神経前駆細胞(NPC、神経幹および前駆細胞とも呼ばれる)は、神経変性治療戦略1の有望な候補となり得る。未分化NPCは自己再生能力、多効力、増殖能力2,3を有する。以前の研究では、細胞外マトリックスと分子メディエーターがNPCの分化を調節することが報告されています。表皮成長因子(EGF)及び塩基性線維芽細胞増殖因子(bFGF)はNPC増殖を促進し、従って未分化状態を維持する4。
これまでの研究では、電気刺激は、除算5、移動6、7、8、分化1、9、10、および細胞死11のような細胞生理学的活動を調節することができるという報告がある。電気分野(EF)は、中枢神経系の発達12,13,14の発達と再生において重要な役割を果たす。2009年から2019年にかけて、この研究室は、マイクロ流体システム1、6、7、8、15、16、17におけるEFの適用に対する細胞応答を調査した。マルチチャネル、光学的に透明な電気戦術(MOE)チップは、共焦点顕微鏡用の免疫蛍光染色に適するように設計された。このチップは、高い光学透過性と耐久性を有し、1回の研究で3つの独立した刺激実験といくつかの免疫染色条件の同時実施を可能にした。マイクロ流体システムは、全体的な実験のセットアップを簡素化し、さらに試薬とサンプルの消費を簡素化する上で有益です。本論文では、長期細胞分化研究に用いたマイクロ流体細胞培養システムの開発について述べた。

electric-field-induced neural precursor cell differentiation in microfluidic devices

生理的電界(EF)は、細胞の移動、分化、分裂、死に重要な役割を果たしています。本論文では、顕微鏡を用いた長期細胞分化研究に用いたマイクロ流体細胞培養システムについて述べている。マイクロ流体システムは、光学的に透明な電気戦術チップ、透明なインジウムスズ酸化物(ITO)ヒーター、培養メディア充填ポンプ、電力供給、高周波パワーアンプ、EFマルチプレクサ、プログラマブルX-Y-Z電動ステージ、およびデジタルカメラを搭載した逆位相対向顕微鏡の主要コンポーネントで構成されています。マイクロ流体システムは、全体的な実験のセットアップを簡素化し、さらに試薬とサンプルの消費を簡素化する上で有益です。この研究は、直流(DC)パルス刺激によって誘導される神経幹および前駆細胞(NPC)の分化を伴う。幹細胞維持培地では、マウスNPC(mNPC)は、DCパルス刺激後にニューロン、アストロサイト、オリゴデンドロサイトに分化した。この結果は、単純なDCパルス治療がmNPCの運命を制御し、神経系障害の治療戦略を開発するために使用できることを示唆している。このシステムは、複数のチャネルでの細胞培養、長期EF刺激、細胞形態観察、および自動タイムラプス画像取得に使用できます。このマイクロ流体システムは、必要な実験時間を短縮するだけでなく、微小環境の制御精度を向上させます。

本研究は、マイクロ流体系における直流パルス刺激によってのみ誘導される神経前駆細胞の分化に関するプロトコルを提示する。この手法は、実験の段取り時間、サンプル量、試薬量を削減するための有益なアプローチを提供します。さらに、MOEチップは共焦点顕微鏡観察に適しています。単純な直流パルス治療は、マウス神経前駆細胞の運命を制御することができ、神経系障害の治療戦略を開発するために使用することができます。まず、個々の PMMA 層と、適切なソフトウェアを使用して両面テープのパターンを描画します。炭酸ガスレーザースクライバの電源を入れ、パソコンに接続します。ソフトウェアを使用して、設計されたパターンファイルを開きます。レーザースクリバーのプラットフォーム上にPMMAシートまたは両面テープを置き、オートフォーカスツールを使用してPMMAシートまたは両面テープの表面にレーザーを焦点を合わせます。レーザースクリバをプリンタとして選択し、レーザースクリバを使用してPMMAシートまたは両面テープで直接アブレーションを開始します。PMMAシートから保護フィルムを取り出し、窒素ガスを使用して表面を洗浄します。PMMAシートの複数の層を結合するには、1ミリメートルPMMAシートの3枚を積み重ね、110°Cで30分間熱ボンダーで平方センチメートル当たり5キログラムの圧力で結合し、流れまたは電気刺激チャネルアセンブリを形成します。高速作用シアノアクリル接着剤とMOEチップアセンブリの層1の個々の開口部に12個のアダプタを付着させます。チップを組み立てる前に、1ミリメートルPMMA基板、両面テープ、3ミリ光学グレードPMMAを30分間紫外線照射で消毒します。3ミリメートル光学グレードPMMAに1ミリメートルのPMMA基板を両面テープで貼り付け、PMMAアセンブリを完成させます。洗浄したカバーグラスを、両面テープでPMMAアセンブリに貼り付けます。寒天ブリッジアダプターの開口部を白い指密プラグでシールします。フラットボトムコネクタを中型の入口アダプタとコンセントアダプタを介してMOEチップアセンブリに接続し、コーンLuerアダプタを3ウェイストップコックに接続します。0.01%PLL溶液の2ミリリットルを培地入口の3ウェイストップコックに接続する3ミリリットルのシリンジを使用して追加し、空の3ミリリットルのシリンジを媒体出口の3ウェイストップコックに接続します。PLL溶液で細胞培養領域を充填し、ゆっくりとコーティング溶液を前後にポンピングします。2つの三方ストップコックを閉じて、培養地域内の溶液を密封します。5%の二酸化炭素雰囲気で満たされたインキュベーターで一晩摂氏37度でMOEチップをインキュベートします。2つの三方向ストップコックを開き、2つの注射器を使用してチャネル内でコーティング溶液を前後に手動でポンピングすることで、チャネル内の気泡を洗い流します。完全な培地を3ミリリットルのシリンジに3ミリリットル引き込み、培地入口の3ウェイストップコックに接続し、それを加えて細胞培養領域のコーティング溶液を交換します。白い指密プラグをLuerアダプタに交換し、3%ホットアガロースを注入してアダプタを充填します。LuerロックシリンジをLuerアダプタに接続し、黒いゴムボンを通して3%のホットアガロースを注入してLuerロックシリンジを充填します。アガロースが冷めて固めるのに10~20分かかります。セルシードのMOEチップを、摂氏37度に維持されたプログラム可能なXYZ電動ステージに取り付けられている透明なITOヒーターに取り付けます。MNPCを手動で媒体出口を介してMOEチップに送り込み、ITOヒーターのセルシードMOEチップを4時間インキュベートします。電線を使用して、EF マルチプレクサをチップの電極を介して MOE チップに接続します。EF マルチプレクサとファンクション ジェネレータをアンプに接続して、直角波 DC パルスを出力します。MOEチップのフラットボトムコネクタとアダプタをすべて取り外し、アダプタにPBSを充填し、アダプタの開口部をパラフィルムで密封します。免疫染色後、共焦点蛍光顕微鏡を用いて細胞を観察する。DCパルス刺激により、幹細胞維持培地中のニューロン、アストロサイト、オリゴデンドロサイトに同時に分化するmNPCが生まれた。コントロール群および刺激群におけるニューロン、アストロサイト、およびオリゴデンドロサイトの割合をここに示す。DCパルス処理後、mNpcはDIV 7で有意に高い数のニューロン、DIV 3でアストロサイト、DIV 7とDIV 14のオリゴデンドロサイトが対照群よりも刺激群において比較的高いレベルで存在した。30〜40細胞によって形成される神経球は、mNPC分化を開始するために好ましい。mNPCの過剰増殖は分化プロセス中の細胞生存を損なう。
