Xenopus laevis ei-extracten zijn naar voren gekomen als een krachtig modelsysteem voor op beeldvorming gebaseerde studies van verschillende subcellulaire structuren 10,14,15,16,17,18,21,31,32,33,34,35,36 en cytoplasmatische organisatie op een geheel </s...

Het cytoplasma vormt het hoofdvolume van een cel en heeft een aparte organisatie. De ingrediënten van het eukaryote cytoplasma kunnen zichzelf assembleren tot een breed scala aan ruimtelijke structuren, zoals microtubule-asters en het Golgi-apparaat, die op hun beurt dynamisch worden gerangschikt en omgedraaid, afhankelijk van de identiteit en fysiologische toestand van de cel. Het begrijpen van de ruimtelijke organisatie van het cytoplasma en de link met cellulaire functies is dus belangrijk om te begrijpen hoe de cel werkt. Xenopus laevis ei-extracten worden traditioneel gebruikt voor biochemische bulktests 1,2,3,4,5,6,7,8, maar recent werk vestigt ze als een krachtig live beeldvormingssysteem voor mechanistische studies van cytoplasmatische structuren en hun cellulaire functies 9,10,11, 12,13,14,15,16,17,18. Deze onverdunde extracten behouden vele structuren en functies van het cytoplasma, terwijl ze directe manipulaties van cytoplasmatische inhoud mogelijk maken die niet haalbaar zijn in conventionele celgebaseerde modellen 19,20. Dit maakt ze ideaal voor het karakteriseren van cytoplasmatische verschijnselen en het ontleden van hun mechanistische onderbouwing.
Bestaande methoden voor het afbeelden van extracten vereisen chemische oppervlaktemodificaties of fabricage van microfluïdische apparaten. Een op coverslip gebaseerde methode vereist polyethyleenglycol (PEG) passivering van glazen coverslips21. Een op micro-emulsie gebaseerde methode vereist dampafzetting van trichlooriso (1H, 1H, 2H, 2H-perfluorooctyl) silaan op glasoppervlakken22,23. Microfluïdische systemen maken nauwkeurige controle van het volume, de geometrie en de samenstelling van extractdruppels mogelijk, maar vereisen gespecialiseerde microfabricagefaciliteiten 11,12,24.
Hier introduceren we een alternatieve methode voor het afbeelden van ei-extracten die gemakkelijk te implementeren is en gebruik maakt van direct beschikbare, goedkope materialen. Dit omvat de voorbereiding van een beeldkamer met een dia en een coverslip bedekt met gefluoreerde ethyleenpropyleen (FEP) tape. De kamer kan worden gebruikt voor beeldvormingsextracten met een verscheidenheid aan microscopiesystemen, waaronder stereoscopen en rechtopstaande en omgekeerde microscopen. Deze methode vereist geen chemische behandeling van oppervlakken, terwijl een vergelijkbare optische helderheid wordt bereikt die wordt verkregen met bestaande op glas gebaseerde methoden die hierboven zijn besproken. Het is ontworpen om een laag extracten met een uniforme dikte over een 2D-veld af te beelden en kan eenvoudig worden uitgebreid om een 3D-volume aan extracten af te beelden. Het is zeer geschikt voor time-lapse beeldvorming van collectief cytoplasmatisch gedrag over een groot gezichtsveld.
We hebben interfase-arrested ei-extracten gebruikt om onze beeldvormingsmethode te demonstreren. Het extractpreparaat volgt het protocol van Deming en Kornbluth19. Kortom, eieren die van nature zijn gearresteerd in de metafase van meiose II worden verpletterd door een spin met lage snelheid. Deze spin bevrijdt het cytoplasma van meiotische arrestatie en laat het extract in de interfase gaan. Normaal gesproken wordt cytochalasine B toegevoegd voorafgaand aan de verpletterende spin om de vorming van F-actine te remmen. Het kan echter worden weggelaten als F-actine gewenst is. Cycloheximide wordt ook toegevoegd voorafgaand aan de breekspin om te voorkomen dat het interfase-extract de volgende mitose binnendringt. De extracten worden vervolgens in de eerder genoemde beeldvormende kamers geplaatst en op een microscoop gelegd. Ten slotte worden beelden in de loop van de tijd met gedefinieerde intervallen opgenomen door een camera die op de microscoop is aangesloten, waardoor time-lapse-beeldreeksen worden geproduceerd die het dynamische gedrag van het extract in een 2D-veld vastleggen.

