Das Zytoplasma bildet das Hauptvolumen einer Zelle und hat eine ausgeprägte Organisation. Die Inhaltsstoffe des eukaryotischen Zytoplasmas können sich zu einer Vielzahl von räumlichen Strukturen wie Mikrotubuli-Astern und dem Golgi-Apparat zusammensetzen, die wiederum je nach Identität und physiologischem Zustand der Zelle dynamisch angeordnet und umgedreht werden. Das Verständnis der räumlichen Organisation des Zytoplasmas und seiner Verbindung zu zellulären Funktionen ist daher wichtig, um zu verstehen, wie die Zelle funktioniert. Xenopus laevis Ei-Extrakte wurden traditionell für biochemische Massentests 1,2,3,4,5,6,7,8 verwendet, aber neuere Arbeiten etablieren sie als leistungsfähiges Live-Bildgebungssystem für mechanistische Untersuchungen zytoplasmatischer Strukturen und ihrer zellulären Funktionen 9,10,11, 12,13,14,15,16,17,18. Diese unverdünnten Extrakte bewahren viele Strukturen und Funktionen des Zytoplasmas, während sie eine direkte Manipulation des zytoplasmatischen Inhalts ermöglichen, die in herkömmlichen zellbasierten Modellen nicht möglich sind19,20. Dies macht sie ideal für die Charakterisierung zytoplasmatischer Phänomene und die Analyse ihrer mechanistischen Grundlagen.
Bestehende Methoden zur Bildgebung von Extrakten erfordern chemische Oberflächenmodifikationen oder die Herstellung von mikrofluidischen Geräten. Ein Deckglas-basiertes Verfahren erfordert die Polyethylenglykol (PEG)-Passivierung von Glasdeckgläsern21. Ein mikroemulsionsbasiertes Verfahren erfordert die Gasphasenabscheidung von Trichlor(1H,1H,2H,2H-perfluorooctyl)silan auf Glasoberflächen22,23. Mikrofluidische Systeme ermöglichen eine präzise Kontrolle des Volumens, der Geometrie und der Zusammensetzung von Extrakttröpfchen, erfordern jedoch spezielle Mikrofabrikationsanlagen11,12,24.
Hier stellen wir eine alternative Methode zur Abbildung von Eiextrakten vor, die einfach zu implementieren ist und leicht verfügbare, kostengünstige Materialien verwendet. Dazu gehört die Vorbereitung einer Bildkammer mit einem Objektträger und einem Deckglas, das mit fluoriertem Ethylenpropylen (FEP)-Band beschichtet ist. Die Kammer kann für die Bildgebung von Extrakten mit einer Vielzahl von Mikroskopiesystemen verwendet werden, einschließlich Stereoskopen und aufrechten und inversen Mikroskopen. Dieses Verfahren erfordert keine chemische Behandlung von Oberflächen und erreicht gleichzeitig eine ähnliche optische Klarheit, die mit den oben beschriebenen glasbasierten Methoden erzielt wird. Es wurde entwickelt, um eine Schicht von Extrakten mit einer gleichmäßigen Dicke über ein 2D-Feld abzubilden, und kann leicht erweitert werden, um ein 3D-Volumen von Extrakten abzubilden. Es eignet sich gut für die Zeitrafferbildgebung des kollektiven zytoplasmatischen Verhaltens über ein großes Sichtfeld.
Wir haben Interphase-Arrest-Ei-Extrakte verwendet, um unsere bildgebende Methode zu demonstrieren. Die Extraktzubereitung folgt dem Protokoll von Deming und Kornbluth19. Kurz gesagt, Eier, die natürlich in der Metaphase der Meiose II blockiert werden, werden durch einen Spin mit niedriger Geschwindigkeit zerkleinert. Dieser Spin befreit das Zytoplasma aus dem meiotischen Stillstand und ermöglicht es dem Extrakt, in die Interphase überzugehen. Normalerweise wird Cytochalasin B vor dem zerkleinernden Spin hinzugefügt, um die Bildung von F-Aktinen zu hemmen. Es kann jedoch weggelassen werden, wenn F-Aktin gewünscht wird. Cycloheximid wird auch vor dem Zerkleinerungsspin hinzugefügt, um zu verhindern, dass der Interphasenextrakt in die nächste Mitose gelangt. Die Extrakte werden anschließend in die vorgenannten Bildgebungskammern gegeben und auf ein Mikroskop gelegt. Schließlich werden Bilder über die Zeit in definierten Intervallen von einer an das Mikroskop angeschlossenen Kamera aufgenommen, wodurch Zeitraffer-Bildserien entstehen, die das dynamische Verhalten des Extrakts in einem 2D-Feld erfassen.

