S. Enteritidis ist ein wichtiger fakultativer intrazellulärer Erreger, der junge Hühner infizieren kann und Symptome von Enteritis bis hin zu systemischer Infektion und Tod hervorruft17,18. Enteritidis latente Infektionen bei erwachsenen Hühnern, und chronische Träger kontaminieren Geflügelprodukte, was beim Menschen zu lebensmittelbedingten Infektionen führt19. Nicht-kodierende kleine RNAs (sRNAs) sind 40-400 Nukleotide lang, die im Allgemeinen keine Proteine kodieren, aber unabhängig voneinander in bakteriellen Chromosomen transkribiert werden können 1,2,3. Die meisten sRNAs kodieren in den intergenen Regionen (IGRs) zwischen genkodierenden Regionen und interagieren mit Ziel-mRNAs durch Basenpaarungsaktionen und regulieren die Expression von Zielgenen auf posttranskriptioneller Ebene 4,5. Sie spielen eine wichtige Rolle bei der Regulation des Stoffstoffwechsels, der Proteinsynthese der äußeren Membran, der Quorum-Sensing und der Virulenz-Genexpression5.
MicC ist ein kleines RNA-Transkript mit 109 Nukleotiden, das in Escherichia coli und Salmonella enterica serovar Typhimurium vorkommt und die Expression mehrerer Proteine der äußeren Membran wie OmpC, OmpD, OmpN, Omp35 und Omp36 regulieren könnte 6,7,8,9. MicC reguliert die Expression von OmpC, indem es die Bindung des Ribosoms an den ompC-mRNA-Leader in vitro hemmt, und benötigt das Hfq-RNA-Chaperon für seine Funktion in Escherichia coli6. In Salmonella Typhimurium schaltet MicC ompD-mRNA über einen ≤12-bp-RNA-Duplex innerhalb der kodierenden Sequenz (Codons 23-26) aus und destabilisiert dann die endonukleolytische mRNA7. Dieser Regulationsprozess wird durch das Chaperonprotein Hfq10 unterstützt. Das OmpC ist ein reichlich vorhandenes äußeres Membranprotein, von dem angenommen wurde, dass es in Umgebungen mit hohen Nährstoff- und Toxinkonzentrationen, wie z. B. im Darm, wichtig ist6. Das OmpD-Porin ist das am häufigsten vorkommende äußere Membranprotein in Salmonella Typhimurium und macht etwa 1% des gesamten Zellproteins11 aus. OmpD ist an der Adhärenz an menschlichen Makrophagen und Darmepithelzellen beteiligt12. MicC unterdrückt auch die Expression von OmpC- und OmpD-Porinen. Es wird vermutet, dass MicC die Virulenz regulieren kann. Um neue Zielgene zu erforschen, die durch MicC reguliert werden, und um die Virulenzregulationsfunktion von micC zu untersuchen, klonierten wir das micC-Gen im Salmonella Enteritidis (SE)-Stamm 50336 und konstruierten dann die Mutante 50336ΔmicC und die komplementäre Mutante 50336ΔmicC/p micC. Neuartige Zielgene wurden mittels qRT-PCR gescreent. Die Virulenz von 50336ΔmicC wurde durch Mäuse- und Hühnerinfektionen nachgewiesen.

a non-coding small rna micc contributes to virulence in outer membrane proteins in salmonella enteritidis

Eine nicht-kodierende kleine RNA (sRNA) ist ein neuer Faktor zur Regulierung der Genexpression auf posttranskriptioneller Ebene. Eine Art sRNA MicC, wie sie in Escherichia coli und Salmonella Typhimurium bekannt ist, könnte die Expression von Proteinen der äußeren Membran unterdrücken. Um die Regulationsfunktion von micC in Salmonella Enteritidis weiter zu untersuchen, klonierten wir das micC-Gen im Salmonella Enteritidis-Stamm 50336 und konstruierten dann die Mutante 50336Δ micC durch das λ Red-basierte Rekombinationssystem und die komplementäre Mutante 50336Δ micC/p micC, die das rekombinante Plasmid pBR322 trägt, das micC exprimiert. Die Ergebnisse der qRT-PCR zeigten, dass die Transkription von ompD in 50336Δ micC 1,3-fach höher war als die im Wildtyp-Stamm, während die Transkription von ompA und ompC in 50336ΔmicC 2,2-fach und 3-fach höher war als im Wildtyp-Stamm. Diese deuteten darauf hin, dass micC die Expression von ompA und ompC unterdrückt. In der folgenden Studie wurde die Pathogenität von 50336ΔmicC sowohl durch die Infektion von 6 Wochen alten Balb/c-Mäusen als auch durch 1 Tag alte Hühner nachgewiesen. Das Ergebnis zeigte, dass die LD 50 des Wildtyp-Stammes 50336, die Mutanten 50336Δ micC und 50336Δ micC/pmicC für 6 Wochen alte Balb/c-Mäuse 12,59 KBE, 5,01 KBE bzw. 19,95 KBE betrugen. Die LD 50 der Stämme für 1 Tag alte Hühner betrugen 1,13 x 109 KBE, 1,55 x 10 8 KBE bzw.2,54 x 10 8 KBE. Es zeigte sich, dass die Deletion von micC die Virulenz von S erhöht. Die Expression der ompA-, ompC- und ompD-Gene in den Stämmen 50336, 50336 Delta micC und 50336 delta micC pmicC wurde mittels quantitativer realtime PCR analysiert. Die Transkription von ompA und ompC sowie 50336 delta micC erhöhte sich im Vergleich zum Wildtyp-Stamm um das 2,2-fache und dreifache.Während die Transkription von ompD nur geringfügig zunahm. LD 50-Assays zeigten, dass die meisten Mäuse, die mit 50336 Delta micC infiziert waren, nach der Infektion von sechs bis acht Wochen alten BALB/c-Mäusen mit jedem der drei Stämme 48 Stunden nach der Infektion Müdigkeit, Appetitlosigkeit oder Durchfall zeigten und nach 96 Stunden starben. Die Mäuse, die mit dem Wildtyp-Stamm und 50336 delta micC pmicC infiziert waren, zeigten 72 Stunden nach der Infektion die gleichen Symptome und starben nach 120 Stunden.Die LD 50 der drei Stämme zeigten, dass die Virulenz der Mutante 50336 Delta micC im Vergleich zum Wildtyp um das 2,5-fache anstieg. Diese Technik hilft Forschern, die Funktion bakterieller Gene zu untersuchen und den gewünschten gentechnisch veränderten Stamm zu erhalten, indem Zielgene gelöscht werden.