xenopus laevis egg extract preparation and live imaging methods for visualizing dynamic cytoplasmic organization

Xenopus laevis ei-extracten, die traditioneel worden gebruikt voor biochemische bulktests, zijn naar voren gekomen als een krachtig op beeldvorming gebaseerd hulpmiddel voor het bestuderen van cytoplasmatische verschijnselen, zoals cytokinese, mitotische spindelvorming en assemblage van de kern. Voortbouwend op vroege methoden die vaste extracten in beeld brachten die op schaarse tijdstippen waren bemonsterd, beeldden recente benaderingen live-extracten af met behulp van time-lapse-microscopie, waarbij meer dynamische kenmerken met verbeterde temporele resolutie werden onthuld. Deze methoden vereisen meestal geavanceerde oppervlaktebehandelingen van het beeldvormende vat. Hier introduceren we een alternatieve methode voor live beeldvorming van ei-extracten die geen chemische oppervlaktebehandeling vereisen. Het is eenvoudig te implementeren en maakt gebruik van in massa geproduceerde laboratorium verbruiksartikelen voor beeldvorming. We beschrijven een systeem dat kan worden gebruikt voor zowel wide-field als confocale microscopie. Het is ontworpen voor het afbeelden van extracten in een 2-dimensionaal (2D) veld, maar kan eenvoudig worden uitgebreid naar beeldvorming in 3D. Het is zeer geschikt voor het bestuderen van ruimtelijke patroonvorming binnen het cytoplasma. Met representatieve gegevens demonstreren we de typische dynamische organisatie van microtubuli, kernen en mitochondriën in interfase-extracten die met deze methode zijn bereid. Deze beeldgegevens kunnen kwantitatieve informatie verschaffen over cytoplasmatische dynamica en ruimtelijke organisatie.

Xenopus laevis ei-extracten kunnen worden gebruikt om de zelforganisatie van het cytoplasma tijdens de interfase te bestuderen. Figuur 2A toont de resultaten van een succesvol experiment. Deze methode is geschikt voor live beeldvorming van cytoplasmatische dynamica met behulp van kikkereierenextracten, waardoor de studie van ruimtelijke patrooninformatie in het cytoplasma mogelijk wordt. Monsters kunnen worden gevormd tot een laag met een uniforme en instelbare dikte en worden afgebeeld over een 2D-veld of 3D-volume. Deze methode is kosteneffectief en eenvoudig te implementeren.Begin met het aanbrengen van een laag FEP-plakband op een glazen glijbaan met een rollerapplicator. Knip overtollige tape over de randen af met een schoon scheermesje. Bereid vervolgens FEP-tape gecoate afdekslips op dezelfde manier voor.Breng vervolgens een dubbelzijdige kleverige beeldafstandhouder aan op de met FEP-tape gecoate kant van de dia, waarbij de beschermende voering ongepeld blijft. Breng de eieren over in een glazen bekerglas van 400 milliliter en verwijder zoveel mogelijk legbuffer door te decanteren. Incubeer vervolgens de eieren in 100 milliliter vers bereide dejellying-oplossing en draai ze voorzichtig rond.Giet na ongeveer drie minuten de oplossing af en voeg 100 milliliter verse dejellying-oplossing toe. Ga door met de incubatie totdat de eieren stevig zijn verpakt, maar vermijd eieren langer dan vijf minuten in de dejellying-oplossing te laten zitten. Verwijder na de incubatie de dejellying-oplossing zoveel mogelijk en was de eieren in een buffer van 0,25x MMR.Draai de eieren rond en giet vervolgens de buffer af. Herhaal dit een paar keer totdat in totaal een liter van de buffer is gebruikt voor de