. qRT-PCR-resultaten toonden aan dat transcriptie van ompD in 50336Δ micC 1,3-voudig hoger was dan die in de wildtype stam, terwijl de transcriptie van ompA en ompC in 50336Δ micC 2,2-voudig en 3-voudig hoger was dan die in de wildtype stam. Deze gaven aan dat micC de expressie van ompA en ompC onderdrukt. In de volgende studie werd de pathogeniciteit van 50336ΔmicC gedetecteerd door zowel 6 weken oude Balb / c-muizen als 1-dag-oude kippen te infecteren. Het resultaat toonde aan dat de LD 50 van de wilde stam 50336, de mutanten 50336Δ micC en 50336ΔmicC/pmicC voor 6 weken oude Balb/c muizen respectievelijk 12,59 CFU, 5,01 CFU en 19,95 CFU waren. De LD 50 van de stammen voor 1-dag-oude kippen waren respectievelijk 1,13 x 109 CFU, 1,55 x 10 8 CFU en2,54 x 10 8 CFU. Het gaf aan dat deletie van micC de virulentie van S verhoogde. Enteritidis bij muizen en kippen door de expressie van buitenmembraaneiwitten te reguleren.

Dit protocol kan worden gebruikt voor het inactiveren van chromosomale genen om mutanten te isoleren voor genfunctiestudie en bacteriën. Het maakt het homologe recombinatieproces eenvoudig, snel en zeer efficiënt. Met behulp van deze techniek kunnen de virulentiegerelateerde genen van het pathogeen worden verwijderd om de verzwakte mutant te verkrijgen die kan worden gebruikt als kandidaat voor het verzwakte vaccin.Begin met het versterken van de chlooramfenicolcassette met hemologiefragmenten van het micC-gen. Ontwerp voorwaartse en omgekeerde primers om de chlooramfenicolcassette van plasmide PKD3 te versterken, inclusief 50 basepaar homologie-uitbreidingen van de vijf priem- en drie priemuiteinden van het micC-gen. Voer PCR uit zoals beschreven in het tekstmanuscript met behulp van het PKD3-plasmide als sjabloon om de chlooramfenicolcassette te versterken.Bepaal de grootte van het PCR-product met agarosegel-elektroforese. Zuiver het product vervolgens met een DNA-gelherstelkit en bepaal de concentratie van DNA door spectrofotometrie. Voer een tweede PCR-reactie uit om de interferentie van verdere recombinatie door het PKD3-plasmide te elimineren.Verdun het eerste PCR-product in een verhouding van 1 tot 200 en gebruik het als sjabloon voor de secundaire PCR. Om de eerste recombinante stam 50336 delta micC dot cat te construeren, mengt u 100 microliter SE 50336 competente cellen met vijf microliter PKD 46 plasmide en incubeert u gedurende 30 minuten op ijs. Hitteschok het mengsel bij 42 graden Celsius gedurende 90 seconden en breng het snel terug op ijs gedurende twee minuten om het plasmide in de cellen te transformeren.Screen positieve kolonies door de cellen 's nachts bij 30 graden Celsius te kweken op een ampicillineresistente plaat. Voeg de volgende dag 30 milli meer L arabinose toe aan de SE 50336 PKD 46 vloeibare cultuur en induceer recombinase-expressie met een incubatie van één uur bij 30 graden Celsius tijdens het schudden. Verdeel het mengsel op een chlooramfenicol-resistente LB-plaat en kweek 's nachts bij 37 graden Celsius om te screenen op positieve kolonies.Kweek de positieve kolonies twee uur lang op 42 graden Celsius. Vervolgens wordt de kolonie 's nachts gescreend op gevoeligheid voor ampicilline en resistentie tegen chlooramfenicol bij 37 graden Celsius om de eerste recombinante stam zonder PKD 46 te verkrijgen. Na het extraheren van de 50336 Delta micC dot cat genomisch DNA met behulp van PCR en gel elektroforese, galvaniseerde 100 nanogram plasmide PCP 20 in 40 microliter van 50336 Delta micC dot cat competente cellen.Screen positieve transformanten op zowel ampicilline als chlooramfenicol resistente platen bij 30 graden Celsius. Breng de positieve transformanten over in niet-resistent LB vloeibaar medium en kweek ze 's nachts bij 42 graden Celsius. Isoleer vervolgens enkele kolonies op een LB-plaat bij 37 graden Celsius.Selecteer de kolonie die gevoelig is voor zowel ampicilline als chlooramfenicol. Deze mutant is de micC deletie mutant SE 50336 Delta micC. Controleer 50336 Delta micC door PCR uit te voeren zoals beschreven in het tekstmanuscript.Dit protocol werd gebruikt om de deletiemutant 50336 Delta micC te construeren in de gecomplimenteerde mutant 50336 Delta micC, pmicC. De eerste recombinante 50336 Delta micC dot cat werd gevalideerd door PCR met een verwachte bandgrootte van ongeveer 1200 basenparen PCR-producten met de chlooramfenicol-insertie vergeleken met 279 basenparen in de wildtypestam. Bij de tweede recombinatie werd de chlooramfenicolcassette geëlimineerd door PCP 20.De PCR-resultaten in combinatie met sequencing bevestigden dat de isogene micC-mutant met succes was geconstrueerd. De expressie van ompA-, ompC- en ompD-genen in stammen 50336, 50336 Delta micC en 50336 delta micC pmicC werden geanalyseerd door realtime kwantitatieve PCR. Transcriptie van ompA en ompC en 50336 delta micC nam ongeveer 2,2 keer toe en verdrievoudigde in vergelijking met de wildtype soort.Terwijl de transcriptie van ompD slechts licht toenam. LD 50-testen toonden aan dat na het infecteren van zes tot acht weken oude BALB / c-muizen met elk van de drie stammen, de meeste muizen geïnfecteerd door 50336 Delta micC 48 uur na infectie lassitude, inappetentie of diarree vertoonden en na 96 uur stierven. De muizen geïnfecteerd door wild type stam en 50336 delta micC pmicC vertoonden dezelfde symptomen 72 uur na infectie en stierven na 120 uur.De LD 50's van de drie stammen gaven aan dat de virulentie van de mutant 50336 Delta micC 2,5 keer verbeterde in vergelijking met het wilde type. Deze techniek helpt onderzoekers de functie van bacteriële genen te bestuderen en de gewenste gemanipuleerde stam te verkrijgen door doelgenen te verwijderen.
