Xenopus laevis ægekstrakter er opstået som et kraftfuldt modelsystem til billeddannelsesbaserede undersøgelser af forskellige subcellulære strukturer 10,14,15,16,17,18,21,31,32,33,34,35,36 og cytoplasmatisk organisation i det hele taget</s...

Cytoplasmaet udgør hovedvolumenet af en celle og har en særskilt organisation. Ingredienserne i det eukaryote cytoplasma kan selv samles i en bred vifte af rumlige strukturer, såsom mikrotubulus asters og Golgi-apparatet, som igen er dynamisk arrangeret og vendt afhængigt af cellens identitet og fysiologiske tilstand. At forstå den rumlige organisation af cytoplasmaet og dets forbindelse til cellulære funktioner er således vigtigt for at forstå, hvordan cellen fungerer. Xenopus laevis ægekstrakter er traditionelt blevet brugt til bulk biokemiske assays 1,2,3,4,5,6,7,8, men nyere arbejde etablerer dem som et kraftfuldt levende billeddannelsessystem til mekanistiske undersøgelser af cytoplasmatiske strukturer og deres cellulære funktioner 9,10,11, 12,13,14,15,16,17,18. Disse ufortyndede ekstrakter bevarer mange strukturer og funktioner i cytoplasmaet, samtidig med at de tillader direkte manipulationer af cytoplasmatisk indhold, der ikke kan opnås i konventionelle cellebaserede modeller19,20. Dette gør dem ideelle til at karakterisere cytoplasmatiske fænomener og dissekere deres mekanistiske underlag.
Eksisterende metoder til billeddannelsesekstrakter kræver kemiske overflademodifikationer eller fremstilling af mikrofluidiske enheder. En coverslip-baseret metode kræver polyethylenglycol (PEG) passivering af glasdæksler21. En mikroemulsionsbaseret metode kræver dampaflejring af trichlor(1H,1H,2H,2H-perfluoroctyl)silan på glasoverflader22,23. Mikrofluidbaserede systemer muliggør præcis kontrol af volumen, geometri og sammensætning af ekstraktdråber, men kræver specialiserede mikrofabrikationsfaciliteter11,12,24.
Her introducerer vi en alternativ metode til billeddannelse af ægekstrakter, der er let at implementere og bruger let tilgængelige, billige materialer. Dette omfatter fremstilling af et billeddannelseskammer med et dias og en dæksel belagt med fluoreret ethylenpropylen (FEP) tape. Kammeret kan bruges til billeddannelsesekstrakter med en række mikroskopisystemer, herunder stereoskoper og opretstående og inverterede mikroskoper. Denne metode kræver ingen kemisk behandling af overflader, samtidig med at der opnås lignende optisk klarhed opnået med eksisterende glasbaserede metoder, der er diskuteret ovenfor. Det er designet til at afbilde et lag af ekstrakter med en ensartet tykkelse over et 2D-felt og kan let udvides til at afbilde et 3D-volumen ekstrakter. Det er velegnet til time-lapse-billeddannelse af kollektiv cytoplasmisk adfærd over et stort synsfelt.
Vi har brugt interfase-arresterede ægekstrakter til at demonstrere vores billeddannelsesmetode. Ekstraktpræparatet følger protokollen for Deming og Kornbluth19. Kort fortalt knuses æg, der naturligt arresteres i metafase af meiose II, af et lavt hastighedsspin. Dette spin frigiver cytoplasmaet fra meiotisk anholdelse og tillader ekstraktet at fortsætte ind i interfase. Normalt tilsættes cytochalasin B før knusningsspin for at hæmme dannelsen af F-actin. Det kan dog udelades, hvis F-actin ønskes. Cycloheximid tilsættes også før knusningsspin for at forhindre interfaseekstraktet i at komme ind i den næste mitose. Ekstrakterne placeres efterfølgende i de førnævnte billeddannelseskamre og placeres på et mikroskop. Endelig optages billeder over tid med definerede intervaller af et kamera, der er forbundet til mikroskopet, hvilket producerer time-lapse-billedserier, der fanger ekstraktets dynamiske opførsel i et 2D-felt.

xenopus laevis egg extract preparation and live imaging methods for visualizing dynamic cytoplasmic organization

Traditionelt anvendt til bulk biokemiske assays, Xenopus laevis ægekstrakter er opstået som et kraftfuldt billeddannelsesbaseret værktøj til at studere cytoplasmatiske fænomener, såsom cytokinesis, mitotisk spindeldannelse og samling af kernen. Baseret på tidlige metoder, der afbildede faste ekstrakter, der blev samplet på sparsomme tidspunkter, har de seneste tilgange billede live ekstrakter ved hjælp af time-lapse mikroskopi, der afslører mere dynamiske funktioner med forbedret tidsmæssig opløsning. Disse metoder kræver normalt sofistikerede overfladebehandlinger af billeddannelsesbeholderen. Her introducerer vi en alternativ metode til levende billeddannelse af ægekstrakter, der ikke kræver kemisk overfladebehandling. Det er nemt at implementere og bruger masseproducerede laboratorieforbrugsstoffer til billeddannelse. Vi beskriver et system, der kan bruges til både bredfelts- og konfokalmikroskopi. Det er designet til billeddannelsesekstrakter i et 2-dimensionelt (2D) felt, men kan let udvides til billeddannelse i 3D. Det er velegnet til at studere rumlig mønsterdannelse inden for cytoplasmaet. Med repræsentative data demonstrerer vi den typiske dynamiske organisering af mikrotubuli, kerner og mitokondrier i interfaseekstrakter fremstillet ved hjælp af denne metode. Disse billeddata kan give kvantitativ information om cytoplasmisk dynamik og rumlig organisation.

