生体試料における脂質保存分析の従来のアプローチには、生化学的アッセイや脂溶性染料による染色プロトコルが含まれます。質量分析(MS)を含む生化学的定量アッセイは、その分子の解決性において比類のないものですが、非常に大きなサンプル量を必要とし、サンプル調製には通常数時間かかり、生体システムのリアルタイムイメージングへの応用を制限します5。これらのアッセイのもう一つの大きな制限は、空間情報の欠如である。一方、オイルレッドOやスーダンブラックなどの親油性色素は、脂質貯蔵小器官の組織および細胞分布を提供し、MS技術と比較して、これらの染色法も低コストで、かつ実行しやすい。しかし、これらの染色プロトコルは固定を必要とし、脂質滴の疎水性に影響を与え、その構造に人工的な変化を生じ、実験6の間に矛盾を生じる。生化学的および染色技術に関連する技術的な困難は、脂質分子を画像化するためのラベルフリーの方法の探索と、脂質イメージングにおけるコヒーレントラマン散乱(CRS)顕微鏡の使用の急速な増加につながっている。
ラマン効果は、ラマンとクリシュナンによって最初に認識され、光子と相互作用すると、分子が波長の変化(レイリー散乱と呼ばれる)または変化した波長(ラマン散乱と呼ばれる)でめったに散乱光を生成することができ、この波長の変化は分子内の官能化学基の特徴であると報告した。分子内の化学結合がポンプ光子と呼ばれる入射光子によってより高い振動エネルギーレベルに励起されると、ストークス光子と呼ばれる散乱光子のエネルギーが低くなる。さもなければ、化学結合はもともとより高いレベルにあるならばより低い振動エネルギーレベルに達し、散乱された光子は反ストークス光子になるエネルギーを得る。インシデントと散乱フォトンの周波数差は、ラマンシフトと呼ばれます。分子内の各化学結合は、特徴的かつ定量的なラマンシフトを有する。例えば、CH2結合は、2,845cm-1のラマンシフトを有し、これは脂肪酸鎖8に豊富である。この自発的なラマン信号は一般的に非常に弱く、従来の自発的なラマン顕微鏡ではイメージング速度が大幅に制限されています。また、自発ラマン顕微鏡のイメージング速度と感度を高めるさまざまなアプローチが長年にわたって開発されてきました。コヒーレント・アンチ・ストークス・ラマン散乱(CARS)顕微鏡検査や刺激ラマン散乱(SRS)顕微鏡など、コヒーレントラマン散乱顕微鏡が最新の進歩である。CARSとSRSは、わずかに異なる動作原理を持っていますが、どちらもライブイメージング機能を備え、脂質貯蔵ダイナミクスに関する空間的および時間的な情報を生成することができ、小さなサンプルサイズしか必要としなくて済むラベルフリーの技術です。CARS顕微鏡は、様々な非線形プロセスに由来する非共振性バックグラウンドに苦しんでおり、CARS信号も分子濃度と非線形関係を有し、定量プロセス9を一緒に複雑にする。CARS顕微鏡とは異なり、SRS顕微鏡は非共振性のバックグラウンド信号を生成せず、対象分子の濃度に直線的に依存します。従って、現在SRS顕微鏡は、より広く脂質イメージングに使用されている。
SRS顕微鏡では、弱い自発的なラマン信号は、化学結合振動周波数と一致する周波数差を持つ2つの同期レーザービームによって励起されたときに増幅することができる。分子は、一貫した励起のために励起状態への遷移を増強する。その結果、ストークスフォトン生成の速度が上昇する。その結果、送信された「ストークス」ビームの強度が増加し(刺激されたラマンゲイン、SRG)、送信された「ポンプ」ビームの強度が減少する(刺激されたラマン損失、SRL)。SRGまたはSRL信号の検出は、特異的化学結合10を有する分子の刺激ラマン散乱(SRS)顕微鏡イメージングの基礎となる。2つのレーザービーム間の周波数差が目的の分子内の化学結合の振動周波数と一致しない場合、SRG信号もSRL信号も生成されません。SRS顕微鏡のイメージング速度は、ピクセルあたり約2μsecまたはフレームあたり1秒で、自発的なラマン顕微鏡11よりもはるかに高速です。SRS顕微鏡の典型的な横解像度は、回折限であり、約300nmである。さらに、SRS顕微鏡の2光子光学プロセスにより、相対厚組織サンプルの体積3Dイメージングが可能となり、イメージング深度は300〜500μmに達する可能性があります。全体として、SRS顕微鏡は、特定の生体分子、特に脂質を検出するための効率的でラベルフリーなイメージング技術を提示します。
