L. Enteritidis est un important agent pathogène intracellulaire facultatif qui peut infecter les jeunes poulets et produire des symptômes allant de l’entérite à l’infection systémique et à la mort17,18. De plus, S. Enteritidis provoque des infections latentes chez les poulets adultes et les porteurs chroniques contaminent les produits avicoles, ce qui entraîne des infections d’origine alimentaire chez les humains19...

Les petits ARN non codants (ARNs) ont une longueur de 40 à 400 nucléotides, qui ne codent généralement pas pour des protéines mais pourraient être transcrits indépendamment dans les chromosomes bactériens 1,2,3. La plupart des ARNs sont codés dans les régions intergéniques (IGR) entre les régions codant pour les gènes et interagissent avec les ARNm cibles par des actions d’appariement de bases, et régulent l’expression des gènes cibles au niveau post-transcriptionnel 4,5. Ils jouent un rôle régulateur important dans le métabolisme des substances, la synthèse des protéines de la membrane externe, la détection du quorum et l’expression des gènes de virulence5.
MicC est un petit transcrit d’ARN de 109 nucléotides présent dans Escherichia coli et Salmonella enterica sérovar Typhimurium, qui pourrait réguler l’expression de plusieurs protéines de la membrane externe telles que OmpC, OmpD, OmpN, Omp35 et Omp36 6,7,8,9. MicC régule l’expression de l’OmpC en inhibant la liaison du ribosome au leader de l’ARNm ompC in vitro et nécessite le chaperon d’ARN Hfq pour sa fonction chez Escherichia coli6. Chez Salmonella Typhimurium, MicC réduit au silence l’ARNm ompD via un duplex d’ARN ≤12-bp dans la séquence codante (codons 23-26) puis déstabilise l’ARNm endonucléolytique7. Ce processus de régulation est assisté par la protéine chaperonne Hfq10. L’OmpC est une protéine membranaire externe abondante que l’on croyait importante dans les environnements où les concentrations de nutriments et de toxines étaient élevées, comme dans l’intestin6. La porine OmpD est la protéine membranaire externe la plus abondante chez Salmonella Typhimurium et représente environ 1% de la protéine cellulaire totale11. L’OmpD est impliqué dans l’adhérence aux macrophages humains et aux cellules épithéliales intestinales12. MicC réprime également l’expression des porines OmpC et OmpD. On pense que MicC peut réguler la virulence. Pour explorer de nouveaux gènes cibles régulés par MicC et étudier la fonction de régulation de la virulence de micC, nous avons cloné le gène micC dans la souche 50336 de Salmonella Enteritidis (SE), puis construit le mutant 50336ΔmicC et le mutant complété 50336ΔmicC/p micC. De nouveaux gènes cibles ont été criblés par qRT-PCR. La virulence de 50336ΔmicC a été détectée par des souris et des infections de poulet.

Un petit ARN non codant (ARNs) est un nouveau facteur pour réguler l’expression des gènes au niveau post-transcriptionnel. Une sorte d’ARNs MicC, connu chez Escherichia coli et Salmonella Typhimurium, pourrait réprimer l’expression des protéines de la membrane externe. Pour étudier plus avant la fonction de régulation du micC chez Salmonella Enteritidis, nous avons cloné le gène micC dans la souche 50336 de Salmonella Enteritidis, puis construit le mutant 50336Δ micC par le système de recombinaison à base de λ Red et le mutant 50336Δ micC/p micC complété portant le plasmide recombinant pBR322 exprimant micC. Les résultats de la qRT-PCR ont démontré que la transcription de l’ompD dans 50336Δ micC était 1,3 fois plus élevée que celle de la souche de type sauvage, tandis que la transcription de ompA et ompC dans 50336ΔmicC était 2,2 fois et 3 fois plus élevée que celles de la souche de type sauvage. Ceux-ci indiquent que micC réprime l’expression de ompA et ompC. Dans l’étude suivante, la pathogénicité de 50336ΔmicC a été détectée par des souris Balb/c âgées de 6 semaines et des poulets âgés de 1 jour. Le résultat a montré que la DL50 de la souche de type sauvage 50336, les mutants 50336Δ micC et 50336Δ micC/p micC pour les souris Balb/c âgées de 6 semaines étaient respectivement de 12,59UFC, 5,01 UFC et 19,95 UFC. La DL 50 des souches pour les poulets âgés de 1 jour était de 1,13 x 109 UFC, 1,55 x 10 8 UFC et2,54 x 10 8 UFC, respectivement. Il a indiqué que la délétion de micC améliorait la virulence de S. Enteritidis chez la souris et le poulet en régulant l’expression des protéines de la membrane externe.