Xenopus laevis-Ei-Extrakte, die traditionell für biochemische Massentests verwendet werden, haben sich zu einem leistungsstarken bildgebenden Werkzeug für die Untersuchung zytoplasmatischer Phänomene wie Zytokinese, mitotische Spindelbildung und Aufbau des Kerns entwickelt. Aufbauend auf frühen Methoden, bei denen feste Extrakte abgebildet wurden, die zu spärlichen Zeitpunkten abgetastet wurden, nähern sich neueren Ansätzen Live-Extrakten mit Zeitraffermikroskopie und zeigen dynamischere Merkmale mit verbesserter zeitlicher Auflösung. Diese Methoden erfordern in der Regel anspruchsvolle Oberflächenbehandlungen des bildgebenden Gefäßes. Hier stellen wir eine alternative Methode zur Live-Bildgebung von Ei-Extrakten vor, die keine chemische Oberflächenbehandlung erfordern. Es ist einfach zu implementieren und verwendet massenproduzierte Laborverbrauchsmaterialien für die Bildgebung. Wir beschreiben ein System, das sowohl für die Weitfeld- als auch für die konfokale Mikroskopie eingesetzt werden kann. Es ist für die Abbildung von Extrakten in einem 2-dimensionalen (2D) Feld konzipiert, kann aber leicht auf die Bildgebung in 3D erweitert werden. Es eignet sich gut zur Untersuchung der räumlichen Musterbildung im Zytoplasma. Mit repräsentativen Daten zeigen wir die typische dynamische Organisation von Mikrotubuli, Zellkernen und Mitochondrien in Interphasenextrakten, die mit dieser Methode hergestellt werden. Diese Bilddaten können quantitative Informationen über zytoplasmatische Dynamik und räumliche Organisation liefern.

Xenopus laevis Ei-Extrakte können verwendet werden, um die Selbstorganisation des Zytoplasmas während der Interphase zu untersuchen. Abbildung 2A zeigt die Ergebnisse eines erfolgreichen Experiments. Diese Methode eignet sich für die Live-Bildgebung der zytoplasmatischen Dynamik mit Froschei-Extrakten, die die Untersuchung von räumlichen Musterinformationen im Zytoplasma ermöglichen. Proben können zu einer Schicht mit einer gleichmäßigen und einstellbaren Dicke geformt und über ein 2D-Feld oder 3D-Volumen abgebildet werden. Diese Methode ist kostengünstig und einfach zu implementieren.Beginnen Sie mit dem Auftragen einer Schicht FEP-Klebeband auf einen Glasobjektträger mit einem Rollenapplikator. Schneiden Sie überschüssiges Klebeband über die Kanten mit einer sauberen Rasierklinge ab. Als nächstes bereiten Sie FEP-bandbeschichtete Deckblätter auf die gleiche Weise vor.Tragen Sie dann einen doppelseitigen, klebrigen Abstandshalter auf die mit FEP-Klebeband beschichtete Seite des Objektträgers auf, wobei die Schutzfolie nicht geschält bleibt. Die Eier in einen 400-Milliliter-Glasbecher geben und durch Dekantieren so viel Eiablagepuffer wie möglich entfernen. Dann die Eier in 100 Milliliter frisch zubereiteter Dejelly-Lösung bebrüten und vorsichtig schwenken.Nach etwa drei Minuten gießen Sie die Lösung ab und fügen Sie 100 Milliliter frische Dejelly-Lösung hinzu. Setzen Sie die Inkubation fort, bis die Eier dicht gepackt sind, aber vermeiden Sie es, Eier länger als insgesamt fünf Minuten in der Dejelly-Lösung zu lassen. Entfernen Sie nach der Inkubation die Dejelly-Lösung so weit wie möglich und waschen Sie die Eier in 0,25x MMR-Puffer.Die Eier schwenken und dann den Puffer abgießen. Wiederholen Sie dies einige Male, bis insgesamt ein Liter des Puffers für die Wäschen verwendet wird. Als nächstes waschen