Los ARN pequeños no codificantes (ARNs) tienen una longitud de 40-400 nucleótidos, que generalmente no codifican proteínas, pero podrían transcribirse independientemente en cromosomas bacterianos 1,2,3. La mayoría de los ARNs están codificados en las regiones intergénicas (IGR) entre las regiones codificantes de genes e interactúan con los ARNm diana a través de acciones de apareamiento de bases, y regulan la expresión de genes diana a nivel post-transcripcional 4,5. Desempeñan importantes funciones de regulación en el metabolismo de sustancias, la síntesis de proteínas de la membrana externa, la detección de quórum y la expresión génica de virulencia5.
MicC es un pequeño transcrito de ARN de 109 nucleótidos presente en Escherichia coli y Salmonella enterica serovar Typhimurium, que podría regular la expresión de múltiples proteínas de membrana externa como OmpC, OmpD, OmpN, Omp35 y Omp36 6,7,8,9. MicC regula la expresión de OmpC inhibiendo la unión del ribosoma al ARNm de ompC líder in vitro y requiere la chaperona de ARN Hfq para su función en Escherichia coli6. En Salmonella Typhimurium, MicC silencia el ARNm ompD a través de un ARN dúplex de ≤12 pb dentro de la secuencia codificante (codones 23-26) y luego desestabiliza el ARNm endonucleolítico7. Este proceso de regulación es asistido por la proteína chaperona Hfq10. La OmpC es una proteína abundante de la membrana externa que se pensaba que era importante en ambientes donde las concentraciones de nutrientes y toxinas eran altas, como en el intestino6. La porina OmpD es la proteína de la membrana externa más abundante en Salmonella Typhimurium y representa aproximadamente el 1% de la proteína celular total11. OmpD está implicado en la adhesión a macrófagos humanos y células epiteliales intestinales12. MicC también reprime la expresión de las porinas OmpC y OmpD. Se cree que MicC puede regular la virulencia. Para explorar nuevos genes diana regulados por MicC y estudiar la función de regulación de virulencia de micC, clonamos el gen micC en la cepa 50336 de Salmonella Enteritidis (SE), y luego construimos el mutante 50336ΔmicC y el mutante complementario 50336ΔmicC/p micC. Para investigar más a fondo la función de regulación de micC en Salmonella Enteritidis, clonamos el gen micC en la cepa 50336 de Salmonella Enteritidis, y luego construimos el mutante 50336Δ micC mediante el sistema de recombinación λ de base roja y el mutante complementario 50336Δ micC/p micC portador del plásmido recombinante pBR322 que expresa micC. Los ensayos LD 50 mostraron que después de infectar ratones BALB/c de seis a ocho semanas de edad con cada una de las tres cepas, la mayoría de los ratones infectados por 50336 Delta micC mostraron lasitud, inapetencia o diarrea 48 horas después de la infección y murieron después de 96 horas. Los ratones infectados por cepa de tipo salvaje y 50336 delta micC pmicC mostraron los mismos síntomas 72 horas después de la infección y murieron después de 120 horas.Los LD 50 de las tres cepas indicaron que la virulencia del mutante 50336 Delta micC mejoró 2,5 veces en comparación con el tipo salvaje. Esta técnica ayuda a los investigadores a estudiar la función de los genes bacterianos y obtener la cepa de ingeniería deseada mediante la eliminación de genes objetivo.
