ノンコード低分子RNA(sRNA)は40〜400ヌクレオチド長であり、一般にタンパク質をコードしないが、細菌染色体1,2,3で独立して転写することができる。ほとんどのsRNAは、遺伝子コード領域間の遺伝子間領域(IGR)にコードされており、塩基対形成作用を介して標的mRNAと相互作用し、転写後レベルで標的遺伝子の発現を調節しています4,5。それらは、物質代謝、外膜タンパク質合成、クォーラムセンシング、および病原性遺伝子発現において重要な調節的役割を果たします5。
MicCは、大腸菌およびサルモネラ・エンテリカ・セロバー・チフィムリウムに存在する109ヌクレオチドの低分子RNA転写産物であり、OmpC、OmpD、OmpN、Omp35およびOmp36 6,7,8,9などの複数の外膜タンパク質発現を調節することができる。MicCは、in vitroでompC mRNAリーダーへのリボソーム結合を阻害することによってOmpCの発現を調節し、大腸菌での機能のためにHfq RNAシャペロンを必要とします6。ネズミチフス菌では、MicCはコード配列(コドン23〜26)内の≤12 bp RNA二重鎖を介してompD mRNAをサイレンシングし、エンドヌクレアーゼmRNA7を不安定にします。この調節プロセスは、シャペロンタンパク質Hfq10によって支援される。OmpCは豊富な外膜タンパク質であり、腸管など栄養・毒素濃度が高い環境では重要であると考えられていました6。OmpDポリンは、ネズミチフス菌の中で最も豊富な外膜タンパク質であり、全細胞タンパク質の約1%を占めています11。OmpDは、ヒトマクロファージおよび腸上皮細胞への接着に関与する12。MicCはまた、OmpCおよびOmpDポリンの両方の発現を抑制する。MicCは病原性を調節すると考えられています。MicCによって制御される新しい標的遺伝子を探索し、micCの病原性制御機能を研究するために、サルモネラ・エンテリティディス(SE)株50336のmicC遺伝子をクローニングし、変異体50336ΔmicCと相補変異体50336ΔmicC/pmicCを構築しました。新規標的遺伝子をqRT-PCRによりスクリーニングした。50336ΔmicCの病原性は、マウスおよびニワトリ感染によって検出された。

ノンコーディング低分子RNA(sRNA)は、転写後レベルで遺伝子発現を制御する新しい因子です。大腸菌やネズミチフス菌で知られている一種のsRNA MicCは、外膜タンパク質の発現を抑制する可能性があります。サルモネラ・エンテリティディスにおけるmicCの制御機能をさらに調べるために、サルモネラ・エンテリティディス50336株のmicC遺伝子をクローニングし、λRedベースの組換えシステムによって変異体50336Δ micCを構築し、miccを発現する組換えプラスミドpBR322を保有する相補変異体50336ΔmicC/p micCを構築しました。 qRT-PCRの結果、50336Δ micCにおけるompDの転写は野生株のそれより1.3倍高く、50336ΔmicCにおけるompAおよびompCの転写は野生株のそれより2.2倍および3倍高いことが示された。 これらは、micCがompAおよびompCの発現を抑制することを示した。以下の研究では、50336ΔmicCの病原性は、6週齢のBalb / cマウスと1日齢のニワトリの両方に感染することによって検出されました。その結果、6週齢のBalb/cマウスの野生株50336のLD50、変異株50336ΔmicCおよび50336Δ micC/pmicCは、それぞれ12.59 CFU、5.01 CFU、および19.95 CFUであることが示された。1日齢ニワトリのLD50株は,それぞれ1.13 x 109 CFU,1.55 x 10 8 CFU,2.54 x 108 CFUであった。micCの欠失がSの病原性を亢進させることを示した。 外膜タンパク質の発現を調節することにより、マウスおよびニワトリにおける腸炎。
