Er zijn verschillende primaire hepatocytenisolatieprotocollen ontwikkeld. Ze zijn ook geoptimaliseerd en aangepast op basis van verschillende behoeften (tabel 1). Isolatieprotocollen bestaan over het algemeen uit twee delen: perfusie (inclusief enzymvertering) en zuivering.
De perfusie kan worden uitgevoerd met de gehele lever in vivo 2,20,21,22,23 of met ontlede leverkwabben </sup...

De lever dient als een van de belangrijkste organen in het gewervelde lichaam vanwege de vitale rol die het speelt in tal van levensondersteunende functies zoals voedselvertering, bloedcirculatie en ontgifting. Het gebruik van primaire hepatocyten in vitro kweek van muizen wordt steeds populairder in studies naar koolhydraatmetabolisme en levercarcinoom. Daarom is het belangrijk om een handige methode te ontwikkelen voor primaire hepatocytenisolatie bij muizen met behoud van de aangeboren fysiologische functie. Vanwege zijn functie als een hub van glucosemetabolisme, staat de lever ook centraal in de glucoseproductie en -opslag1. Experimenten met primaire hepatocyten in vitro zijn een must voor de meeste glucosemetabolismestudies. Daarom hebben verschillende onderzoeksgroepen jarenlang protocollen ontwikkeld voor primaire hepatocytenisolatie van muizen.
De algemene procedure van muishepatocytenisolatie is om eerst bloed in de lever weg te spoelen met een isosmotische vloeistof zoals fosfaat-gebufferde zoutoplossing (PBS) of Hanks 'Balanced Salt Solution (HBSS) en vervolgens collagenase-bevattende oplossing te gebruiken om hepatocyten te dissociëren. Deze protocollen delen een algemene procedure, maar verschillen in reagentia en stappen op basis van verschillende behoeften. Het voorbereiden van vereiste reagentia en het uitvoeren van isolatiestappen kost echter tijd. Bij de ontwikkeling van het huidige protocol is efficiëntie voorop gesteld, waarbij alle reagentia gebruiksklaar en uit de markt leverbaar zijn, en zo min mogelijk stappen. Het algemene doel van dit protocol is om een snelle en arbeidsefficiënte methode te bieden om primaire hepatocyten van muizen te isoleren, zonder de geïsoleerde primaire hepatocytenzuiverheid en levensvatbaarheid in gevaar te brengen.

an improved time- and labor- efficient protocol for mouse primary hepatocyte isolation

Primaire hepatocyten worden op grote schaal gebruikt in lever in vitro onderzoek, vooral in glucosemetabolisme studies. Een basistechniek is aangepast op basis van verschillende behoeften, zoals tijd, arbeid, kosten en primair hepatocytengebruik, wat resulteert in verschillende primaire hepatocytenisolatieprotocollen. De vele stappen en tijdrovende reagenspreparaten in primaire hepatocytenisolatie zijn echter grote nadelen voor de efficiëntie. Na het vergelijken van verschillende protocollen voor hun voor- en nadelen, werden de voordelen van elk gecombineerd en werd een snel en efficiënt primair hepatocytenisolatieprotocol geformuleerd. Binnen slechts ~ 35 minuten zou dit protocol evenveel, zo niet meer, gezonde primaire hepatocyten kunnen opleveren als andere protocollen. Verder valideerden glucosemetabolisme-experimenten uitgevoerd met behulp van de geïsoleerde primaire hepatocyten het nut van dit protocol in in vitro levermetabolismestudies. We hebben ook uitgebreid de betekenis en het doel van elke stap in deze studie beoordeeld en geanalyseerd, zodat toekomstige onderzoekers dit protocol verder kunnen optimaliseren op basis van behoeften.

Om de efficiëntie te testen, werd het huidige primaire hepatocytenisolatieprotocol uitgevoerd op 8 weken oude vrouwelijke C57BL / 6-muizen. De hechting en zuiverheid van geïsoleerde primaire hepatocyten werden getest. An Improved Time- and Labor- Efficient Protocol for Mouse Primary Hepatocyte Isolation

Primaire hepatocyten zijn een waardevol hulpmiddel om leverrespons en metabolisme in vitro te bestuderen. Met behulp van in de handel verkrijgbare reagentia werd een verbeterd tijd- en arbeidsefficiënt protocol voor primaire hepatocytenisolatie bij muizen ontwikkeld.