Denne metode er egnet til levende billeddannelse af cytoplasmisk dynamik ved hjælp af frøægekstrakter, hvilket muliggør undersøgelse af rumlig mønsterinformation i cytoplasmaet. Prøver kan formes til et lag med en ensartet og justerbar tykkelse og afbildes på tværs af et 2D-felt eller 3D-volumen. Denne metode er omkostningseffektiv og nem at implementere.Begynd med at påføre et lag FEP-tape på et glasglas med en rulleapplikator. Afskær overdreven tape over kanterne med et rent barberblad. Forbered derefter FEP tape belagte dæksler på samme måde.Påfør derefter en dobbeltsidet klæbrig billedafstandsstykke på den FEP-båndbelagte side af glideren, så beskyttelsesforingen ikke skrælles. Overfør æg til et 400 ml glasbægerglas og fjern så meget æglægningsbuffer som muligt ved dekantering. Derefter inkuberes æggene i 100 ml frisklavet dejellying opløsning og hvirvler dem forsigtigt.Efter ca. tre minutter hældes opløsningen af og tilsættes 100 ml frisk dejellying opløsning. Fortsæt inkubationen, indtil æggene er tæt pakket, men undgå at efterlade æg i dejellyopløsningen i mere end i alt fem minutter. Efter inkubationen fjernes dejellyopløsningen så meget som muligt og vaskes æggene i 0,25x MMR buffer.Hvirvl æggene, og hæld derefter bufferen af. Gentag et par gange, indtil i alt en liter af bufferen er brugt til vaskene. Vask derefter æggene et par gange med i alt 400 ml æglysebuffer, og fjern unormale æg mellem vaskene.Brug derefter en overførselspipette med en bred skåret spids til at overføre æggene til et 17 ml rundt bundcentrifugerør indeholdende en milliliter æglysebuffer. Drej røret i en klinisk centrifuge ved 400 g i 15 sekunder for at pakke æggene. Fjern derefter så meget af æglysebufferen som muligt ved hjælp af en Pasteur-pipette.Bestem det omtrentlige volumen af de pakkede æg. Derefter tilsættes fem mikrogram pr. milliliter hver af aprotinin, leupeptin og cytochalasin B og 50 mikrogram pr. milliliter cyclohexamid direkte oven på de pakkede æg. Knus æggene ved at centrifuse røret i 15 minutter ved 12.000 g og fire grader Celsius i en svingende spandrotor.Fastgør derefter en 18 gauge nål til en sprøjte. Med nålespidsen skrå opad punkterer røret fra siden i bunden af det cytoplasmatiske lag og genvinder ekstraktet ved at trække langsomt. Overfør det genvundne cytoplasmatiske ekstrakt til et nyt mikrocentrifugerør og hold det på is.Når du er klar til at afbilde, skal du supplere ekstraktet med de ønskede reagenser og fluorescensbilledprober. Fjern den øverste beskyttelsesforing fra billedafstandsstykket på det forberedte dias og deponer ca. syv mikroliter ekstrakt i midten af brønden. Påfør straks FEP-tapebelagt dæksel med FEP-siden vendt mod ekstraktet for at forsegle brønden og hurtigt fortsætte med at billeddanne.Indstil diaset på et omvendt eller opretstående mikroskop med et motoriseret trin og et digitalt kamera. Billede ekstrakterne på de ønskede rumlige positioner i tidsintervaller i både lyse felt- og fluorescenskanaler. Et time-lapse-eksperiment ved hjælp af xenopus laevis-ægekstrakter til at studere cytoplasmaets selvorganisering under interfase er vist her.Efter ca. 20 minutters inkubation ved stuetemperatur var områder udtømt for lysspredende cytoplasmatiske komponenter synlige i både lyse felt- og mitokondrierkanaler. Ved ca. 60 minutter ved stuetemperatur skal et rumligt mønster bestående af cellelignende rum være veletableret med mikrotubuli, der danner en hul kranslignende struktur og mitokondrier tydeligt opdelt i hvert rum. En sammenligning af ekstraktydelse i billeddannende kamre med og uden FEP-bånd på glas er vist her.I kammeret lavet med FEP tapet glas ekstraktet selvorganiseret i normale cellelignende mønstre. I kammeret, hvor glasoverfladerne ikke var dækket af FEP-båndet, viste ekstraktet unormale lyse felt- og mikrotubulimønstre, der blev mere og mere forstyrret over tid. Der blev ikke observeret signifikante forskelle i den nukleare import af GST-mCherry-NLS-proteinet.De vigtige ting at huske er at passivere glasoverfladerne med FEP-tape og fjerne alle dårlige æg under ekstraktforberedelsen. Med dette system kan vi nemt kontrollere tykkelsen af ekstraktprøven og bane vejen for billeddannelse af cytoplasmatiske mønstre i 3D ved hjælp af lysarkmikroskopi.
