암의 특정 미세 환경을 모델링하는 것은 암 발달을 이해하고 적절한 치료를 탐구하는 데 필수적입니다. 동물 모델은 강한 생리적 관련성 때문에 널리 사용되었지만 종 차이 및 윤리적 문제와 관련된 문제는5. 더욱이, 기존의 2D 및 3D 모델은 심층 분석을 위해 암세포의 조작 및 실시간 이미징을 허용하지만, 그들의 건축 및 세포 복잡성은 완전히 재구성될 수 없다. 예를 들어, 암 스페로이드 모델은 스페로이드에서 암세포 응집이 코어에서 저산소증을 자연적으로 생성할 수 있기 때문에 널리 사용되고 있다. 더욱이, 균일한 크기의 셀룰러 스페로이드의 다수는 플라스틱 또는 실리콘 기반 멀티웰 시스템을 사용하여 생산되었다6,7. 그러나, 종래의 플랫폼을 통해 암 조직의 정확한 이질적 구조를 포착하는 것과 관련하여 유연성이 낮을수록 암 연구를 개선하기 위한 고생물학적 플랫폼을 구축하기 위한 첨단 생체 제조 기술의 확립이 요구되고있다 8.
3D 미생물학적 시스템(MPS)은 암세포9의복잡한 기하학적 및 병리학적 진행을 재구성하는 유용한 도구이다. 암세포가 생화학적 그라데이션을 감지함에 따라 성장인자와 케모키인의 생화학적 그라데이션과 시스템에서 재현된 기계적 이질성, 암 발달의 중요한 특징은 시험관내에서 조사될 수 있다. 예를 들어, 암 생존가능성, 전이성 악성, 및 다양한 산소 농도에 따른 약물 내성은 MPSS10,11을사용하여 연구되고 있다. 최근의 발전에도 불구하고, 체외 모델의 저산소 조건을 생성하는 것은 물리적 가스 펌프와의 연결을 포함하여 복잡한 제조 절차에 의존합니다. 따라서 암별 마이크로 환경을 구축하는 간단하고 유연한 방법이 필요합니다.
3D 세포 인쇄 기술은 토착 생물학적 구조를 재구성하기 위해 생체 재료의 공간 적 배치를 정밀하게 제어하여 상당한 주목을 받고있다(12) 특히, 이 기술은 암 미세 환경의 공간 적 특징을 구축하기 위한 높은 제어력과 타당성으로 인해 3D 저산소증 모델의 기존 한계를 극복한다. 또한 3D 프린팅은 층별 공정을 통해 컴퓨터 지원 제조를 용이하게 하여 실제 조직 아키텍처를 모방하기 위해 복잡한 형상을 신속하고 정확하며 재현 가능한 시공을 제공합니다. 3D MPS에 대한 기존 제조 전략의 장점 외에도, 암 진행의 병리학적 특징은 생화학적, 세포 및 생물물리학성분(13,14)을패턴화하여 재현될 수 있다.

3d cell-printed hypoxic cancer-on-a-chip for recapitulating pathologic progression of solid cancer
