Eiwitconformatie en bijbehorende hogere ordestructuur (HOS) zijn de belangrijkste determinanten van een goede biologische functie en afwijkend gedrag1. Hetzelfde geldt voor biofarmaceutica, waarvan de structuur en functionele activiteit afhankelijk zijn van verschillende aspecten van hun productie en omgeving. Biofarmaceutische veranderingen in HOS zijn gekoppeld aan bijwerkingen (ADR) toegeschreven aan ongewenste farmacologie en immunologische respons van de patiënt2,3. Het verschijnen van ADR's heeft de biofarmaceutische industrie gewaarschuwd voor de cruciale rol die eiwit-HOS speelt in de veiligheid en werkzaamheid van biotherapeutica, en ze hebben de behoefte aan nieuwe en verbeterde HOS-analyses vastgesteld4.
Hydroxyl Radical Protein Footprinting (HRPF) is een veelbelovende techniek om de verandering in eiwit HOS te volgen. HRPF omvat de onomkeerbare etikettering van de buitenkant van een eiwit met ▪OH gevolgd door massaspectrometrie (MS) analyse om het oplosmiddel toegankelijke oppervlak van het eiwit5,6,7te identificeren . HRPF is met succes gebruikt om defecten in eiwit HOS en zijn functie te detecteren8,9, karakteriseren de HOS van monoklonale antilichamen (mAb)10,11,12,13, bepalen de binding Kd van een ligand14, en nog veel meer15,16,17,18,19. Een veel voorkomende methode om de ▪OH voor HRPF te genereren is Fast Photochemical Oxidation of Proteins (FPOP), die gebruik maakt van energierijke, snelle UV-lasers om ▪OH te produceren uit fotolyse van H2O2. FPOP maakt voor het grootste deel gebruik van dure excimerlasers die gevaarlijk gas (KrF) gebruiken en die aanzienlijke waarborgen vereisen om ademhalings- en oogletsel te voorkomen20. Om inhalatiegevaren te voorkomen, hebben anderen frequentie verviervoudigde neodymium yttrium aluminium granaat (Nd:YAG) lasers21gebruikt, die het gebruik van giftig gas elimineert, maar nog steeds duur is, aanzienlijke operationele expertise vereist en uitgebreide verdwaalde lichtcontroles vereist om gebruikers te beschermen tegen oogletsel.
Hoewel er voldoende informatie kan worden verkregen met behulp van HRPF, is niet voldaan aan een brede acceptatie in biofarma. Twee barrières voor de beperkte HRPF-goedkeuring zijn: 1) het gebruik van gevaarlijke en dure lasers die aanzienlijke veiligheidsmaatregelen vereisen20; en 2) de onherproduceerbaarheid van HRPF veroorzaakt door achtergrondopruiming van ▪OH die vergelijkende studies beperken22. Om lasergebruik te verdringen, werd een high-speed, high energy plasmaflitser fotolyse-eenheid ontwikkeld om FPOP veilig op een facile manier uit te voeren. Om de onherproductieve hrpf-experimenten te verbeteren, wordt real-time radicale dosimetrie geïmplementeerd.
De praktijk van HRPF is beperkt door onherkenbaarheid toegeschreven aan achtergrondopruiming van ▪OH22. Hoewel ▪OH uitstekende sondes van eiwittopografie zijn, reageren ze ook met veel bestanddelen die in preparaten worden aangetroffen, waardoor het noodzakelijk is om de effectieve concentratie van radicalen te meten die beschikbaar is om een doeleiwit te oxideren. Variaties in buffervoorbereiding, waterstofperoxideconcentratie, ligandeigenschappen of fotolyse kunnen leiden tot oxidatieverschillen tussen controle- en experimentele groepen die ambiguïteit creëren in HOS-differentiaalstudies. De toevoeging van real-time radicale dosimetrie maakt het mogelijk om het effect ▪ OH-belasting aan tepassen en verhoogt daardoor het vertrouwen en de reproduceerbaarheid tijdens een HRPF-experiment. Het gebruik van radicale dosimetrie in FPOP is elders beschreven23,24,25, en wordt verder in detail besproken in een recente publicatie26. Hier beschrijven we het gebruik van een nieuw flitsfotolysesysteem en real-time dosimetrie om equine apo-myoglobine (aMb) te labelen, waarbij we de niveaus van peptideoxidatie in een FPOP-experiment vergelijken met die van verkregen bij het gebruik van een excimerlaser.

