De kendte nucleocapsid-lignende partikel (NCLP) strukturer af nonsegmented negative-sense (NNS) RNA-vira viser, at de samlede NCLPs er de komplekse N med vært cellulære RNA'er, når overekspresseret i bakterielle eller eukaryote udtrykssystemer15,16,17,18,19. Ikke-segmenterede RNA-vira (negative-sense) omfatter mange betydelige menneskelige patogener, såsom rabies, ebola og respiratorisk syncytial virus (RSV)1,2. RSV er den hyppigste årsag til luftvejssygdomme såsom bronchiolitis og lungebetændelse hos små børn og ældre voksne over hele verden3. I øjeblikket er der ingen effektive vacciner eller antivirale behandlinger til rådighed til at forebygge eller behandle RSV4. Som en del af livscyklussen fungerer RSV-genomet som skabelon for replikation af RSV RNA-afhængig RNA-polymerase for at producere et antigenom, som igen fungerer som skabelon til at generere et afkomgengenom. Både genom og antigenome RNA'er er helt indkapslet af nucleoprotein (N) til at danne nucleocapsids (NCs)3. Da NC'erne fungerer som skabeloner for både replikering og transskription af RSV-polymerasen, er korrekt NC-samling afgørende for, at polymerasen kan få adgang til skabelonerne til RNA-syntese5. Interessant, baseret på de strukturelle analyser af NNS virale polymeraser, er det hypotese, at flere N proteiner forbigående dissociate fra NCs at give adgang til polymerase og rebind til RNA efter RNA syntese6, 7,8,9,10,11,12.
I øjeblikket er RSV RNA polymeriseringsanalysen blevet etableret ved hjælp af renset RSV polymerase på korte nøgne RNA-skabeloner13,14. RSV-polymerase's aktiviteter når imidlertid ikke optimale, som observeret i de ikke-procesive og abortive produkter, der genereres af RSV polymerase, når du bruger nøgne RNA-skabeloner. Manglen på NC med virusspecifikt RNA er en primær barriere for den yderligere mekanistiske forståelse af RSV RNA-syntesen. Derfor bliver brug af en autentisk RNA-skabelon et kritisk behov for at fremme den grundlæggende viden om RSV RNA-syntese. De kendte strukturer af nucleocapsid-lignende partikler (NCPI'er) fra RSV og andre NNS RNA-vira afslører, at RNA'erne i NCLP'erne enten er tilfældige cellulære RNA'er eller gennemsnitlige virale genomiske RNA'er15,16,17,18,19. Sammen er den største forhindring, at N binder ikke-specifikt til cellulære RNA'er for at danne NCLPs, når N er overekspresseret i værtscellerne.
For at overvinde denne forhindring etablerede vi en protokol for at opnå RNA-fri (N0)først og samle N0 med autentisk viral genomisk RNA i NCLPs20. Princippet i denne protokol er at opnå en stor mængde rekombinant RNA-fri N (N0)ved at co-udtrykke N med en anstandsdame, N-terminal domæne RSV phosphoprotein (PNTD). Den rensede N0P kunne stimuleres og samles i NCB'er ved at tilføje RSV-specifikke RNA oligoer, og under samlingsprocessen forskydes chaperon PNTD ved tilsætning af RNA oligos.
Her beskriver vi en protokol for generering og samling af RSV RNA-specifikke NCs. I denne protokol beskriver vi molekylær kloning, proteinpræparat, in vitro-samling og validering af den komplekse samling. Vi fremhæver kloningsstrategien for at generere bi-cistronic konstruktioner til protein coexpression til molekylær kloning. Til proteinpræparat beskriver vi procedurerne for cellekultur, proteinudvinding og rensning af proteinkomplekset. Derefter diskuterer vi metoden til in vitro-samling af RSV RNA-specifikke NCs. Endelig bruger vi størrelsesudelukkelseskromatografi (SEC) og negativ pletelektronmikroskopi (EM) til at karakterisere og visualisere de samlede NCKLU'er.

Brugen af en autentisk RNA-skabelon er afgørende for at fremme den grundlæggende viden om viral RNA-syntese, der kan guide både mekanistisk opdagelse og analyseudvikling inden for virologi.
