허혈/재관류(SCI)에 의해 유도된 척수 손상은 감소된 결과1,2,3,4와관련된 대동맥 수리의 가장 치명적인 합병증 중 하나입니다. SCI에 대한 현재 예방 및 치료 옵션은 척수관류 압력2,5,5,6,7,8,9를개선하기 위해 뇌척수액 압력(CSP)의 정상화뿐만 아니라 거형역학적 파라미터의최적화를포함한다. 이러한 기동의 구현에도 불구하고 SCI의 발생률은 여전히 대동맥수리10, 11,12의복잡성에 따라 2 %와31% 사이입니다.
최근에는 미세순환이 주목을 받고있으며, 13,14. 미세 순환은 세포 산소 섭취 및 대사 교환의 영역이므로 장기 기능 및 세포 무결성13에서중요한 역할을한다. 손상된 미세 순환 혈류는 증가된 사망률15,16,17,18,19와관련된 조직 허혈의 주요 결정자이다. 척수 미세 순환의 손상은 신경 기능 저하및 결과20,21,22, 23과관련이있다. 따라서 SCI 치료를 위한 미세 관전의 최적화는 가장 유망한 접근법입니다. 거시순환 적 최적화에도 불구하고 미세 순환 장애의 지속성은26,27,28,29로기술되었다. 혈역학 일관성의 이러한 손실은 허혈/재관류를 포함한 다양한 조건에서 자주 발생하며, 직접 미세 순환 평가 및 미세 순환 표적 요법26,27,30의필요성을 강조한다.
지금까지 척수 미세 순환 행동20,31의실시간 평가를 위해 레이저 도플러 프로브를 사용한 연구는 거의 없습니다. 기존 연구는 종종 간헐적 인 사용 및 사후 분석(32,33)에의해 제한되는 마이크로 스피어 주입 기술을 사용했다. 마이크로스피어 사출 기법을 이용한 상이한 측정 횟수는 파장이 다른 마이크로스피어의 가용성에 의해 제한됩니다. 더욱이, 레이저 도플러 기술과는 대조적으로, 이 방법에 대한 사후 조직 처리 및 분석이 필요하기 때문에, 마이크로 퍼퓨전의 실시간 평가는 불가능하다. 여기서, 우리는 허혈/재퍼퓨전의 돼지 큰 동물 모형에서 척수 미세 순환의 실시간 평가를 위한 실험 프로토콜을 제시합니다.

여섯 마리의 동물은 모두 프로토콜이 완료될 때까지 살아남았습니다. 동물 중량은 48.2 ± 2.9 kg; 5마리의 동물이 수컷이고, 한 마리의 동물은 암컷이었다. 척수 바늘 프로브 삽입뿐만 아니라 척수 플럭스 측정은 모든 동물에서 가능했다.