Primaire hepatocyten van muizen zijn belangrijk in leverstudies. Vooral voor glucosemetabolisme en hepatische medicijntests. Qua tijd kan het isolatieproces tijdrovend zijn en de energie kostbaar.Dit protocol kan een efficiënte manier bieden om primaire hepatocyten van muizen te isoleren. Het is tijd- en arbeidsvriendelijk. Vereist zeer weinig stappen met alle regio's commercieel beschikbaar.Begin met het gebruik van de peristaltische pomp om opgewarmd dubbel gedestilleerd water op te pompen met een snelheid van 4 ml per minuut, gedurende vijf minuten. Vervang vervolgens de pompbuis van water naar opgewarmd perfusiemedium. Bewaking van de luchtbel tussen het water en het perfusiemedium.Helpt bij het volgen van de stroom van het perfusiemedium door het systeem. Desinfecteer vervolgens de buik van een verdoofde acht weken oude C57 zwarte 6J vrouwelijke muis met 70% ethanol. En knip met een schaar de buik open om de lever, poortader en inferieure vena cava bloot te leggen.Stop dan de peristaltische pomp. Na ervoor te hebben gezorgd dat de pompbuis perfusiemedium bevat en geen water. Plaats een 24 gauge katheter in de inferieure vena cava.Schakel de pomp weer in en snijd de poortader open. Terwijl het perfusiemedium wordt gepompt, drukt u elke minuut op de poortader om de vloeistof elke hoek van de lever te laten bereiken. Blijf ongeveer drie tot vijf minuten pompen of totdat de uitgespoelde vloeistof helder is.Vervang vervolgens de pompbuis van het perfusiemedium naar het collagenase-dispasemedium. En druk elke minuut op de poortader om de vloeistof elke hoek van de lever te laten bereiken. Ga door met pompen totdat alle 25 ml van het collagenase dispase medium zijn uitgeput.Isoleer vervolgens de hele lever zonder de galblaas. En plaatste het in 30 ml wasmedium in een petrischaaltje op ijs. Met behulp van een tang scheurt de lever in stukken om primaire hepatocyten in de oplossing vrij te geven.In dit stadium verandert het wasmedium in een troebele oplossing met vrijgekomen primaire hepatocyten en kleine leverstukjes. Filter deze troebele oplossing door een celzeef van 70 micron, in 20 ml 1X per oproep HBSS in een buis van 50 ml op ijs. En meng de oplossing door de buis 20 keer om te keren.Centrifugeer vervolgens de oplossing. Adem vervolgens in de weefselkweekkap het supernatant op. En was de pellet met 30 ml koud wasmedium.Pellet de cellen nogmaals met centrifugeren. Verwijder het supernatant en suspensie de pellet opnieuw in 25 ml kweekmedium. Tel de cellen en plaats ze op gewenste kweekplaten.Volgens het experimentele ontwerp. Na plating werden geïsoleerde primaire hepatocyten met een uur stevig bevestigd en na 12 uur volledig uitgebreid. In de geïsoleerde hepatocyten waren de mRNA-niveaus van hepatocytmarkers, zoals transthyretine, CD95, ASGR1 en ASGR2 significant verhoogd in vergelijking met de hele lever.Daarentegen was de aanwezigheid van immuuncellen, stellaatcellen en endotheelcellen lager. Zoals blijkt uit een sterke afname van CD45, collageen type 1 alfa 1 en endotheelcel 2 kinase 2 mRNA niveaus. Wat suggereert dat dit protocol de interferentie van andere levercellen aanzienlijk kan verminderen.Na plating daalden de ASGR1- en ASGR2-niveaus met de tijd, maar blijven ze vergelijkbaar met de hele lever tot het 12-uurstijdpunt. Speciaal voor ASGR1. Het expressieniveau van membraangebonden CD81 was consistent gedurende maximaal 48 uur.Ter vergelijking: de tolachtige receptor 4-expressie nam over het algemeen toe en werd hoger dan in vivo niveaus na 48 uur. Insulinegevoeligheidstests toonden aan dat insuline de fosforylering van beide AKT bij serine 473 significant bevorderde. En vorkkopdoos 01 bij serine 256, in de hepatocyten.Dit geeft de gevoeligheid van primaire hepatocyten voor insuline aan. Fosfoenolpyruvaat carboxykinase eiwit niveaus significant verhoogd na glucagon behandeling. Wat suggereert dat de glucoseproductieroute werd geactiveerd.Deze activering werd verder bevestigd door een verhoogde glucoseproductie. En met andere glucoseproductiestimulatoren zoals Forskolin + IBMX. Dit protocol kan zowel tijd als energie besparen voor studies met primaire hepatocyten van muizen.Volgens dit protocol kunnen gerelateerde experimenten zoals het hypothetische glucosemetabolisme en farmaceutische biomarkertests worden uitgevoerd.
