1. 将成人成纤维细胞直接转化为神经元祖细胞

<ol><li>使用主要的人类皮肤成纤维细胞，可以从细胞库或皮肤活检31获得。在 5% CO2 组织培养孵化器中培养 37 °C 的培养细胞。在实验中使用之前，至少1-2个通道的通道细胞在解冻后。细胞准备好后，在室温下用人体纤维素（PBS稀释1：200）涂上6井板，时间为15-20分钟。</li>

	<li>与所有四个逆转录病毒载体（10月3/4，Klf4，Sox2和c-Myc）一口井 - 使用多种感染（MOI）为10的快速生长或15缓慢生长成纤维细胞（转导效率应超过70%或以上的正细胞为每个载体）。将常规成纤维细胞介质添加到病毒混合物中，每口井的总体积为 1 mL，并在 5% CO2 组织培养孵化器中以 37 °C 的温度在一夜之间孵化。将第二个井未经处理，并将细胞返回组织培养孵化器。 注：逆转录病毒载体可以由供应商购买或内部制作，协议可在线32。</li>

<li>继续观察细胞并每天更换介质（转换介质的 1-2 mL）。</li>

	<li>在转换介质中大约 5-6 天后（对于困难的细胞系最多 12 天），或当细胞变得非常密集时，通过它们 1：2 或最大 1：3。 注意：在整个转换过程中保持细胞的高密度非常重要。</li>
</ol>2. 转换和 iNPC 拆分程序
<ol><li>加热细胞分离溶液（如 Accutase），在室温下涂抹适当数量的 6 井与纤维素（PBS 中 1：200，涂层体积 1 mL）的 6 井，时间为 15-20 分钟。纤维素涂层可以更长，没有负面影响，但应小心不要缩短涂层时间。</li>
	<li>使用PBS小心地清洗细胞，而不会分离任何细胞（使用井壁轻轻地将PBS涂抹在细胞上）。小心地用移液器或吸入器取出 PBS。</li>
	<li>加入 0.5 mL 的细胞分离溶液，在 37 °C 的组织培养孵化器中孵化 2-3 分钟。在显微镜下检查，以验证大多数细胞是否已经分离。如果所有细胞都脱落，添加 2 mL 的新鲜转换介质，并轻轻地上下吹笛 2-3 次，以分离团块（如果需要）。</li>
	<li>将细胞收集在 15 mL 管中，并添加额外的 3 mL 介质，以进一步稀释细胞分离溶液。首先用额外的介质清洗油井，以确保所有细胞都已收集完好。</li>

</ol>3. 从NPC生成诱导星形细胞
