抽出物をイメージングするための既存の方法は、化学的表面修飾、またはマイクロ流体デバイスの製造を必要とします。カバーガラスベースの方法の1つは、ガラスカバーガラス21のポリエチレングリコール(PEG)パッシベーションを必要とする。マイクロエマルジョンベースの方法では、ガラス表面22,23へのトリクロロ(1H,1H,2H,2H-パーフルオロオクチル)シランの蒸着が必要です。マイクロ流体ベースのシステムは、抽出液滴の量、形状および組成の正確な制御を可能にするが、特殊な微細加工設備を必要とする11、12、24。

我々は、イメージング方法を実証するために、間期停止卵抽出物を使用しました。抽出物の調製は、デミングおよびコーンブルース19のプロトコルに従います。簡単に言えば、減数分裂IIの中期に自然に停止した卵は、低速スピンによって押しつぶされます。このスピンは細胞質を減数分裂停止から解放し、抽出物が間期に進むことを可能にします。通常、サイトカラシンBは、F-アクチン形成を阻害するために、破砕スピンの前に添加される。但し、F-アクチンを希望する場合は省略することができる。シクロヘキシミドは、間期抽出物が次の有糸分裂に入るのを防ぐために、粉砕スピンの前にも添加されます。続いて、抽出物を前述のイメージングチャンバーに入れ、顕微鏡上に配置します。最後に、顕微鏡に接続されたカメラによって定義された間隔で画像が経時的に記録され、2Dフィールドでの抽出物の動的挙動をキャプチャするタイムラプス画像シリーズが生成されます。

この方法は、カエルの卵抽出物を用いた細胞質動態のライブイメージングに適しており、細胞質内の空間パターン情報の研究を可能にします。サンプルは、均一で調整可能な厚さの層に成形し、2Dフィールドまたは3Dボリュームにわたって画像化することができます。この方法は費用効果が高く、実装も簡単です。ローラーアプリケーターでスライドガラスにFEP粘着テープの層を塗布することから始めます。きれいなかみそりの刃で端の余分なテープを切り取ります。次に、FEPテープでコーティングされたカバースリップを同じ方法で準備します。次に、両面粘着性イメージングスペーサーをスライドのFEPテープコーティングされた側に適用し、保護ライナーを剥がさないようにします。卵を400ミリリットルのガラスビーカーに移し、デカントしてできるだけ多くの産卵バッファーを取り除きます。次に、100ミリリットルの新たに調製したデゼリー溶液で卵をインキュベートし、穏やかに回転させます。約3分後、溶液を注ぎ、100ミリリットルの新鮮なデゼリー溶液を加えます。卵がしっかりと詰まるまで孵卵を続けますが、合計5分以上卵を脱ゼリー溶液に残さないでください。インキュベーション後、脱ゼリー溶液をできるだけ取り除き、0.25倍のMMRバッファーで卵を洗います。卵を渦巻き、バッファーを注ぎます。合計1リットルのバッファーが洗浄に使用されるまで、数回繰り返します。次に、合計400ミリリットルの卵溶解バッファーで卵を数回洗浄し、洗浄の間に異常な卵を取り除きます。次に、先端が広いトランスファーピペットを使用して、1ミリリットルの卵溶解バッファーを含む17ミリリットルの丸底遠心チューブに卵を移します。チューブを400 gの臨床用遠心分離機で15秒間回転させて、卵を詰めます。次に、パスツールピペットを使用してできるだけ多くの卵溶解バッファーを除去します。パックされた卵のおおよその量を決定します。次に、アプロチニン、ロイペプチン、サイトカラシンBをそれぞれ1ミリリットルあたり5マイクログラム、シクロヘキサミドを1ミリリットルあたり50マイクログラムをパックされた卵の上に直接加えます。スイングバケットローターで12, 000 g、摂氏4度でチューブを15分間遠心分離して卵を粉砕します。次に、18ゲージの針を注射器に取り付けます。針先ベベルを上に向けて、細胞質層の下部にある側からチューブを穿刺し、ゆっくりと描画して抽出物を回収します。回収した細胞質抽出物を新しいマイクロ遠心チューブに移し、氷上に保管します。イメージングの準備ができたら、目的の試薬と蛍光イメージングプローブを抽出物に補充します。準備したスライドのイメージングスペーサーから上部の保護ライナーを取り外し、ウェルの中央に約7マイクロリットルの抽出物を堆積させます。FEP側を抽出物に向けてFEPテープでコーティングされたカバースリップをすぐに適用してウェルを密閉し、すぐにイメージングに進みます。電動ステージとデジタルカメラを備えた倒立顕微鏡または正立顕微鏡にスライドをセットします。明視野チャンネルと蛍光チャンネルの両方で時間間隔で目的の空間位置で抽出液を画像化します。アフリカツメガエル卵抽出物を使用して、間期中の細胞質の自己組織化を研究するタイムラプス実験をここに示します。室温で約20分間インキュベートした後、光散乱細胞質成分が枯渇した領域は、明視野チャネルおよびミトコンドリアチャネルの両方で可視であった。室温で約60分までに、細胞状の区画からなる空間パターンが十分に確立され、微小管が中空の花輪のような構造を形成し、ミトコンドリアが各区画に明確に仕切られるはずです。ここでは、ガラス上のFEPテープを使用した場合と使用しない場合のイメージングチャンバーでの抽出性能の比較を示します。FEPテープガラスで作られたチャンバー内で、抽出物は正常な細胞様パターンに自己組織化しました。ガラス表面がFEPテープで覆われていないチャンバーでは、抽出物は異常な明視野と微小管パターンを示し、時間の経過とともにますます破壊されました。GST-mCherry-NLSタンパク質の核内輸入に有意差は観察されなかった。覚えておくべき重要なことは、FEPテープでガラス表面を不動態化し、抽出物の調製中にすべての悪い卵を取り除くことです。このシステムでは、抽出サンプルの厚さを簡単に制御でき、ライトシート顕微鏡を使用して細胞質パターンを3Dでイメージングする道が開かれます。