laser-free hydroxyl radical protein footprinting to perform higher order structural analysis of proteins

Hydroxyl Radical Protein Footprinting (HRPF) is een opkomende en veelbelovende hogere orde structurele analysetechniek die informatie biedt over veranderingen in eiwitstructuur, eiwit-eiwit interacties of eiwit-ligand interacties. HRPF maakt gebruik van hydroxylradicalen(▪OH) om het oplosmiddeltoegankelijke oppervlak van een eiwit onomkeerbaar te labelen. De inherente complexiteit, kosten en gevaarlijke aard van het uitvoeren van HRPF hebben een aanzienlijk beperkte brede acceptatie in biopharma. Deze factoren omvatten: 1) het gebruik van gecompliceerde, gevaarlijke en dure lasers die aanzienlijke veiligheidsmaatregelen vereisen; en 2) de onherproduceerbaarheid van HRPF veroorzaakt door achtergrondopruiming van ▪OH die vergelijkende studies beperken. Deze publicatie biedt een protocol voor de werking van een laservrij HRPF-systeem. Dit laservrije HRPF-systeem maakt gebruik van een hoge energie, hoge druk plasma lichtbron flash oxidatie technologie met in-line radicale dosimetrie. De plasmalichtbron is veiliger, gemakkelijker te gebruiken en efficiënter in het genereren van hydroxylradicalen dan lasergebaseerde HRPF-systemen, en de in-line radicale dosimeter verhoogt de reproduceerbaarheid van studies. Gecombineerd pakt het laservrije HRPF-systeem de genoemde tekortkomingen en beperkingen van lasergebaseerde technieken aan en overtreft het deze.

De hogedrukplasmabron in combinatie met real-time dosimetrie maakt een betere controle van ▪OH-opbrengst mogelijk om veranderingen in de eiwitstructuur van hogere orde nauwkeuriger te observeren. De toevoeging van adenine zorgt voor een effectieve real-time radicale dosimeter. Bij oxidatie verliest adenine uv-absorptie bij 265 nm (figuur 2A). Laservrije hydroxylradicale eiwitvoetafdruk maakt het identificeren van eiwitinteractieplaatsen en gebieden van bevestigingsverandering veel gemakkelijker, waardoor onderzoek in eiwitaggregatie-, structuur- en stabiliteitsstudies wordt versneld. Met inline radicale dosimetrie kunnen gebruikers de effectieve hydroxylradicale belasting in realtime aanpassen, en met realtime radicale dosimetrie kunt u experimentele tijd en kostbaar monster besparen terwijl de reproduceerbaarheid van etiketten wordt verbeterd. Om te beginnen, splijten een 250 micrometer binnendiameter silica capillair tot 27 inch met een silica kloven steen, het controleren van de uiteinden van het capillair voor een schone en rechte snede.Maak twee vensters van ongeveer 15 millimeter lengte. Van de onderkant van het capillair, brand de polyimide coating 90 millimeter tot aan het fotolysevenster en 225 millimeter voor het dosimetervenster weg. Plaats het onderste uiteinde van het capillair net voorbij het conische uiteinde van een noot en ferrule.Voeg het capillair toe aan poort vijf en draai net voorbij de vinger vast met een moersleutel. Verwijder de fotolyseceldop van de fotolysemodule door deze er recht uit te trekken en verwijder vervolgens het magnetisch gemonteerde metalen masker, dat het capillair op zijn plaats houdt. Open de dosimetercel door op het lipje aan de linkerkant te drukken en de dosimetercel naar rechts te zwaaien.Verwijder de magnetisch gemonteerde clips die het capillair op hun plaats houden. Plaats vervolgens het capillair in het gegroefde kanaal aan de basis van de fotolysecel en centreer het capillaire venster met het fotolysecelvenster. Houd het capillair op zijn plaats en voeg het magneetmasker toe.Plaats de fotolysedop weer op zijn plaats. Plaats het capillair in het gegroefde kanaal aan de basis van de dosimetercel en centreer het tweede capillaire venster op het kleine diafragma in het midden van de dosimetercel. Terwijl u het capillair met één hand op zijn plaats houdt, plaatst u de twee magnetische clips in positie om het capillair op zijn plaats te houden.Sluit de dosimetercel totdat deze dichtklikt. Plaats ten slotte het capillair door de gekartelde knop bovenop de capillaire lift van de productcollector en verleng het capillair tot net boven de onderkant van de flacon. Snijd de gewenste lengte teflonslang met een frees en controleer de uiteinden op een schone rechte snede.Steek het ene uiteinde van de nieuwe injectielus door een van de moeren en plaats een nieuwe ferrule op het uiteinde van de buis. Houd de moer en de ferrule op hun plaats terwijl u de buis in poort zes van de injectieklep steekt totdat deze uitvalt. Draai de moer vast met een moersleutel en bevestig vervolgens of de moer en de ferrule goed op hun plaats zitten.Zodra beide uiteinden een moer en een vaste ferrule hebben, schroeft u het ene uiteinde losjes op poort drie en het andere uiteinde op poort zes. Eenmaal in positie, draai beide zijden vast