ホーム熱ブランケットに関連付けられている潤滑直腸温度プローブをマウス直腸に挿入します。マウス温度を平衡化して、温度を37°Cに設定します。これには最大 5 ~ 10 分かかります。上部メニューまたはスキャナ設定アイコンからアクセスできる[スキャナ設定]を選択します。スキャン領域は、対象領域に合わせて X、Y 座標を変更して調整する必要があります。スキャン速度は、スキャンの解像度に依存します。解像度が高いほど、スキャン時間が長くなります。反復スキャンでは、全球の拡散に焦点を当て、解剖灌流に焦点を当てた高分解能とは対照的に、ピクセル当たり4ミリ秒の最高スキャン速度で十分である。繰り返しスキャンを実行する場合は、[繰り返しおよびライン スキャン] タブを選択します。スキャンの数は変更できます。この場合、3回のスキャンと繰り返し間隔。繰り返し間隔の最小時間は、スキャン領域とスキャン解像度によって決定される、ボックスの右側のグレー表示領域に表示される推定スキャン時間です。数秒を追加すると、ユーザーはスキャンの間に必要に応じてマウスを一時停止し、マウスの位置を変更できます。この場合のスキャン時間の合計は 3 分 21 秒です。[イメージ スキャン] タブを選択し、[マーク] ボタンを選択します。これにより、レーザーがスキャン領域の輪郭を描きます。スキャン対象のターゲットがマークされた領域内になるように、マウスの位置を調整します。繰り返しスキャンアイコンを選択し、再生ボタンを押してスキャンを開始することで、測定を繰り返し開始します。スキャン距離を確認するポップアップウィンドウが表示されます。[OK] をクリックしてスキャンを開始します。後肢虚血の誘導前のマウスの最初のスキャンは、リアルタイムで示されている。2回目と3回目の繰り返しスキャンは、16倍の速度に加速されます。メインウィンドウには、ソフトウェアによって生成されたスキャンの結果が表示されます。右下の差し込みは、マウスのフットパッドをスキャンするレーザーを示しています。フットパッドまたはフットパッドおよび後頭のふくらはぎのスキャンのために、後肢を尾部に向かって伸ばした状態で位置を決めやすく、より一貫した位置合わせが得られます。大腿動脈、伏在動脈および副次表は、大腿部およびふくらはぎの腹側表面に非常に近い。したがって、これらの対象領域を使用する場合は、スピーニンポジショニングが好ましい。マウスの動きをスキャン中にマウスを監視します。マウスがスキャン中にスキャン領域に足パッドが表示されなくなるほど動く場合は、スキャンを再開します。マウスフットパッド位置の小さなバリエーションは、解析ソフトウェアに対応できます。また、ホームソアミックブランケットを使用しても変動する可能性があるため、スキャンプロセス中にマウス温度を監視します。マウス温度に過度な変動がある場合、スキャンの間に大きな変動が生じる可能性があります。一般に、36.8 ~ 37.2 の温度範囲は許容可能なデータになります。キャプチャしたスキャンを [名前を付けて保存] ウィンドウの下に、マウス識別子とタイム ポイントを含むファイル名を付けて保存し、データ分析を容易にします。必要に応じて、マウスとタイムポイントの詳細を[件名の詳細]ウィンドウに入力します。マウスを上肢の位置でスキャンした腹側後肢は、スキャン領域を調整した後にここに示される。マウスの上向きの提案は、足の側面の可変部分がスキャンされることになる点に注意してください。直腸温度プローブを取り外し、70%エタノールで消毒して、次のマウスの準備が整います。イソフルランを0%に減らす アイコンを選択します。これにより、対象地域の統計情報を表示するウィンドウが表示されます。データレビューでは、平均拡散単位(PU)では、各フットパッドの繰り返しスキャンで値が100~150個を超えるPUは変化しません。これは許容可能なスキャンであり、スキャン中にマウスが十分に平衡化されたことを示します。100 から 150 PU を超える変動を持つ平均 PU 値は、繰り返しスキャンを行う場合に、操作されていないマウス フットパッドの平均灌流の 10% を超える値に相当すると、マウスが十分に平衡化されておらず、繰り返しスキャンが強く推奨されることを示します。スキャンが完了した直後にクイック分析を実行すると、マウスの麻酔中に繰り返しスキャンを実行でき、スキャンが後日分析され、スキャンが繰り返しスキャンの間に有意な変動が生じた場合に発生する可能性のあるデータポイントを失うことを防ぐことができます。結果は、マウスが最初に血管化し、その次に固有の副次的な組み合せを開発するとして、制御後肢灌流に対する外科的後肢拡散の比率として表され、LDPIによる血流回復は術後の時間経過を通じて見るべきである。回復の程度は、マウス株および後肢虚血モデルの重症度に依存する。この図は、LDPIが大腿動脈結紮およびP27ノックアウトおよび野生型C57Bl/6マウスの後の時間の経過に関するフットパッド血流回復を測定するために使用された28日間にわたる完成したタイムコース実験を示している。野生型マウスのP27ノックアウトはいずれも、一般化マトリックスメタロプロテイナーゼ阻害剤である経口ドキシサイクリンで治療した場合の血流回復を損なっていたため、MMP活性が動脈新生を促進することを示唆した。ゲイン設定とバックグラウンドしきい値は、時間コースの実験全体で一定に保つ必要があります。背景のしきい値が低すぎると、バックグラウンド の拡散単位に増やすと、どの解析にも含まれない灰色の背景になります。フラックス画像のパレットまたはカラースケールは、測定ソフトウェアと画像レビューソフトウェアの両方で調整できます。スケールの最大値を超えるすべての拡散値は、最大の拡散色として表示されます。この場合は、赤。たとえば、ダイナミック レンジを 0 から 1500 に変更すると、1000 ~ 1500 のプロフュージョン単位の間の領域が赤で表示されるのではなく、色のグラデーションを使用して表示されるようになったので、繰り返しスキャンのバリエーションが目にわかりやすくなります。ここで示すスキャンは、繰り返しスキャンの開始時にコア体温が最初に36.8を下回ったマウスで得られ、繰り返しスキャンを繰り返すと増加した。なお、対照フットパッド平均拡散は虚血性フットパッドよりも寒いコア体温によってより影響を受けた。より涼しい体温を1回スキャンすると、このデータポイントに対して高い拡散比が記録される。C57Bl/6マウスで見られるようなマウス皮膚色素沈着の変動は、スキャン領域が背景閾値を下回ることになり、色素領域を含む対象領域の分析が困難になる可能性がある。マウスのフットパッドは、一般的には色素沈着が多く、このマウス株をスキャンするのに最も便利な領域です。研究者が腹側後肢を研究したい場合は、白いマウス株を選択する必要があります。LDPIは、全体的な動脈灌流の反射として後肢真皮拡散を測定するための効果的で、容易に行われ、反復可能な方法である。LDPI
