De kjente nukleocapsid-lignende partikkelstrukturene (NCLP) til de ikke-segmenterte negative sansene (NNS) RNA-virusene viser at de monterte NCLPene er de komplekse N med vertscelle-RNAer når de overeksponeres i bakterielle eller eukaryote uttrykkssystemer15,16,17,18,19. Ikke-segmentert negativ sans (NNS) RNA-virus inkluderer mange betydelige menneskelige patogener, for eksempel rabies, ebola og respiratorisk synytialvirus (RSV)1,2. RSV er den ledende årsaken til luftveissykdom som bronkiolitt og lungebetennelse hos små barn og eldre voksne over hele verden3. For tiden er ingen effektive vaksiner eller antivirale terapier tilgjengelige for å forhindre eller behandle RSV4. Som en del av livssyklusen fungerer RSV-genomet som mal for replikering av RSV RNA-avhengig RNA-polymerase for å produsere et antigenom, som igjen fungerer som mal for å generere et avkomgenom. Både genom og antigenom RNAer er helt innkapslet av nukleoproteinet (N) for å danne nukleocapsids (NCs)3. Fordi NCene fungerer som maler for både replikering og transkripsjon av RSV-polymerasen, er riktig NC-montering avgjørende for polymerase for å få tilgang til malene for RNA-syntese5. Interessant, basert på de strukturelle analysene av NNS virale polymeraser, er det hypoteset at flere N-proteiner midlertidig dissosierer fra NCene for å tillate tilgang til polymerase og rebind til RNA etter RNA-syntesen6,7,8,9,10,11,12.
For tiden er RSV RNA-polymerisasjonsanalysen etablert ved hjelp av renset RSV-polymerase på korte nakne RNA-maler13,14. RSV-polymerasens aktiviteter når imidlertid ikke optimale, som observert i de ikke-prosessive og abortive produktene som genereres av RSV-polymerasen ved bruk av nakne RNA-maler. Mangelen på NC med virusspesifikk RNA er en primærbarriere for den videre mekanistiske forståelsen av RSV RNA-syntesen. Derfor blir bruk av en autentisk RNA-mal et kritisk behov for å fremme den grunnleggende kunnskapen om RSV RNA-syntese. De kjente strukturene til nukleocapsid-lignende partikler (NCLPer) fra RSV og andre NNS RNA-virus avslører at RNAene i NCLPene enten er tilfeldige cellulære RNAer eller gjennomsnittlig viral genomiske RNAer15,16,17,18,19. Sammen er det viktigste hinderet at N binder seg ikke spesifikt til cellulære RNAer for å danne NCLPer når N er overekspressert i vertscellene.
For å overvinne dette hinderet etablerte vi først en protokoll for å få RNA-fri (N0) og montere N0 med autentisk viral genomisk RNA i NCLPer20. Prinsippet i denne protokollen er å oppnå en stor mengde rekombinant RNA-fri N (N0) ved å uttrykke N sammen med en anstand, N-terminaldomenet til RSV fosfoprotein (PNTD). Den rensede N0P kan stimuleres og monteres i NCLPer ved å legge til RSV-spesifikke RNA oligos, og under monteringsprosessen forskyver chaperone PNTD ved tillegg av RNA oligos.
Her beskriver vi en protokoll for generering og montering av RSV RNA-spesifikke NCer. I denne protokollen beskriver vi molekylær kloning, proteinpreparat, in vitro-montering og validering av den komplekse samlingen. Vi fremhever kloningsstrategien for å generere bi-cistronic konstruksjoner for proteinkoekspressjon for molekylær kloning. For proteinpreparat beskriver vi prosedyrene for cellekultur, proteinutvinning og rensing av proteinkomplekset. Deretter diskuterer vi metoden for in vitro-montering av RSV RNA-spesifikke NCer. Til slutt bruker vi størrelseseksklusjonskromatografi (SEC) og negativ flekkelektronmikroskopi (EM) for å karakterisere og visualisere de monterte NCLPene.

Bruken av en autentisk RNA-mal er avgjørende for å fremme den grunnleggende kunnskapen om viral RNA-syntese som kan lede både mekanistisk oppdagelse og analyseutvikling i virologi. Basert på UV-absorbanskjernens renhetsforhold inneholdt N0 P både N og N-terminal P i full lengde, men inneholdt ikke cellulær RNA.Den rensede N0 P kunne deretter stimuleres og monteres i nukleocapsid som partikler på elektronmikroskopigitter via inkubasjon med spesifikke RNA oligos som demonstrert. Ved rensing av proteinet unngå luftbobler under kolonnerensing og fortynn prøven med QA-buffer for å justere pH- og saltkonsentrasjonen før du laster prøven på kolonnen.