脂質滴は、トリアシルグリセロール(TAG)およびコレステロールエステル(CE)を含む中性脂質の主な細胞貯蔵部位である単膜小器官である。これらの脂質分子の脂肪酸鎖中のCH2結合は、励起された2,845cm-1で強いSRSシグナルを生成し、インタクトな細胞、組織切片、さらには生物全体12、13、14、15の保存脂質レベルの検出および定量を可能にする。特に、C.エレガンスは、その透明性のために脂質イメージング研究に有用である。哺乳類と同様に、C.エレガンスも脂質滴に脂質を保存し、脂質分子の合成および分解経路は高度に保存されている16である。このプロトコルでは、SRS顕微鏡の原理を提供し、その基本的な設定を行い、C.エレガンスにおける脂質イメージングにおける使用法について説明します。

刺激されたラマン散乱顕微鏡は、脂質の迅速かつ定量的な検出を可能にし、生きている動物の脂質をラベルフリーで追跡することを可能にする化学画像技術です。従来の脂質検出技術に対するSRS顕微鏡の最も重要な利点は、ラベルフリーであり、したがって、他の蛍光および染色の問題ほどフォト漂白に持続可能ではないということです。SRS顕微鏡の使用は、モデル生物、特にカエノルハブディティスエレガンスに利用可能な遺伝的および生化学的ツールと組み合わせて使用すると、脂質生物学と代謝の正常な調節因子を研究するための強力なフレームワークを提供します。レーザービームをSRS顕微鏡に送り込むには、まず、ピコ秒光源出口から顕微鏡のスキャナー上の赤外線レーザー入力ポートにビームを持ち上げ、レーザー制御ソフトウェアを開いてレーザーを開始するようにペリスコープを設定します。ポンプレーザーパワーを50ミリワットに下げ、ポンプ信号を750ナノメートルに設定します。ビームエキスパンダーを使用して、顕微鏡の目的の背面開口に合わせてビーム直径を調整します。M1とM2の2つのリレーミラーを使用して、レーザービームをペリスコープに導き、ペリスコープのうなずきをチューニング範囲の中心に設定します。レーザービームが鏡の中心に当たって顕微鏡にビームを打ち上げ、顕微鏡の目的砲塔の空のポートを使用してコースアライメントを行うよう、各ミラーの初期位置と角度を選択します。パワーメータープローブを目的ポートに置き、透過光のパワーを測定し、顕微鏡のスキャナーを最高ズーム50倍に設定します。焦点を合わせレーザビームスポットで伝送光のパワーを測定し、ミラーM1とM2のノブを反復的に最適化して、最高の透過レーザーパワーを達成します。アライメントツールを使用して、透過光の微調整を行い、光が目的の中心を通過していることを確認し、空の目標シートに蛍光ターゲットアライメントキャップを配置します。最初のステアリングミラーM1のノブを調整してレーザービームスポットを中央に配置し、レーザービームスポットを監視するための空の目的シートにもう一方の端にアライメントキャップを持つ延長管を導入します。次に、2つ目のステアリングミラーM2のノブを調整して、レーザービームスポットを延長管のもう一方の端に再び中央に配置します。SRS検出モジュールを設定するには、コンデンサの後にビームスプリッターキューブを配置し、送信されたレーザーをフォトダイオードモジュールに向けます。エレクトロニクス接続を設定するには、ピコ秒のチューン可能なレーザーの中に20メガヘルツの内蔵電気光学変調器を使用して、ストークスビームの強度を調節し、フォトダイオードの出力信号をロックインアンプに供給して、刺激されたラマン損失を復調させます。次に、ロックインアンプ出力をマイクロスコープのアナログボックスに送り込み、電気アナログ信号をデジタル信号に変換します。イメージング条件を最適化するには、5マイクロリットルのオレイン酸をミニ顕微鏡スライドチャンバーに加え、カバースリップでサンプルを覆います。サンプルを顕微鏡のステージに置き、パッドの端を使用して液滴を見つけて焦点を当てます。コーラー照明用のコンデンサを調整し、赤外線センサーカードと赤外線ビューアを使用して、ポンプビームパスがまだ正しいかどうかを確認します。遅延ステージがフェムト秒ゼロに設定されていることを確認し、ポンプビーム波長を795.8ナノメートルに設定します。ポンプビームのパワーを50ミリワットに設定し、ストークビームを100ミリワットに設定します。