om de vinger strak te houden, dan een kwartslag voorbij vinger strak met een moersleutel. Schakel het laservrije HRPF-systeem in door de fluidics-module te starten, gevolgd door de fotolysemodule, dosimetermodule, productcollector en de systeemcomputer.Start de systeemsoftware. Dompel de slang volledig onder in 10 milliliter buffer vanuit de kleppositie op de spuitpomp. Leid de slang vanuit de afvalpositie en de slang van poort twee op de spuitpoort naar een lege container om afval op te halen.Plaats 1,5 milliliter microcentrifugebuizen op de met HN6 gemarkeerde positie op de carrousel van de productverzamelaar. Spoel de injectielus vijf keer uit door 25 microliters HPLC-water te injecteren met de klep in de belastingsstand. Draai de injectieklep handmatig om de rest van het systeem door te spoelen.Selecteer Proces op de besturingssoftware om te beginnen met stromend water totdat er een druppel ontstaat. Maak een oplossing met twee millimolar adenine en 10 micromolar eiwit. Maak vervolgens een quench oplossing met 0,3 milligram per milliliter catalase en 35 millimolar methianineamide.Aliquot 25 microliter blusoplossing in een 200 microliter microbuis. Verdun waterstofperoxide tot 200 millimolar, zodat het op ijs blijft. Start de flitsspanning op nul volt op de besturingssoftware.Deze nul volt regeling zal elke achtergrond oxidatie op het eiwit van belang bepalen. Selecteer Gegevens automatisch starten, gevolgd door Verwerken en vervolgens Gereed. Draai ten slotte de injectieklep terug naar de belastingspositie.Plaats de blusoplossing in positie één op de carrousel van de productverzamelaar en verander vervolgens de productflacon in één op de systeembesturingssoftware. Meng vlak voor injectie 12,5 microliter van het adenine- en eiwitmengsel met 12,5 microliter waterstofperoxide. Pipetteer het mengsel op en neer om te mengen en draai het vervolgens snel naar beneden.Injecteer 25 microliters van deze oplossing met behulp van de injectiepoort binnen 10 seconden na het mengen. Zet de injectieklep in de injectiestand en wacht terwijl het monster wordt verwerkt. Zet de flitsspanning op 750 volt op de besturingssoftware en herhaal de etiketteringsstappen.Noteer vervolgens de absorptie van elk monster. Klik eerst op Selecteren en selecteer vervolgens handmatig het begin en einde van de piekabsorpance. Schrijf in de beschikbare ruimte een beschrijving van het voorbeeld.Herhaal dit voor alle verkregen gegevens. Kopieer en plak de gegevens in een spreadsheet om de gemiddelde verandering in adenineabsorpiteit voor elke spanning te berekenen. Nadat alle monsters zijn verzameld, spoelt u de spuitpoort en de monsterlus door de injectieklep in de belastingsstand te zetten en injecteert u vijf keer 25 microliters HPLC-water.Draai de injectieklep in de injectiepositie om de rest van het systeem met HPLC-water te spoelen. Eenmaal doorgespoeld, stopt u de stroom, verlaat u de systeembesturingssoftware en schakelt u de modules uit, te beginnen met de productcollector, dosimetermodule, fotolysemodule en ten slotte de fluidics-module. Bij oxidatie vermindert adenine dat wordt gebruikt voor dosimetrie de UV-absorptie met 265 nanometer.Buffer 2 bevat een radicale aaseter, die de verandering in adenineabsorptantie vermindert in vergelijking met Buffer 1. Apomyoglobin werd gewijzigd in aanwezigheid van waterstofperoxide en adenine bij vier toenemende plasmalampspanningen. Zes peptiden werden gedetecteerd, met een lineaire toename van oxidatie met betrekking tot de verandering van adenineabsorptantie bij 265 nanometer.Deze zes geoxideerde peptiden zijn gelabeld op een kristalstructuur van myoglobine. De mate van oxidatie gedetecteerd door de hogedrukplasmalamp is veel hoger dan de lasergebaseerde methode. Deze toename komt voort uit het brede uv-emissiespectrum van de hogedrukplasmalamp.Door een breed UV-spectrum te produceren, overlapt de hogedrukplasmalamp het absorptiedomein van waterstofperoxide beter, wat resulteert in een efficiëntere productie van hydroxylradicalen door de fotolyse van waterstofperoxide, in vergelijking met de Kr F Excimer Laser en Nd YAG Laser. De hogedrukplasmalamp verhoogt de productie van hydroxylradicalen aanzienlijk verder dan wat doorgaans wordt waargenomen met behulp van een Kr F Excimer Laser. Het gebruik van 100 millimolar peroxide kan leiden tot hoge niveaus van oxidatie op de achtergrond.Als het eiwitsysteem gevoelig is voor door peroxide veroorzaakte oxidatie, verlaag dan de concentratie tot slechts 10 millimol. HRPF's onomkeerbare label maakt downstream monsterbehandeling mogelijk, inclusief lange proteaseverteringen, u kunt tandemmassatag toevoegen voor multiplexing en 2D-chromatografie uitvoeren, allemaal ter verbetering van peptidedetectie en conccoördineerde structurele informatie.
