このプロトコルは、生体内での脂肪組織の生理学を忠実に再現する白および茶色の脂肪細胞の細胞文化モデルを生成するための簡単な戦略を提供する。このプロトコルは、新生児マウスからの白色および褐色の前読細胞と、この方法を用いて単離する細胞の両方を同時に分離することを可能にし、高い増殖能力および分化可能性を評価する。このアプローチを用いて単離された細胞は、他の培養モデルよりも脂肪組織の複雑さを反映している。肥満や糖尿病のアディポサイト機能をより正確に研究するために使用することができます。.私たちは主に肥満における脂肪組織機能不全の理解に焦点を当てています。しかし、この方法で調製された細胞は、発生生物学における細胞に関する基本的な質問を研究するためにも使用することができる。実験を成功させる鍵は、白い脂肪の貯蔵所をこれらの拠点として識別する方法を決定することですが、これは小さいですが、非常に豊富で増殖する豊富な豊富な前読細胞は、子犬で区別するのが難しい場合があります。視覚的なデモンストレーションは、このプロトコルがいかに簡単であるかを示し、白と茶色の脂肪のデポを見つけて解剖するためのヒントを提供することを可能にします。デポの隔離を見ることは非常に役に立ちます。皮下白脂肪組織を採取するには、安楽死させた新生児の腹部の周りの皮膚を切断し、脚の下の皮膚をそっと引っ張る。皮膚組織を採取することなく、皮膚の内側または四頭筋の上に付着した透明または白い細長い組織として現れる脂肪貯蔵所を慎重に収穫する。採取したデポをPBSでリンスし、250マイクロリットルのPBSと2Xの2X分離バッファーを含む1.5ミリリットルのチューブに脂肪組織を氷の上に置きます。間肩蓋間の茶色の脂肪組織を収集するには、頭の上に肩甲骨から皮膚を引っ張り、肩甲骨の間に位置する深い赤茶色の脂肪組織を持ち上げます。慎重に体からそれを取り外し、一貫性と色のために組織をチェックするために脂肪の周りに切開を行います。すすいだ後、250マイクロリットルのPBSと2X分離バッファーの200マイクロリットルを含む2番目の1.5ミリリットルのチューブに組織を氷の上に置きます。すべてのデポが収集されたら、はさみを使用して各デポをチューブ内で4〜6回直接軽くミンチし、各チューブに2X分離バッファで10Xコラゲナーゼの50マイクロリットルを追加します。その後、チューブを素早く反転させて、温度制御ミキサーで30分間摂氏37度、毎分1400回転で組織を混合して配置します。消化の終わりに、チューブを氷の上に戻し、100ミクロンの細胞ストレーナーを通して消化組織を新しい50ミリリットルチューブにフィルター処理します。細胞収量を最大化するには、各チューブを1ミリリットルの分離培地で洗い流し、この洗浄をストレーナーにフィルターします。茶色と白の脂肪組織溶液を、新鮮な分離培地でデポあたりウェルあたり2ミリリットルの組織懸濁液に希釈します。その後、白い脂肪組織の懸濁液を4〜6つの井戸のそれぞれに2ミリリットル、茶色の脂肪組織懸濁液の2ミリリットルを、井戸あたり1100ミクロンフィルターを通して6つのウェルプレートの2〜3つの井戸のそれぞれにプレートします。すべての細胞がめっきされたら、60〜90分間細胞培養インキュベーターにプレートを置きます。インキュベーションの終わりに、2ミリリットルの無血清培地で各ウェルをよく洗い、プレートを穏やかに攪拌してウェルの底から血液細胞を取り外します。3回の清水の後、2ミリリットルの新鮮な分離培地をウェルに加え、プレートを細胞培養インキュベーターに戻します。細胞が80〜90%の合流度に達したら、新しい目的地プレートに無菌0.1%ゼラチンを37°Cで10分間蒸留水でコーティングします。プレートがインキュベートされている間に、細胞培養インキュベーターで3分間トリプシンで処理する前にPBSで細胞を洗浄します。細胞が剥離したら、細胞の懸濁液を数回ピペットして細胞の回復を最大化し、細胞を新しいチューブに移します。すべての細胞が採取されたら、1ミリリットルの分離培地でそれぞれをよく洗い、細胞懸濁液で洗浄します。遠心分離によって細胞を収集し、カウント用の分離培地の3〜5ミリリットルにペレットを再懸濁します。新鮮な分離培地でめっきに適した濃度に細胞を希釈し、余分なゼラチンを除去するためにPBSでゼラチンコーティングされたプレートを洗浄します。次に、ゼラチンコーティングされたプレートに細胞を播種し、細胞を細胞培養インキュベーターに戻します。細胞が合流したら、分離培地を分化培地に置き換えます。褐色脂肪組織を予見細胞に分化する場合は、ナノモルトリヨードサイロニンを1つ加える。48時間後、培地を培養ごとに適切な量のメンテナンス培地でリフレッシュします。このとき、細胞内の小さな液滴の蓄積は、明るいフィールド顕微鏡の下で見えるようになるはずです。濾過後も、細胞の残骸や血液細胞の中には細胞懸濁液の中に残るものもあります。めっきの1時間後に穏やかに打つ場合、前細孔細胞がウェルの底に急速に付着するので、関連性のない細胞が除去されます。新生児から分離された白色および褐色の前向き細胞は両方とも高い増殖能力を有するが、白い前置細胞の収率は一般的に、デポ当たり茶色の予後細胞の2倍である。したがって、同期培養が望ましい場合、白色の前置細胞のこの開始密度をそれに応じて計算しなければならない。分化の終わりに、細胞は脂質滴を積み込んでいるように見え、それぞれ白色脂肪細胞と茶色の脂肪細胞の古典的なマーカーを発現する。この基礎条件下での白色および褐色のジポサイトのミトコンドリアストレス試験における酸素消費量の比較分析において、ミトコンドリア機能の既知の刺激装置に応答して、各細胞タイプのこの特定の生体エネルギー能が観察できる。私たちは生きた細胞を隔離しており、数日間培養したくてはなることを覚えておいてください。したがって、汚染を避けるために、分離中に無菌状態を維持し、その後の培養を損なう可能性があります。この方法は、機能アッセイ、遺伝的または化学的スクリーン、またはオミックスプロファイリングを含むさまざまな用途に使用できる完全に成熟したアディポサイトを生成し、アディポサイト機能に影響を与える細胞自律的メカニズムを研究する。