Sie die Eier einige Male mit insgesamt 400 Milliliter Eilysepuffer und entfernen Sie abnormale Eier zwischen den Wäschen.Verwenden Sie dann eine Transferpipette mit einer breit geschnittenen Spitze, um die Eier in ein 17-Milliliter-Zentrifugenröhrchen mit rundem Boden zu überführen, das einen Milliliter Eilysepuffer enthält. Drehen Sie das Röhrchen in einer klinischen Zentrifuge bei 400 g für 15 Sekunden, um die Eier zu verpacken. Entfernen Sie dann mit einer Pasteur-Pipette so viel Eilysepuffer wie möglich.Bestimmen Sie das ungefähre Volumen der verpackten Eier. Dann fügen Sie je fünf Mikrogramm pro Milliliter Aprotinin, Leupeptin und Cytochalasin B und 50 Mikrogramm pro Milliliter Cyclohexamid direkt auf die verpackten Eier hinzu. Zerkleinern Sie die Eier, indem Sie das Rohr 15 Minuten lang bei 12.000 g und vier Grad Celsius in einem schwingenden Eimerrotor zentrieren.Als nächstes befestigen Sie eine 18-Gauge-Nadel an einer Spritze. Mit der Nadelspitze nach oben stechen Sie das Röhrchen von der Seite am Boden der zytoplasmatischen Schicht und gewinnen Sie den Extrakt durch langsames Ziehen zurück. Den gewonnenen zytoplasmatischen Extrakt in ein neues Mikrozentrifugenröhrchen geben und auf Eis halten.Wenn Sie zum Abbilden bereit sind, ergänzen Sie den Extrakt mit den gewünschten Reagenzien und Fluoreszenz-Bildgebungssonden. Entfernen Sie die obere Schutzfolie aus dem Bildabstandshalter auf dem vorbereiteten Objektträger und legen Sie etwa sieben Mikroliter Extrakt in der Mitte des Bohrlochs ab. Tragen Sie sofort den mit FEP-Klebeband beschichteten Abdeckstreifen auf, wobei die FEP-Seite dem Extrakt zugewandt ist, um das Bohrloch zu versiegeln und schnell mit der Bildgebung fortzufahren.Stellen Sie den Objektträger auf ein inverses oder aufrechtes Mikroskop mit einem motorisierten Tisch und einer Digitalkamera. Bilden Sie die Extrakte an den gewünschten räumlichen Positionen in Zeitintervallen sowohl in hellen Feld- als auch in Fluoreszenzkanälen ab. Ein Zeitrafferexperiment mit Xenopus laevis Ei-Extrakten zur Untersuchung der Selbstorganisation des Zytoplasmas während der Interphase ist hier gezeigt.Nach etwa 20 Minuten Inkubation bei Raumtemperatur waren Bereiche mit lichtstreuenden zytoplasmatischen Komponenten sowohl in hellen Feld- als auch in Mitochondrienkanälen sichtbar. Nach etwa 60 Minuten bei Raumtemperatur sollte ein räumliches Muster aus zellartigen Kompartimenten gut etabliert sein, wobei Mikrotubuli eine hohle kranzartige Struktur bilden und Mitochondrien klar in jedes Kompartiment unterteilt sind. Ein Vergleich der Extraktleistung in Bildgebungskammern mit und ohne FEP-Bänder auf Glas wird hier gezeigt.In der Kammer aus FEP-Klebeglas organisiert sich der Extrakt selbst in normale zellartige Muster. In der Kammer, in der die Glasoberflächen nicht vom FEP-Band bedeckt waren, zeigte der Extrakt abnormale helle Feld- und Mikrotubulimuster, die im Laufe der Zeit zunehmend gestört wurden. Es wurden keine signifikanten Unterschiede beim nuklearen Import des GST-mCherry-NLS-Proteins beobachtet.Es ist wichtig, die Glasoberflächen mit FEP-Klebeband zu passivieren und alle schlechten Eier während der Extraktvorbereitung zu entfernen. Mit diesem System können wir die Dicke der Extraktprobe leicht kontrollieren und den Weg für die Abbildung zytoplasmatischer Muster in 3D mit Hilfe der Lichtblattmikroskopie ebnen.