wasbeurten. Was vervolgens de eieren een paar keer met een totaal van 400 milliliter eierlysisbuffer, waarbij abnormale eieren tussen de wasbeurten worden verwijderd.Gebruik vervolgens een transferpipet met een brede gesneden punt om de eieren over te brengen naar een 17 milliliter ronde bodemcentrifugebuis met één milliliter eierlysisbuffer. Draai de buis in een klinische centrifuge op 400 g gedurende 15 seconden om de eieren te verpakken. Verwijder vervolgens zoveel mogelijk van de eierlysisbuffer met een Pasteur-pipet.Bepaal het geschatte volume van de verpakte eieren. voeg vervolgens vijf microgram per milliliter elk van aprotinine, leupeptine en cytochalasine B en 50 microgram per milliliter cyclohexamide direct bovenop de verpakte eieren toe. Plet de eieren door de buis gedurende 15 minuten te centrifugeren op 12.000 g en vier graden Celsius in een zwaaiende emmerrotor.Bevestig vervolgens een naald van 18 gauge aan een spuit. Met de schuine kant van de naaldpunt naar boven prikt u de buis vanaf de zijkant aan de onderkant van de cytoplasmatische laag door en herstelt u het extract door langzaam te tekenen. Breng het teruggewonnen cytoplasmatische extract over naar een nieuwe microcentrifugebuis en bewaar het op ijs.Wanneer u klaar bent om te imagen, vult u het extract aan met de gewenste reagentia en fluorescentiebeeldvormingsondes. Verwijder de bovenste beschermende voering van de beeldvormende afstandhouder op de voorbereide dia en deponeer ongeveer zeven microliter extract in het midden van de put. Breng onmiddellijk de FEP tape gecoate afdekslip aan met de FEP-kant naar het extract gericht om de put af te dichten en ga snel over tot beeldvorming.Plaats de dia op een omgekeerde of rechtopstaande microscoop met een gemotoriseerd podium en een digitale camera. Beeld de extracten af op de gewenste ruimtelijke posities in tijdsintervallen in zowel heldere veld- als fluorescentiekanalen. Een time-lapse-experiment met xenopus laevis-ei-extracten om de zelforganisatie van het cytoplasma tijdens interfase te bestuderen, wordt hier getoond.Na ongeveer 20 minuten incubatie bij kamertemperatuur waren gebieden uitgeput van lichtverstrooiing cytoplasmatische componenten zichtbaar in zowel heldere veld- als mitochondriënkanalen. Tegen ongeveer 60 minuten bij kamertemperatuur moet een ruimtelijk patroon bestaande uit celachtige compartimenten goed zijn vastgesteld met microtubuli die een holle kransachtige structuur vormen en mitochondriën die duidelijk in elk compartiment zijn verdeeld. Een vergelijking van de extractprestaties in beeldkamers met en zonder FEP-tapes op glas wordt hier getoond.In de kamer gemaakt met FEP getapt glas het extract zelf georganiseerd in normale celachtige patronen. In de kamer waar de glasoppervlakken niet door de FEP-tape werden bedekt, vertoonde het extract abnormale heldere veld- en microtubulepatronen die in de loop van de tijd steeds meer verstoord raakten. Er werden geen significante verschillen waargenomen in de nucleaire import van het GST-mCherry-NLS eiwit.De belangrijke dingen om te onthouden zijn om de glazen oppervlakken te passiveren met FEP-tape en om alle slechte eieren te verwijderen tijdens de extractvoorbereiding. Met dit systeem kunnen we eenvoudig de dikte van het extractmonster regelen, waardoor de weg wordt vrijgemaakt voor het in beeld brengen van cytoplasmatische patronen in 3D, met behulp van light sheet microscopie.