Ce protocole peut être utilisé pour inactiver les gènes chromosomiques afin d’isoler les mutants pour l’étude de la fonction des gènes et des bactéries. Il rend le processus de recombinaison homologue simple, rapide et très efficace. En utilisant cette technique, les gènes liés à la virulence de l’agent pathogène peuvent être supprimés pour obtenir le mutant atténué qui peut être utilisé comme candidat pour le vaccin atténué.Commencez par amplifier la cassette de chloramphénicol contenant des fragments hémologiques du gène micC. Concevoir des amorces avant et arrière pour amplifier la cassette de chloramphénicol à partir du plasmide PKD3, y compris 50 extensions d’homologie de paires de bases des cinq extrémités premières et des trois extrémités premières du gène micC. Effectuer la PCR comme décrit dans le manuscrit en utilisant le plasmide PKD3 comme modèle pour amplifier la cassette de chloramphénicol.Déterminer la taille du produit PCR avec électrophorèse sur gel d’agarose. Purifiez ensuite le produit avec un kit de récupération de gel d’ADN et déterminez la concentration d’ADN par spectrophotométrie. Effectuer une deuxième réaction PCR pour éliminer l’interférence d’une recombinaison ultérieure par le plasmide PKD3.Diluez le premier produit de PCR dans un rapport de 1 à 200 et utilisez-le comme modèle pour la PCR secondaire. Pour construire la première souche recombinante 50336 delta micC dot cat, mélanger 100 microlitres de cellules compétentes SE 50336 avec cinq microlitres de plasmide PKD 46 et incuber sur de la glace pendant 30 minutes. Choquer le mélange à 42 degrés Celsius pendant 90 secondes et le transférer rapidement sur la glace pendant deux minutes pour transformer le plasmide en cellules.Dépistage des colonies positives en cultivant les cellules pendant la nuit à 30 degrés Celsius sur une plaque résistante à l’ampicilline. Le lendemain, ajoutez 30 milli de L arabinose supplémentaire à la culture liquide SE 50336 PKD 46 et induisez l’expression de la recombinase avec une incubation d’une heure à 30 degrés Celsius tout en agitant. Mélanger 100 nanogrammes du produit PCR purifié et 40 microlitres de cellules compétentes SE 50336 PKD 46 dans une coupelle à choc électrique.Ensuite, effectuez la transformation par choc électrique. Après électrotransformation, transférer le mélange dans un millilitre de milieu SOC et un incubateur de culture à agitation à 150 tr / min et 30 degrés Celsius pendant une heure. Étaler le mélange sur une plaque LB résistante au chloramphénicol et la cultiver à 37 degrés Celsius pendant la nuit pour dépister les colonies positives.Culture des colonies positives à 42 degrés Celsius pendant deux heures. Ensuite, la colonie a examiné la sensibilité à l’ampicilline et la résistance au chloramphénicol à 37 degrés Celsius pendant la nuit pour obtenir la première souche recombinante sans PKD 46. Après avoir extrait l’ADN génomique du chat point 50336 Delta micC par PCR et électrophorèse sur gel, électroplaque 100 nanogrammes de plasmide PCP 20 dans 40 microlitres de cellules compétentes 50336 Delta micC point cat.Criblez les transformants positifs sur des plaques résistantes à l’ampicilline et au chloramphénicol à 30 degrés Celsius. Transférer les transformants positifs dans un milieu liquide LB non résistant et les cultiver pendant la nuit à 42 degrés Celsius. Ensuite, isolez les colonies individuelles sur une plaque LB à 37 degrés Celsius.Sélectionnez la colonie sensible à l’ampicilline et au chloramphénicol. Ce mutant est le mutant de délétion micC SE 50336 Delta micC. Vérifiez 50336 Delta micC en effectuant une PCR comme décrit dans le manuscrit texte.Ce protocole a été utilisé pour construire le mutant de délétion 50336 Delta micC dans le mutant 50336 Delta micC complimenté, pmicC. Le premier chat recombinant 50336 Delta micC point cat a été validé par PCR avec une taille de bande attendue d’environ 1200 paires de bases de produits PCR avec l’insertion de chloramphénicol contre 279 paires de bases dans la souche de type sauvage. Dans la deuxième recombinaison, la cassette de chloramphénicol a été éliminée par PCP 20.Les résultats de la PCR combinés au séquençage ont confirmé que le mutant micC isogénique a été construit avec succès. L’expression des gènes ompA, ompC et ompD dans les souches 50336, 50336 Delta micC et 50336 delta micC pmicC a été analysée par PCR quantitative en temps réel. La transcription de ompA et ompC et 50336 delta micC a augmenté d’environ 2,2 fois et triplé par rapport à la souche de type sauvage.Alors que la transcription de ompD n’a augmenté que légèrement. Les tests DL 50 ont montré qu’après avoir infecté des souris BALB / c âgées de six à huit semaines avec chacune des trois souches, la plupart des souris infectées par 50336 Delta micC présentaient une lassitude, une inappétence ou une diarrhée 48 heures après l’infection et mouraient après 96 heures. Les souris infectées par la souche de type sauvage et 50336 delta micC pmicC ont présenté les mêmes symptômes 72 heures après l’infection et sont mortes après 120 heures.Les DL 50 des trois souches ont indiqué que la virulence du mutant 50336 Delta micC était multipliée par 2,5 par rapport au type sauvage. Cette technique aide les chercheurs à étudier la fonction des gènes bactériens et à obtenir la souche modifiée souhaitée en supprimant les gènes cibles.
