ウイルスの動的な複製および組み立てプロセスのキャプチャは、ウイルスの核酸とタンパク質の高感度かつ同時標識を可能にする堅牢なin situハイブリダイゼーション(ISH)技術の欠如によって妨げられてきました。従来のDNA蛍光in situハイブリダイゼーション(FISH)法は、多くの場合、免疫染色に適合しません。そこで、DNA、RNA、タンパク質のシングルセル同時可視化を可能にするイメージングアプローチMICDDRP(DNA、R NA、PROTEINの究極の蛍光cELベースのデテクション)を開発しました。従来のDNA FISHと比較して、MICDDRPは分岐DNA(bDNA)ISH技術を利用しており、オリゴヌクレオチドプローブの感度と検出を劇的に向上させます。MICDDRPの小さな修飾により、異なる鎖の核酸(RNAまたはDNA)と同時にウイルスタンパク質のイメージングが可能になります。これらのプロトコルを適用して、ヒト免疫不全ウイルス(HIV)-1、ヒトTリンパ球向性ウイルス(HTLV)-1、B型肝炎ウイルス(HBV)、C型肝炎ウイルス(HCV)、ジカウイルス(ZKV)、インフルエンザAウイルス(IAV)など、複数のウイルス病原体のライフサイクルを研究しています。私たちは、さまざまなウイルスのウイルス核酸とタンパク質を効率的に標識できることを実証しました。これらの研究は、複数のウイルスシステムの機構的理解を向上させることができ、さらに、幅広い細胞系にわたるDNA、RNA、およびタンパク質のマルチプレックス蛍光イメージングのアプリケーションのためのテンプレートとして役立ちます。

MICDDRP の回路図を 図 1 に示します。DNAおよびRNAの標識に続いて免疫染色を行う。分岐増幅器を使用するとシグナルが増加し、単一の核酸分子の検出が可能になります。
