Under gonadogenesis gjennomgår den embryonale gonad, og spesielt stamcellenisjen i den mannlige gonad15, raske morfologiske endringer. Drosophila melanogaster testis har fungert som et paradigme for vår forståelse av mange dynamiske cellulære prosesser. Studier av denne modellen har belyst stamcelledivisjonsregulering 1,2,3, bakteriecelleutvikling 4,5, piRNA-biologi 6,7,8 og nisje-stamcellesignaleringshendelser 9,10,11,12,13. Denne modellen er fordelaktig fordi den er genetisk gjennomførbar14,15 og er en av de få der vi kan leve-bilde stamceller i deres naturlige miljø 3,16,17,18. Imidlertid har levende avbildning av denne modellen vært begrenset til voksenvev og tidlige embryonale stadier, og etterlater et gap i vår kunnskap om gonadaldynamikk i sen embryo, det nøyaktige stadiet når nisjen først dannes og begynner å fungere.
Den senfase embryonale gonad er en sfære, bestående av somatiske nisjeceller i fremre, og bakterieceller som er viklet inn av somatiske gonadale celler i flere bakre regioner19. Dette organet kan avbildes levende in vivo frem til tidlig embryonal stage 17 17,20,21. Videre avbildning er forhindret på grunn av initiering av store muskelsammentrekninger. Disse sammentrekningene er så alvorlige at de skyver gonad ut av bilderammen, og slik bevegelse kan ikke korrigeres med bildeprogramvare. Laboratoriet vårt er interessert i å avduke mekanismene for nisjedannelse, som oppstår i denne unnvikende perioden for levende avbildning. Derfor genererte vi en ex vivo-tilnærming til levende bilde gonad som starter på embryonal trinn 16, og letter studiet av celledynamikken i denne avgjørende perioden med gonadutvikling. Tidligere arbeid fra laboratoriet vårt viser at denne ex vivo-avbildningen trofast recapitulates in vivo gonad utvikling17. Denne teknikken er den første og eneste av sitt slag for Drosophila embryonal gonad.
Her presenterer vi disseksjonsprotokollen som kreves for ex vivo live-imaging av gonad under sene embryonale stadier. Denne protokollen kan kombineres med farmakologiske behandlinger, eller transgen manipulering av spesifikke celleforinger i gonad. Ved hjelp av denne teknikken har vi vellykket avbildet trinnene i stamcellenisjedannelse17. Denne bildemetoden er dermed medvirkende for stamcellebiologien, da den vil muliggjøre visualisering av de første stadiene av nisjedannelse i sanntid i sitt naturlige miljø15,17. Selv om denne metoden er gunstig for stamcellebiologi, er den i tillegg anvendelig for visualisering av dynamiske prosesser som forekommer i gonad under dette utviklingstidspunktet, inkludert cellulære omorganiseringer22, celleadhesjon 2,12,23 og cellemigrasjon23. Etter disseksjon kan gonader avbildes ved hjelp av et konfokalt mikroskop for å visualisere dynamiske cellulære prosesser. Disseksjonsprosedyren krever presis timing og fingerferdighet, men vi gir innsikt i hvordan vi kan forhindre vanlige feil og hvordan vi kan overvinne disse utfordringene. Så vidt vi vet er dette den første disseksjonsprotokollen for Drosophila embryonal gonad, og vil tillate levende avbildning i et ellers utilgjengelig tidsvindu.