両方のビームのシャッターとレーザーのメインシャッターを開きます。サンプルをスキャンし、コンピュータ画面上の画像を確認します。表示モードを高低に変更し、約 50% の彩度を表示するように範囲を調整します。飽和が画像の中央にあるかどうかを確認し、ピコ秒のチューン可能なレーザーシステム内のステアリングミラーを慎重に調整して、画像強度を最大化し、必要に応じてピーク強度を中央に配置します。各イメージングセッションの前に、100マイクロリットルの暖かい2%アガロースを、2層のラボテープを備えたサポートスライドの間に配置されたきれいなガラススライドに加え、最初のスライドの上に2番目のスライドを素早く配置します。やさしく押して、薄いアガロースパッドを作成し、アガロースを冷却します。イメージング用のワームを取り付けるには、10~20個のワームにつき4~5マイクロリットルの麻酔薬をカバースリップに置き、解剖顕微鏡を使用して画像化するワームを選び、収集された麻酔の液滴にワームを置きます。すべてのワームが収集されたら、アガロースパッドでガラススライドでワームを覆います。イメージングの場合は、ウォームサンプルをカバースリップで対物レンズに向けて取り付け、明視野光源を接眼レンズに向けて、ワームを見つけます。ワームに焦点を当て、それに応じてコンデンサーの位置を調整します。ポンプレーザーパワーを200ミリワットに、IRパワーを400ミリワットに設定して、レーザーパワーを調整します。ワームSRS信号の復調用ロックインアンプを設定します。最初のワームのスキャンを高速スキャン速度で開始し、対象領域を見つけるために細かい焦点を調整します。信号が最適化されたら、SRSイメージを取得するために、より低速なスキャンレートと高いピクセル解像度に切り替え、高解像度の画像を可能にする形式で保存します。すべてのサンプルがイメージ化されたら、レーザー光源をスタンバイ状態にして、関連する機器をすべてオフにします。画像を解析するには、ImageJ でファイルを開き、解析を選択して、分析するプロパティを選択して測定を設定します。ポリゴン選択ツールを使用して、定量化するワーム腸の領域を概説し、[解析]をクリックしてもう一度測定します。測定結果ウィンドウに測定値が表示されます。すべてのワームの概要が作成されたら、特定の遺伝子型の各ワームの測定値をコピーしてスプレッドシートに貼り付けます。背景の測定値については、SRS信号を持たないワームの近傍の領域を選択し、個々のワームの測定値から背景平均グレー値を差し引き、所定の遺伝子型または試験群内のすべてのワームから差し引いた背景の平均値と標準偏差を計算します。これらの値は、制御グループの平均に正規化することができます。この代表的な分析では、成人期にインスリン受容体を欠いた年齢同期性野生型ワームおよびdaf-2変異体における腸内脂質レベルを定量した。予想通り、野生型ワームでは脂質レベルの低下が観察され、1日目から成人期の9日目まで続きます。しかし、daf-2変異体の脂質レベルは、5日後まで増加し、その時点で野生のタイプよりも約2.5〜3倍高いままである。Daf-16の不活性化は、daf-2変異体の脂質レベルの増加を抑制する。daf-16aaまたはdaf-16bのアイソフォームの発現は、daf-2において、daf-16二重変異体は脂質レベルを回復しない。しかし、daf-16d/f/hアイソフォームの発現は、daf-2、daf-16二重変異体における脂質レベルをdaf-2単一変異体で観察されたレベルに復元するのに十分である。これらの結果は、daf-16d/f/hアイソフォームがdaf-2変異型ワームにおける脂質代謝を特異的に調節することを示す。イメージングセッション全体を通して、ポンプとストークスのビームに同じレーザーパワーを使用し、6つおよび4つの一貫した定量化ごとに制御グループを含めることが重要です。脂質保存の定量に加えて、SRS顕微鏡検査は重水素などの生体直交タグと結合し、脂質の組み込み、合成および分解のダイナミクスを可視化するために実施することができる。SRS顕微鏡は多重化を可能にし、異なる細胞下のコンパートメント内の複数の生体分子をイメージするために実施することができる。いわゆるハイパースペクトルSRS顕微鏡は、代謝ダイナミクスを探求する上で大きな未来を持っています。
